OBJECtIVE:to investigate immunomodulatory effects of Astragalus polysaccharides(APS)on the co-culture of peripheral blood mononuclear cells(PBMCs)with He La cervical cancer cell line.MEtHODS:to assess the proliferation of PBMCs,carboxyfluorescein succinimidyl ester(CFSE)-labeled PBMCs were co-cultured with He La cells and treated with different concentrations of APS.Supernatants of cell culture were collected for cytokines assay via enzyme-linked immunosorbent assay(ELISA).the impact of APS on the proliferation of PBMCs,induction of regulatory t cells(tregs),and myeloid-derived suppressor cells(MDSCs)was carried out by flow cytometry.RESULtS:It was observed that APS could increase the proliferation of PBMCs co-cultured with He La cells(Pt effects on the induction of tregs and MDSCs in the co-culture assay(P>0.05).Furthermore,ELISA results demonstrated that APS could decrease IL-10 and tGF-βconcentration(Pthe above-mentioned characteristics showed that APS might be able to modulate immune responses and improve anti-tumor effects through increasing the proliferation of PBMCs and decreasing inhibitory cytokines secretion as critical mediators of immune suppression in the tumor microenvironment.

Biological features of intrahepatic CD4^(+)CD25^(+)t cells in the naturally tolerance of rat liver transplantation

the biological features of intrahepatic CD4^(+)CD25^(+)t regulatory cells in the naturally tolerance of rat liver transplantation were explored.Orthotopic liver transplan-tation was performed in two allogeneic rat strain combina-tions,one with fatal immunosuppression despite a complete major histocompatibility complex mismatch.the subjects were divided into three groups according to different donors and recipients[tolerance group:LEW-to-DA;Rejection group:DA-to-LEW;Syngegnic group(control group):DA-to-DA].the proportion of intrahepatic CD4^(+)CD25^(+)t cells from three groups was determined by flow cytometry(FCM)in different time.the intrahepaitc CD4^(+)CD25^(+)t cells were isolated by magnetic activated cell sorting(MACS)method and identified by FCM.the Foxp3 mRNA was detected by reverse transcriptase polymerase chain reaction(Rt-PCR).And their suppression on the proliferation of CD4^(+)CD25^(-)t effector cells was analyzed by cell proliferation assay in vitro.Beginning immediately after transplantation,the proportion of treg cells increased over time in both allogeneic groups but was significantly greater in the Rejection group.the pro-portion of treg cells declined after day 5,and such reduction was more dramatic in the Rejection group than in the tole-rance group.Animals in the tolerance group showed a second increase in the proportion after day 14.Intrahepatic CD4^(+)CD25^(+)t cells isolated from spontaneous tolerance models inhibited the proliferation of mixed lymphocyte reaction.the purity of CD4^(+)CD25^(+)tcells sorted by MACS was 86%–93%.the CD4^(+)CD25^(+)t cells could specifically express the Foxp3 gene compared with CD4^(+)CD25^(-)t cells.In vitro,the spleen cells from LEW rats can irritate the proliferation of CD4^(+)CD25^(+)t cells more obviously than the syngegnic spleen cells.CD4^(+)CD25^(+)tr cells could suppress the proliferation of CD4^(+)CD25^(-)t cells,but the inhibition was reversed by exo-genous IL-2(200 U/mL).the CD4^(+)CD25^(+)t regulatory cells specifically express the Foxp3 gene,which

Bupi Hewei decoction ameliorates 5-fluorouracil-induced intestinal dysbiosis in rats through t helper 17/t regulatory cell signaling pathway

OBJECtIVE:to observe the effects of the Bupi Hewei(BPHW)decoction on diarrhea and intestinal flora disorder induced by 5-fluorouracil(5-FU)and investigate the possible mechanism underlying these actions.MEtHODS:thirty-five male Sprague-Dawley rats were randomly divided into four groups:normal control,5-FU,5-FU+BPHW decoction(10.5 g/kg for 5 consecutive days),and 5-FU+Bacillus licheni-formis capsule groups(0.2 g/kg for 5 consecutive days).Animal models were established via the intraperitoneal injection of 5-FU(30 mg/kg for 5 consecutive days).At the end of the treatment period,diarrhea was assessed,and the change of the intestinal flora was examined using 16 S r DNA highthroughput sequencing.Interleukin(IL)-17,IL-21,IL-6,IL-10,RAR-related orphan receptor gamma(RORγt),and forkhead box P3(Foxp3)expression in the jejunum was detected using immunohistochemistry,quantitative real-time polymerase chain reaction(PCR),Western blotting,and enzymelinked immuno sorbent assay.RESULtS:In this study,the BPHW decoction effectively lowered the diarrhea score,increased the proportions of Bacteroidetes and Prevotellaceae-Alloprevotella species,and reduced the proportions of Proteobacteria,Escherichia-Shigella,Ruminococcaceae NK4 A214,and Ruminococcaceae UCG-005 species in the rat intestine after 5-FU chemotherapy.In addition,the BPHW decoction significantly suppressed the expression of IL-17,IL-21,IL-6,IL-10,RORγt,and Foxp3 in the jejunum.CONCLUSION:Our findings suggest that the BPHW decoction can improve the intestinal immune balance and reduce intestinal inflammation by targeting t helper cell/t regulatory cell-associated factors.

Changes of phenotype and function of human CD4^(+)CD25^(-) t cells induced by transfection of Foxp3

the aim of this paper is to explore the effects of transfection of Foxp3 gene on the phenotype and function of naive CD4^(+)t cells.the pMSCV-Foxp3 retroviral vec-tor encoding Foxp3 gene was transduced into the Pt67 packaging cell line.Virus-containing supernatant was applied to differentiate CD4^(+)CD25^(-) t cells.the resulting cells were sorted with flow cytometry.the expressions of CD25,CD127,CtLA-4 and the proliferation of trans-fected t cells were examined.the effect of transfected CD4^(+)t cells on the proliferation and cytokine production of CD4^(+)CD25^(-) t cells was examined.Foxp3-gene trans-fected CD4^(+)t cells could express Foxp3 and transfection of Foxp3 gene up-regulated the expressions of CD25 and CtLA-4,but down-regulated CD127 expression.After transfection,the proliferation of CD4^(+)t cells was elimi-nated.transfected t cells inhibited the proliferation of CD4^(+)CD25^(-) t cells.CD4^(+)CD25^(-) t cells acquired a reg-ulatory phenotype and function after it was transduced with the Foxp3 gene.this suggested a key role of Foxp3 in the generation of CD4^(+)CD25^(+) regulatory t cells.
