t-type calcium channel expression in cultured human neuroblastoma cells

Human neuroblastoma cells （SH-SY5Y） have similar structures and functions as neural cells and have been frequently used for cell culture studies of neural cell functions.Previous studies have revealed L-and N-type calcium channels in SH-SY5Y cells.However,the distribution of the low-voltage activated calcium channel （namely called t-type calcium channel,including Cav3.1,Cav3.2,and Cav3.3） in SH-SY5Y cells remains poorly understood.the present study detected mRNA and protein expres-sion of the t-type calcium channel （Cav3.1,Cav3.2,and Cav3.3） in cultured SH-SY5Y cells using real-time polymerase chain reaction （PCR） and western blot analysis.Results revealed mRNA and protein expression from all three t-type calcium channel subtypes in SH-SY5Y cells.Moreover,Cav3.1 was the predominant t-type calcium channel subtype in SH-SY5Y cells.

We found t-type calcium channel blocker Ni2+ can efficiently induce the formation of cement gland in Xenopus laevis animal cap explants. Another t-type specific calcium channel blocker Amiloride can also induce the formation of cement gland, while L-type specific calcium channel blocker Nifedipine has no inductive effect. these results may offer us an new approach to study the differentiation of cement gland through the change of intracelluax calcium concentration.

Role of t-type calcium channels in Generating Hyperexcitatory Behaviors during Emergence from Sevoflurane Anesthesia in Neonatal Rats

In the current study,we sought to investigate whether t-type Ca^(2+)channels(tCCs)in the brain are involved in generating post-anesthetic hyperexcitatory behaviors(PAHBs).We found that younger rat pups(postnatal days 9-11)had a higher incidence of PAHBs and higher PAHB scores than older pups(postnatal days16-18)during emergence from sevoflurane anesthesia.the power spectrum of the theta oscillations(4 Hz-8 Hz)in the prefrontal cortex was significantly enhanced in younger pups when PAHBs occurred,while there were no significant changes in older pups.Both the power of theta oscillations and the level of PAHBs were significantly reduced by the administration of tCC inhibitors.Moreover,the sensitivity of tCCs in the medial dorsal thalamic nucleus to sevoflurane was found to increase with age by investigating the kinetic properties of tCCs in vitro.tCCs were activated by potentiated GABAergic depolarization with a sub-anesthetic dose of sevoflurane(1%).these data suggest that(1)tCCs in the brain contribute to the generation of PAHBs and the concomitant electroencephalographic changes;(2)the stronger inhibitory effect of sevoflurane contributes to the lack of PAHBs in older rats;and(3)the contribution of tCCs to PAHBs is not mediated by a direct effect of sevoflurane on tCCs.

Estrogen is a steroid hormone,regulated a variety of physiological functions,and distributed in many parts of the brain.Previous report demonstrated that estrogen regulation of t-type calcium channel activity in the barin through binding to its receptors.the lateral habenula（LHb） is an important hinge linked the limbic system and the midbrain,it involved in many physiological functions,meanwhile play a role on the estrogen-dependent behaviours,such as increase the sexual receptivity and maternal behavior during pregnancy and postpartum.LHb contained estrogen receptor,and also expressed a higher density of t-type calcium channels subtypes,Cav3.1 and Cav3.3,which play an important role in regulation neuronal excitability.We speculate estrogen may affect the LHb neurons function by regulating the activation of t-type calcium channel.In this study,we assessed the effect of estrogen on t-type calcium current in LHb neurons by patch clamp techniques with ovariectomized oil-treated and E2-treated rats.We evaluated the expression of t-type calcium channels protein and mRNA by real-time PCR,western blot.We found that compare with oil-treated group,the expression of Cav3.1 and Cav3.3 mRNA decrease significantly in LHb in E2-treated group,the protein expression of the two subunits also decrease in E2-treated group.Whole-cell patch recordings in LHb neurons revealed that E2 treatment significantly attenuated t-type current,and increased decay time and recovery time of t-type calcium channels,increased the slope factor of activation and inactivation curve.taken together,these data indicate the impact of estrogen to LHb may be related to change the activity of t-type calcium channels.

In the inner retina,a neuronal circuit formed by ganglion cells（RGCs）,amacrine cells（ACs） and bipolar cells（BCs） integrates and modulates visual information.We investigated how activation of the cannabinoid CB1receptor（CB1R）,expressed on the terminals of ACs and BCs,as demonstrated by multiple labeling immunohistochemistry,regulates mtPSCs and mEPSCs of RGCs in rat retina.WIN55121-2（WIN）,a CB1R agonist,suppressed mtPSCs of RGCs by inhibiting L-type calcium channels of ACs.In contrast,the existence of inhibitory inputs from ACs to BCs was indispensable for WtN-induced suppression of mEPSCs of RGCs.Furthermore,the depolarized membrane potential of BCs in the absence of inhibitory inputs from ACs inactivated the t-type calcium channel that was responsible for the generation of RGC mEPSCs,so mat this channel could not be regulated.When the inactivation was eliminated by the hyperpolarization caused by the inhibitory inputs,the t-type channel was inhibited by activation of presynaptic CBlRs on BCs,which resulted in the suppression of the mEPSCs.this unique feature of CB lR-mediated retrograde regulation provides a novel mechanism for modulating excitatory synaptic transmission in the inner retina.

Objective: t-type calcium channels are widely distributed in central and peripheral nervous system including spinal lamina II(also named substantia gelatinosa, SG) neurons. Activation of t-type channels induces an inward Ca current(I) which contribute to regulation of nociceptive transmission. In order to know in detail the cell-specific expression and function of I in SG neurons, we investigated the pharmacological characterization of I and the morphology of the SG neurons with or without I. Methods: Whole-cell patch-clamp recordings were applied to SG neurons in spinal cord slices prepared from male juvenile Sprague–Dawley rats. Neurobiotin 488 was added to the intracellular solutions to label the somata, dendrites and axon in a fraction of SG neurons recorded. Results: I was recorded in nearly 46% of the SG neurons(n = 177/384) with the average amplitude of 85.80 ± 7.49 p A. the slope of activation curve and voltage for half-maximal current(V) of I is 3.75 and-53.18 m V, respectively. the slope and V of inactivation curve was-7.31 and V =-79.69 m V, respectively. Moreover, NiCl, an antagonist of ttype calcium channels, inhibited I in a concentration-dependent manner and the IC50 was 89.2 mM when fitted with a Hill equation(Hill coefficient = 1.7). In morphological experiments, 111 SG neurons were used for analysis. Five major cell types were identified: islet, central, radial, vertical, and unclassified cells. Among them, I was recorded in 23 neurons(21 %). Moreover, the proportion of I in each subtypes of SG neurons were: islet(35 %, n = 8/23), central(21 %, n = 5/23), radial(9 %, n = 2/23), vertical(22 %, n = 5/23), and unclassified(13%, n = 3/23) neurons. Conclusions: Most SG neurons showed a functional I that was blocked by NiCl in a concentration-dependent and reversible manner. the expression ratio of I is highest in islet neurons and lowest in radial neurons. Altogether, current study will help us better understand the role of calcium channels in nociceptive processing circuits in the spinal dorsal horn
