THE tremendous impact of large models represented by ChatGPT[1]-[3]makes it necessary to con-sider the practical applications of such models[4].However,for an artificial intelligence(AI)to truly evolve,it needs to possess a physical“body”to transition from the virtual world to the real world and evolve through interaction with the real environments.In this context,“embodied intelligence”has sparked a new wave of research and technology,leading AI beyond the digital realm into a new paradigm that can actively act and perceive in a physical environment through tangible entities such as robots and automated devices[5].

生物转化反式茴脑生成的苯丙素类芳香物质被视为天然原料,能广泛应用于食品加工、香料生产及医药等领域。以能代谢反式茴脑的假单胞菌(Pseudomonas sp.)AT39为出发菌株,采用转座子(pTnMod-RKm’)插入突变构建了含有6137个突变体的突变体库,筛选出44株反式茴脑代谢受影响的突变株。检测突变菌及野生菌的反式茴脑利用能力,发现突变菌AT39-15的反式茴脑代谢能力是野生菌的14%。利用hiTAIL-PCR技术对突变菌AT39-15转座子侧翼序列扩增,获得插入突变的基因,测序结果显示基因大小为1047 bp,基因序列与恶臭假单胞菌(Pseudomonas sp.)JYR-1的反式茴脑氧化酶基因(TAO)一致性达100%。研究结果为下一步构建基因工程菌提供了基因资源。

从早期使用数学工具探索神经活动机理,到采用人工神经网络模型模拟人类智能,再到基于深度网络解决特定问题,人工智能历经80年发展,当前已进入大模型时代,其特点是立足基础大模型、服务广大社会群体。基础大模型的发展面临一系列挑战:一是大模型训练所必需的数据,质与量有待提高;二是高端算力设备受限制,模型训练高电耗、高碳排放带来环境问题;三是大模型的社会化带来歧视与偏见、虚假信息传播、隐私侵犯、影响就业等社会发展问题。在当今去全球化的国际形势下,为克服以上挑战,健全包含联邦数据、联邦控制、联邦管理和联邦服务的联邦生态系统,构建基于区块链保真(trustable+reliable+usable+effective/efficient,TRUE)和去中心化自治组织和分布式自治运营(DAO)的智能系统联邦,发展基于TAO(TRUE DAO)的基础智能(foundation intelligence,FI),为大模型时代人工智能的发展提供必要和有力的支撑。

Professor Weisun TAO is a distinguished educator, a pioneering scientist of biochemical researches and one of the founders of the protein chemistry research in China (Fig. 1). She is a main founder of the Department of Chemistry at Utopia University, the Department of Chemistry and the Department of Biology at Jilin University. She was appointed as a professor in Utopia University, Northeast Institute of Technology, Jilin University, and the honorary director of the Chinese Biochemical Society. She also established the disciplines of organic chemistry and bio chemistry at Jilin University.

Psychopathological outcomes and defence mechanisms in clinically healed adults with a paediatric cancer history: an exploratory study

Background The incidence of paediatric cancers has increased in recent years;however,with advances in the treatment of paediatric cancer,almost 80%of children and adolescents who receive a diagnosis of cancer become long-term survivors.Given the high stress levels associated with cancer,it becomes important to ascertain the risk and likelihood of psychiatric disorders in adult paediatric cancer survivors.Aims This study aims to investigate the relationship between defence styles and predisposition to psychiatric diseases in adults with a history of paediatric cancer.Methods We performed an explorative study on a sample of 66 clinically healed adults with a history of paediatric cancer(survivors)during follow-up visits at the University Hospital‘Policlinico G Rodolico’of Catania(Italy)and 98 healthy controls among medicine students.We administered the Defence Mechanism Inventory(DMI)to assess defence styles.The Symptom Checklist-90-Revised(SCL-90-R)and the Davidson Trauma Scale(DTS)were administered to assess psychopathological indices.We conducted comprehensive statistical analysis based on correlation analysis and mediation analysis to investigate the relationship between defence styles and psychopathological outcomes in survivors compared with controls.Results The survivors obtained statistically significant lower values in TAO,PRO and TAS defence styles and a higher value in REV.Both groups showed non-pathlogical mean scores in DTS and SCL-90-R(with an exception of the obsessive-compulsive subscale),with lower mean values among survivors.The results of mediation analysis showed that TAS had mediation effects on interpersonal sensitivity,anxiety,PSDI,GSI and avoidance,while TAO had mediation effects on DTS total score and intrusivity.Thus,for these psychopathological indices,the effect of the oncological pathology was indirect and mediated by TAO or TAS.Our analysis exlcluded mediation effects between the remaining variables and defence styles.Conclusion Integrating data from mediation and correlation analysis,

