N1-methyladenosine(mA),first characterized more than 50 years ago, has recently been found as a reversible and dynamic modification in ***,mA-induced mispairing and stall during reverse transcription were developed for transcriptome-wide mapping of mA sites. Herein,we present a straightforward method for the site-specific detection of mA in mRNA by ligation-assisted *** method utilizes the fact that the methyl group in mA disrupts the Watson-Crick base pairing,which therefore compromises the nick ligation efficiency of *** decreased ligation efficiency will result in less ligated products,thus enabling the subsequent quantitative real-time PCR detection and evaluation of the site of *** proposed method was successfully employed to accurately determine mA in HEK293 t cells and lung carcinoma tissues.
