this study was conducted to investigate the effects of AFB on t-cell subsets and cytokines mRNA expression in the small intestine of *** hundred and fifty six one-day-old healthy Cobb broilers were randomly divided into control group(0 mg/kg AFB)and AFB group(0.6 mg/kg AFB) with three replicates per group and 26 birds per replicate for 21 days,*** 7,14,and 21 days of age,the duodenum,jejunum and ileum were sampled for analyzing t cell subsets(CD3+,CD3+CD4+and CD3+CD8+)by flow cytometry as well as IL-2,IL-4,IL-6,IL-10,IL-17,IFN-γ and tNF-α mRNA expression by *** percentages of t-cell in the intra-epithelial lymphocytes(IELs) and lamina propria lymphocytes(LPLs) of duodenum,jejunum and ileum in the AFB group showed a decreased tendency in comparison to the control *** mRNA expression of cytokines in the three intestinal segments in the AFB group presented a general decline compared with the control *** data demonstrated that 0.6 mg/kg AFB in broilers diet could reduce the percentages of t-cell subsets and the expression level of cytokines mRNA in the small intestine,implying that the immune function of the intestinal mucosa might be *** reduction of cytokines mRNA expression may be closely associated with the decreased proportions of t cells subsets induced by AFB.

Mechanisms underlying lineage commitment and plasticity of human yo t cells

Phenotypic and functional heterogeneity are the hallmarks of effector and memory t cells. Upon antigen stimulation, y t cells differentiate into two major types of memory t cells： central memory cells, which patrol the blood and secondary lymphoid organs, and effector memory cells, which migrate to peripheral tissues, y t cells display in vitroa certain degree of plasticity in their function that is reminiscent of that which is observed in conventional CD4 t cells. Similar to CD4 t cells, in which a plethora of specialized subsets affect the host response, y8 t cells may readily and rapidly assume distinct thl-, th2-, th17-, tFH and t regulatory-like effector functions, suggesting that they profoundly influence cell-mediated and humoral immune responses. In addition to differences in cytokine repertoire, y~ t cells exhibit diversity in homing, such as migration to lymph node follicles, to help B cells versus migration to inflamed tissues. Here, we review our current understanding of y t-cell lineage heterogeneity and flexibility, with an emphasis on the human system, and propose a classification of effector y t cells based on distinct functional phenotypes.

A Randomized Controlled trial Comparing total Glucosides of Paeony Capsule and Compound Glycyrrhizin tablet for Alopecia Areata

Objective： to observe the efficacy and safety of total glucosides of paeony capsule （tGPC） in patients with mild and moderate alopecia areata. Methods： A total of 86 outpatients were randomly allocated into two groups of tGPC （treatment, 44 cases） and compound glycyrrhizin tablet （control, 42 cases）. the treatment group was given oral tGPC, three times daily and 600 mg per time; the control group was given oral compound glycyrrhizin tablets, three times daily and 50 mg per time. In addition, both groups were given 10 mg of vitamin B2 and tapped the bold patches with massage. the treatment course was three months for both groups. Peripheral blood t-cell subsets （CD3^＋CD4^＋, CD3^＋CD8^＋, th, ts, th/ts） of 10 patients randomly selected from each group respectively were tested before and after three months of treatment. the effectiveness and adverse reaction of all cases were observed each month. the safety was evaluated according to the incidence rate of adverse reaction. Results： In the treatment group, the cured and markedly effective rate was 36.36% （16/44）, 50.00% （22/44） and 68.18% （30/44） at the end of first, second and third month of treatment, respectively, and the incidence rate of adverse reaction was 13.64% （6/44）. In the control group, the cured and markedly effective rate was 38.10% （16/42）, 57.14% （24/42） and 71.43% （30/42）, respectively, and the incidence rate of adverse reaction was 16.67% （7/42）. the cured and markedly effective rate and the incidence rate of adverse reaction were similar in both groups （P〉0.05）. tGPC and compound glycyrrhizin tablet can inhibit CD3^＋CD4^＋ and CD3^＋CD8^＋, and decrease the ratio of th/ts （P〈0.05）. Conclusion： tGPC is effective and safe in the treatment of alopecia areata.

Study on Clinical Effect and Mechanism of Jianpi Qingre Huayu Recipe (健脾清热化瘀方) in treating Patients with Gastric Ulcer

Objective： to study the effects of Jianpi Qingre Huayu Recipe （健脾清热化瘀方, JQH） in curing gastric ulcer and to preliminarily probe into its pathogenic mechanism, Methods： Fifty patients with gastric ulcer of Pi （脾）-insufficiency and stasis-heat syndrome type were assigned to the treated group （30 patients） and the control group （20 patients）. they were treated respectively with JQH and Ranitidine. At the same time, another group consisting of 20 healthy persons was set up for normal control. the clinical effect on gastroscopic figure and traditional Chinese medicine （tCM） syndrome were observed. Changes of t-cell subsets and interleukin-8 （IL-8） in serum as well as IL-8 in mucosa around the gastric ulcer were determined before and after treatment by flow cytometry and ELISA. Results： Comparison of the total effective rate on gastroscopic figure in the treated group and the control group （86.7% vs 80.0%） showed insignificant difference, but the cure rate and markedly effective rate in the former （50.0% and 20.0%） was higher than that in the latter （40.0% and 15.0%） respectively. Comparison of the total effective rate on tCM syndrome in the treated group and in the control group （96.7% vs 70.0%） showed insignificant difference, but the cure rate and markedly effective rate in the former （63.3% and 23.3%） was higher than that in the latter （50.0% and 20.0%） respectively. Serum levels of CD3^＋, CD4^＋, CD8^＋ got restored to normal range in the treated group after treatment but it was not so in the control group. IL-8 level in gastric mucosa was improved in both groups but the improvement in the treated group was better. Cenclusien： JQH could effectively treat gastric ulcer and partly reduce its recurrence through improving patients＇ immune function.
