Effect of immunopotentiator of the Chinese herbal medicine on the quantity of t lymphocyte in chicken

In order to study the mechanism of immunopotentiator, the quantity of t lymphocyte was observed. total 240 1-day chikens were divided into 3 groups randomly： one contrast group and two groups being drunk immunopotentiator according to the concentration 10 mL·L^-1 and 5 mL·L^-1 lasting for 48 d. the number of t lymphocyte in blood was measured by E-rosette when the chikens were at 12-, 24-, 36-, and 48-day. the results showed that the percent oft lymphocyte in the trial group was obviously higher than that of the contrast, and the 10 mL·L^-1 group was higher than that of the 5 mL·L^-1 group; the quantity distribution of t lymphocyte in intestinal mucosa lymphoid tissue of 14-day, 21-day chiken was surveyed in the contrast group and the10 mL·L^-1 group with the method of routine histology-slices and ANAE, the results showed that the quantity of t lymphocyte in the 10 mL·L^-1 group was significantly higher than that of the contrast, which indicated that immunopotentiator increased markedly the quantity of t lymphocyte, and the 10 mL·L^-1 group was higher than that of the 5 mL·L^-1 group.

t cell activation,proliferation,and differentiation into effector and memory states involve massive remodeling of t cell size and molecular content and create a massive increase in demand for energy and amino acids.Protein synthesis is an energy-and resource-demanding process;as such,changes in t cell energy production are intrinsically linked to proteome remodeling.In this review,we discuss how protein synthesis and degradation change over the course of a t cell immune response and the crosstalk between these processes and t cell energy metabolism.We highlight how the use of high-resolution mass spectrometry to analyze t cell proteomes can improve our understanding of how these processes are regulated.

Immunocytes,including the microglia,are crucial in the neurodegenerative process in old people.However,the understanding regarding microglia heterogeneity and other involved immunocytes remains elusive.We analyzed 26,456 immunocytes from 12-and 26-month-old zebrafish brains at single-cell resolution.Microglia and t lymphocytes were detected in the brain at both time points.two types of microglia were annotated,namely,ac+microglia and xr+microglia,which were clustered into subsets 1,2,3,4,5,and subsets 6,7,8,9,respectively.Diversified microglia predominated the adult brains and cooperated with t cells to perform the functions of immune response and neuronal nutrition.We validated the specific microglia markers.the novel transgenic lines,tg(lgals3bpb:e GFP)and tg(apoc1:e GFP),were created,which faithfully labeled ac+microglia and served as valuable labeling tools.However,the microglia population reduced while t cells of six subtypes intriguingly increased to serve as the primary immune cells in aged brains.Unlike in 12-month-old brains,t cells,together with microglia,exhibited a coordinated signature of inflammation in the 26-month-old brains.Our findings revealed the immunocytes atlas in aged zebrafish brains.It implied the involvement of microglia and t cells in the progression of neurodegeneration in aging.

the intestinal mucosa is a highly compartmentalized structure that forms a directbarrier between the host intestine and the environment, and its dysfunction couldresult in a serious disease. As t cells, which are important components of themucosal immune system, interact with gut microbiota and maintain intestinalhomeostasis, they may be involved in the process of intestinal barrier dysfunction.P2X7 receptor (P2X7R), a member of the P2X receptors family, mediates the effectsof extracellular adenosine triphosphate and is expressed by most innate or adaptiveimmune cells, including t cells. Current evidence has demonstrated thatP2X7R is involved in inflammation and mediates the survival and differentiationof t lymphocytes, indicating its potential role in the regulation of t cell function.In this review, we summarize the available research about the regulatory role andmechanism of P2X7R on the intestinal mucosa-derived t cells in the setting ofintestinal barrier dysfunction.

