CD4^＋CD25^＋ Regulatory t cells （treg） have been found to down-regulate immune activation in HIV-1 infection. However, whether the depletion of treg benefits to the disease status of HIV infection remains undefined. to address this issue, we enumerated the treg absolute counts and frequency in 75 antiviral-naive HIV-1-infected individuals in this study. It was found that HIV-infected patients displayed a significant decline in treg absolute counts but a significant increase in treg frequency. In addition, with disease progression indicated by CD4 t-cell absolute counts, circulating treg frequency gradually increased; while treg absolute counts were gradually decreased, suggesting that the alteration of treg number closely correlated with disease progression in HIV infection Functional analysis further showed that treg efficiently inhibit both CD4 and CD8 t cell proliferation in vitro. thus, our findings indicates that treg actively participate in pathogenesis of chronic HIV infection, influencing the disease progression.

Objective:to observe the effect of moxibustion on t lymphocyte subsets in peripheral blood of rats with gastric cancer. Methods:Sixty healthy Sprague-Dawley (SD) rats were adaptively fed for one week. By the random number table method, 10 rats were randomly selected as a blank group, and 12 rats were randomly selected to simulate the tumor transplantation process;after transplantation, 10 rats were randomly selected as a sham operation group. the remaining 38 rats were used to prepare gastric cancer models by gastric transplantation of the Walker-256 tumor tissue;8 rats were randomly selected to verify the successful modeling after 7 d;the remaining 30 rats were randomly divided into a model group, a moxibustion group and an infrared group by the random number table method, with 10 rats in each group. From the first day of enrollment, the rats in the moxibustion group received mild moxibustion at Zhongwan (CV 12), Guanyuan (CV 4) and bilateral Zusanli (St 36) (the first group) and bilateral Pishu (BL 20) and Weishu (BL 21) (the second group), and the two groups of acupoints were alternated every other day. the rats in the infrared group received infrared radiation on the stomach area and the area on the back between the t12-t13 spinous processes, the two areas were alternated every other day. Rats in the moxibustion group and the infrared group were treated for 20 min each time, once a day for 21 d. Rats in the blank group, the sham operation group, and the model group were simultaneously grasped and fixed, and no other treatment was performed. After 21 d of intervention, the rats in each group were fasted for 12 h, and blood was collected from the orbits. the numbers of CD3+, CD3+CD4+, CD3+CD8+ t lymphocytes in peripheral blood were determined by flow cytometry, and the ratio of CD3+CD4+/CD3+CD8+ was calculated. the rats were sacrificed and the thymus was dissected under sterile conditions to calculate the thymus index. Objective: to evaluate the role of regulatory t cells (tregs) at late stages of stroke. Methods:Anti-CD25 antibody (or PBS as a control) was injected to reduce the pool of tregs in Wistar rats;then, ischemia was induced transiently by middle cerebral artery occlusion during 60 min and reperfusion was allowed for 7 d. then, treg frequency was analyzed in peripheral blood, spleen and lymph nodes. Neurological score (0-6) and infarct volume were also determined. Results: Nine days after injection, the CD4+CD25+ t cells were reduced by 70.4%, 44.8% and 57.9% in peripheral blood, spleen and lymph nodes, respectively compared to PBS-treated rats. In contrast, the reduction of CD4+FOXP3+ t cells was lower in the same compartments (38.6%, 12.5%, and 29.5%, respectively). the strongest reduction of CD25+CD4+ t cells was observed in those FOXP3-negative cells in blood, spleen and lymph nodes (77.8%, 52.8%, and 60.7%, respectively), most likely corresponding to activated t cells. Anti-CD25-treated transient middle cerebral artery occlusion rats had a lower neurological deficit and did not develop tissue damage compared with PBS-treated animals. Conclusions: these findings suggest that treatment with anti-CD25 in our model preferentially reduce the t cell population with an activated phenotype, rather than the treg population, leading to neuroprotection by suppressing the pathogenic response of effector t cells.