The proliferation and differentiation of adult stem cells must be tightly controlled in order to maintain resident tissue homeostasis. Dysfunction of stem cells is implicated in many human diseases, including cancer. However, the regulation of stem cell proliferation and differentiation is not fully understood. Here we show that the sterile-like 20 kinase, TAO, controls tissue homeostasis by regulating the Hippo pathway in the Drosophila adult midgut. Depletion of TAO in the progenitors leads to rapid intestinal stem cell （ISC） proliferation and midgut homeostasis loss. Meanwhile, we find that the Janus kinase （JAK）/signal transducer and activator of transcription （STAT） signaling activity and cytokine production are significantly increased, resulting in stimulated ISC proliferation. Furthermore, expression of the Hippo pathway downstream targets, Diapl and bantam, is dramatically increased in TAO knockdown intestines. Consistently, we show that the Yorkie （Yki） acts downstream of TAO to regulate ISC proliferation. Together, our results provide insights into our understanding of the mechanisms of stem cell proliferation and tissue homeostasis control.

目的:本研究旨在从甲状腺相关眼病(Thyroid-associated ophthalmopathy,TAO)人群的一般特征、血液学特征、骨性眼眶结构特征出发,探索球后注射糖皮质激素后眼外肌直径改变的影响因素,并进一步构建预后预测模型,便于指导临床应用。方法:回顾性收集和分析2015年1月1日至2021年12月31日在华中科技大学同济医学院附属协和医院眼科行球后注射糖皮质激素的TAO患者的资料和数据,包括患者性别和年龄等一般资料,血常规、大生化、甲状腺功能等实验室检查结果,以及通过眼眶CT测量获得的眼球突出度、轴位眶深、眼外肌直径等数据。然后利用R软件4.0.3和EasyR行数据分析,通过单因素与多因素Logistic回归分析筛选出影响患者治疗效果的因素。最后制定整合各种临床参数的Nomogram预测模型。结果:纳入TAO患者有47例,共77只眼,男女比例为1.06:1.00,平均年龄为49.23±9.40岁。注药后眼外肌变细或基本不变的眼数较多(63/77),眼外肌增粗的眼数较少(14/77)。与眼外肌增粗相关的危险因素有女性(OR:4.71,95%CI:1.72～12.91,P=0.003)和高血压(OR:6.30,95%CI:1.33～29.91,P=0.021),而保护因素有轴位眶深(OR:0.74,95%CI:0.56～0.97,P=0.031)和血红蛋白浓度(OR:0.94,95%CI:0.91～0.98,P=0.002)等,提示轴位眶深更深、血红蛋白浓度更高的患者球后注射糖皮质激素后原增粗眼外肌变细的可能性更大。依据眼球突出度、轴位眶深、性别、肌酐、CAS评分建立的Nomogram模型,预测注药后增粗眼外肌直径改变的准确度为92.70%,敏感度和特异度分别为89.47%和95.45%。结论:与TAO患者球后注射糖皮质激素治疗后眼外肌增粗相关的危险因素有女性、高血压,保护性因素有轴位眶深、血红蛋白浓度等;利用逐步后退法构建的Nomogram模型预测TAO患者球后注射糖皮质激素后原增粗眼外肌直径改变的准确度高。