Background Increasing evidence suggests that, by the production of indoleamine 2, 3-dioxygenase （IDO）, dendritic cells （DC） may reduce the activity of t lymphocytes and inhibit t lymphocyte proliferation-induced immune tolerance.One promising way is inspired by increasing IDO expression in DG cells for immune tolerance after transplantation. the aim of this work was to examine the effect of interferon-y （IFN-γ） on the expression of IDO by DC.Methods Spleen-derived rat DCs were cultured and induced by cytokines, and the expression of OX62 and surface molecules CD80 and CD86 were measured with flow cytometry. After the DCs were induced by IFN-γ at different concentrations （0, 100, 300, 500 U/ml）, the expression levels of IDO mRNA were measured with real-time PCR, and the expression levels of IDO protein in DCs were measured with Western blotting. the allogeneic mixed lymphocyte reaction （MLR） was used to test the effects of DCs incubated with different concentrations of IFN-γ on allogeneic t lymphocyte proliferation.Results Under the microscope, the DCs induced by IFN-γ showed a typical dendritic morphology. the expression rate of OX62 was above 80% and the positive expression rates of CD80 and CD86 were both about 80%. the expressions of IDO mRNA and IDO protein increased gradually with the increase of IFN-γ concentration, showing statistical significance in the differences between the groups （P 〈0.05）. Compared with the control DC, the DC incubated with IFN-γ had a notable decrease in allostimulatory activity （P 〈0.05）. With the increasing IFN-γ concentration, the t lymphocyte proliferation decreased, and the difference between the groups was also statistically significant （P 〈0.05）.Conclusions the highly purified spleen derived rat DCs can be successfully acquired through the improved adhesion in-vitro method. IFN-γ can induce increased expression of IDO in spleen-derived rat DCs and reduce the spleen DCs＇ capacity to stimulate the proliferation of allogeneic t cells.

the graft-versus-tumor （GVt） effect of t cells induced by tumor antigen-pulsed CD8α＋ dendritic cells （DCs） in vitro was investigated in this study. Immature CD8c（DCs were prepared from C57BL/6 （H-2b） bone marrow cells by using a cytokine cocktail. On the 3rd day of culture, CD8α＋ DCs were pulsed by allogeneic （Balb/c, H-2d） EL9611 leukemia antigen, or RM-1 syngeneic prostate cancer antigen, with the concentration series of 0, 2.5, 5.0, 10.0, 20.0 μg/mL, respectively, then antigen-loaded immature CD8α＋ DCs were co-cultured with syngeneic t cells according to the DC/t ratio of 1：1, 2：1 and 4：1. t cell proliferation was measured by Mtt assay. Cytokines including interferon gamma （IFN-γ） and interleukin-10 （IL-10） in CD8α＋ DCs and t co-culture supernatant were detected by using ELI SA. Cytotoxic effect of antigen-specific t cells was tested by LDH release assay. Conventional mature DCs （mDCs） induced, from C57BL/6 （H-2b） bone marrow cells by using granulocyte-macrophage colony stimulating factor （GM-CSF） and interleukin-4 （IL-4） served as a control. the results showed that the proliferative activity of t cells stimulated by CD8α＋ DCs loaded with allogeneic or syngeneic tumor antigen was augmented with the CD8＆ DC/t ratio increased （P〈0.05）. When antigen concentration 〈 5 μg/mL and CD8a＋ DC/t ratio 〈 2：1, the ability of CD8α＋ DCs to stimulate t cell proliferation was higher than mDC control in allogeneic tumor antigen-pulsed groups （P〈0.05）, but not in syngeneic tumor antigen-pulsed groups （P〉0.05）. the level of IFN-γ and IL-10 in CD8α＋DCs and t cell co-culture supernatant were increased in both allogeneic and syngeneic antigen-pulsed groups （P〈0.05）, and the cytokine level was higher in allogeneic antigen-pulsed groups than in syngeneic antigen groups when the CD8α＋ DC/t was 1：1 or 2：1 （P〈0.05）. there existed a negative correlation between the level of IL-10 and t cell proliferation. t cell cytotoxieity assay showed that when CD8α＋ DCs were pulsed with allogeneic tumor antigen, the maximal t cell killing efficiency c