Recombinant human B7.2 IgV-like domain expressed in bacteria maintains its co-stimulatory activity in vitro

OBJECtIVE: to investigate which of the two immunoglobulin (Ig)-like domains, the immunoglobulin variable region homologous domain IgV (hB7.2 IgV) and the immunoglobulin constant region homologous domain IgC (hB7.2 IgC) on the human B7.2 molecule contains receptor binding sites, and to evaluate whether the B7.2 protein expressed in bacteria has biological activity in vitro. MEtHODS: three fragments of hB7.2 IgV,hB7.2 IgC and the complete extracellular region of human B7.2 containing both the IgV and IgC domains,hB7.2 Ig (V+C), were amplified by PCR and subcloned into pGEM-teasy. three recombinants,pGEX-4t-3-hB7.2 IgV,pGEX-4t-3-hB7.2 IgC and pGEX-4t-3-hB7.2 Ig (V+C), were generated by cloning the fragments into a prokaryote expression plasmid (pGEX-4t-3) and transformed into the host strain E. coli DH5alpha. the relevant target fusion proteins consisting of GSt and hB7.2 IgV,hB7.2 IgC and hB7.2 Ig (V+C), were identified by SDS-PAGE and Western blotting. With the presence of the first signal imitated by anti-CD3 antibody, t cell activation was observed by exposing purified t lymphocytes to each soluble form of the three bacterially-produced human B7.2 fusion proteins by [(3)H]-tdR incorporation. RESULtS: three recombinant fusion proteins of human B7.2, GSt-hB7.2 IgV, GSt-hB7.2 IgC and GSt-hB7.2 Ig (V+C) were produced and detected in inclusion body form from engineered bacteria. With the first signal present,t lymphocytes proliferated when co-stimulated by bacterially-produced either GSt-hB7.2 Ig (V+C) or GSt-hB7.2 IgV fusion proteins, but not by GSt-hB7.2 IgC. CONCLUSIONS: Functional human B7.2 fusion protein can be produced in bacteria. the IgV-like domain of human B7.2 is sufficient for B7.2 to interact with its counter-receptors and co-stimulate t lymphocytes.

目的了解NK细胞和.γδt细胞在HIV/AIDS患者不同疾病阶段中的改变特点,探讨其在AIDS发病机制中的作用.方法以311例未接受过抗HIV治疗的HIV/AIDS患者为研究对象,经流式细胞仪检测患者外周血CD4+t淋巴细胞、NK细胞和γδt细胞比例及计数,根据CD4+t淋巴细胞0.35×109/L.将病例分为低、中、高CD4+t淋巴细胞组,进行组间比较.Mann-WhitneyU检验和Kruskal-Wallis检验进行两组和多组独立样本秩和检验,Spearman和Pearson检验进行相关性分析.结果 HIV/AIDS患者外周血NK细胞比例及计数的中位数分别为8.4%和103×106L,γδt细胞比例及计数中位数分别为3.4%和41×106L,均明显低于健康对照组(Z=-5.029,Z=-7.723,Z=-2.437,Z=-6.063;均Pt淋巴细胞组CD4+t淋巴细胞计数中位数分别为0.062×109L、0.276×109L、0.482×109L,NK细胞计数分别为89×106L、97×106L、146×106L,低、中CD4+t淋巴细胞组间的NK细胞计数差异无统计学意义,但均显著低于高CD4+t淋巴细胞组(Z=-3.392,P=0.001;Z=-4.849,Pt淋巴细胞组γδt细胞计数中位数分别为29×106L、43×106L、59×106L,两两之间比较均差异有统计学意义(Pt细胞数量降低程度存在差异.

Objective： to investigate the clinical efficacy and immunological mechanism of herbal cake-partitioned moxibustion for aging process. Method： the herbal cake-partitioned moxibustion was adopted for 223 cases to observe the aging scores before and after the treatment. Apart from that, the t-lymphocyte subsets and changes of IL-2 and β-EP were also detected. Results： After treatment, the aging scores of 223 cases were all substantially reduced, along with an improvement of clinical symptoms, a strengthened cellular immune function, and an increase of total t-lymphocyte count. In addition, the CD4＋/CD8＋ ratio was restored normal, the synthesis or secretion of IL-2 was increased and the β-EP （as the neurotransmitter to modulate immune function） was substantially improved. Conclusion： the aging process is closely associated with the immune function. Moxibustion can modulate abnormal immune function and stabilize homeostasis and thus delay the aging process.