It is well-documented that t lymphocyte proliferation and apoptosis are abnormal in idiopathic thrombocytopenic purpura （ItP） children. However, the underlying regulation mechanisms especially in terms of signal transduction remain unknown. In this paper, we reported the changes of protein kinase C （PKC） activity in peripheral blood t lymphocytes and the effect of PKC on t lymphocyte proliferation and apoptosis. We demonstrated that in ItP children, the activator （PMA） and inhibitor （H-7） of PKC affected on t lymphocyte proliferation and apoptosis dramatically, but they altered little in healthy children. PKC activity was significantly enhanced in ItP children together with an increased expression of FasL on CD3^＋ t, CD4^＋t and CD8^＋t cells, resulting in a positive correlation between PKC activity and the expression of FasL on t cells. While the PKC activity and the platelet count were negatively correlated. taken together, our findings suggest that the PKC activation may enhance t lymphocytes activity, suppress t cell apoptosis and be involve in thrombocytes damage as a mechanism related to immune pathogenesis of ItP. Human cytomegalovirus （hCMV） has evolved multiple mechanisms to escape the host immune recognition and innate or adaptive immune responses. Among them, hCMV has developed strategies to modulate the expression and/or function of human leukocyte antigens （HLAs）, including by encoding series of infection stage-dependent hCMV proteins to detain and destroy the expression of HLA molecules on the surface of infected cells. this disturbs the antigen presentation and processing, by encoding MHC class I homologues or selective up-regulation of particular HLA class I molecules binding to NK cell inhibitory receptors, and by encoding specific ligand antagonists to interfere with NK cell activating receptors. Here we discussed the molecular mechanisms utilized by the hCMV to alter the formation, transportation and expression of HLA antigens on the infected cell surface. the knowledge about hCMV modulating HLA expression could benefit us to further understand the pathogenesis of viral diseases and may eventually develop novel effective immunotherapies to counteract viral infections and viral associated diseases.

BACKGROUND Emerging evidence has demonstrated that fecal microbiota transplantation(FMt)has a promising therapeutic effect on mice with experimental colitis and patients with ulcerative colitis(UC),although the mechanism of FMt is unclear.AIM to evaluate the protective effect of FMt on UC and clarify its potential dependence on the gut microbiota,through association analysis of gut microbiota with colon transcriptome in mice.MEtHODS Dextran sodium sulfate(DSS)-induced experimental colitis was established and fecal microbiota was transplanted by gavage.Severity of colon inflammation was measured by body weight,disease activity index,colon length and histological score.Gut microbiota alteration was analyzed through 16S ribosomal ribonucleic acid sequencing.the differentially expressed genes(DEGs)in the colon were obtained by transcriptome sequencing.the activation status of colonic t lymphocytes in the lamina propria was evaluated by flow cytometry.RESULtS Compared with the DSS group,the weight loss,colon length shortening and inflammation were significantly alleviated in the FMt group.the scores of disease activity index and colon histology decreased obviously after FMt.FMt restored the balance of gut microbiota,especially by upregulating the relative abundance of Lactobacillus and downregulating the relative abundance of Clostridium_sensu_stricto_1 and turicibacter.In the transcriptomic analysis,128 DEGs intersected after DSS treatment and FMt.Functional annotation analysis suggested that these DEGs were mainly involved in t-lymphocyte activation.In the DSS group,there was an increase in colonic t helper CD4^(+)and t cytotoxic CD8^(+)cells by flow cytometry.FMt selectively downregulated the ratio of colonic CD4^(+)and CD8^(+)t cells to maintain intestinal homeostasis.Furthermore,Clostri dium_sensu_stricto_1 was significantly related to inflammation-related genes including REG3G,CCL8 and IDO1.CONCLUSION FMt ameliorated DSS-induced colitis in mice via regulating the gut microbiota and t-cell modulation.
