Lupus nephritis(LN) has a high incidence in systemic lupus erythematosus(SLE) patients, but there is a lack of sensitive predictive markers. The purpose of the study was to investigate the association between the CD4^(+)CD8^(+)double positive T(DPT) lymphocytes and LN. The study included patients with SLE without renal impairment(SLE-NRI), LN, nephritic syndrome(NS), or nephritis. Peripheral blood lymphocyte subsets were analyzed by flow cytometry. Biochemical measurements were performed with peripheral blood in accordance with the recommendations proposed by the National Center for Clinical Laboratories. The proportions of DPT cells in the LN group were significantly higher than that in the SLE-NRI group(t=4.012, Psusceptibility, and the interference of NS and nephritis could be effectively excluded when assessing the risk of renal impairment during SLE with DPT cell proportion.

Antimicrobial Activities of Extracts of Macrosphyra longistyla against Gram-Positive Oral Biofilm-Formers from School Children in Southwestern Nigeria and Toxicity Studies Using Brine Shrimps

The world will benefit from more effective antimicrobial agents against oral conditions arising from the actions of biofilm forming bacteria. Also, information is lacking on the oral biofilm-forming bacterial diversity in Southwestern Nigeria. In this study, we isolate and characterize oral biofilm producing bacteria in the oral cavities of schoolchildren in Southwestern Nigeria. We also investigate the antimicrobial properties of Macrosphyra longistyla extracts against the biofilm-formers and the toxicity of potent extracts. Samples were obtained from 109 schoolchildren aged 4 - 14 years from Lagos, Oyo and Osun States. Agar well diffusion technique was used in the antimicrobial susceptibility testing. Toxicity testing was done using brine shrimps (Artemia salina). Biofilm-formers in this study are Klebsiella sp., Streptococcus sp., Staphylococcus sp., and Micrococcus sp. Ethanol leaf extracts had the highest activity against all biofilm-producing bacteria. Ethanol stem bark extract, which elicited activity against Klebsiella only, was found to be less toxic than the ethanol leaf extract. Staphylococcus showed >10 mm susceptibility to the ethanol and aqueous extracts of Macrosphyra longistyla. Streptococcus and Micrococcus were susceptible to the antimicrobial actions of the ethanolic leaf extracts. Although the ethanol extracts of the leaves had lower minimum inhibitory concentrations than the ethanol extracts of the stem bark, toxicity studies showed ethanol extracts of the stem-bark to be more toxic than the ethanol extracts of the leaves. In conclusion, ethanolic extracts of Macrosphyra longistyla show potential as sources of antimicrobials against gram-positive, oral biofilm-forming bacteria.

VEGFA rs3025039 and biliary atresia susceptibility in Chinese population:a systematic review and meta-analysis

Background Previous studies have suggested an association between vascular endothelial growth factor A(VEGFA)rs3025039 polymorphism and biliary atresia(BA).However,this conclusion is controversial and there is no published pooled evidence of this association.Methods This study was conducted and reported according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses.The protocol was registered with PROSPERO(International Prospective Register of Systematic Reviews).A thorough search was performed on databases including PubMed,Embase,and Chinese Biomedical Database up to August 2020.This study included 846 cases of BA and 2821 controls concerning VEGFA rs3025039 polymorphism.We selected relevant studies based on the following inclusion criteria:(1)the study design was case-control and cohort and(2)the patients carried standard clinical diagnoses of BA,etc.The exclusion criteria were as follows:(1)patients with other related diseases,(2)lack of requisite information and(3)duplicate data.The OR(odd ratio)and the corresponding 95%CI(confidence interval)were calculated to estimate the association.Results This study on VEGFA rs3025039 polymorphism in the Chinese population included 846 cases and 2821 controls.The results showed that there was no significant association between rs3025039 and susceptibility to BA under four genetic models.The results of the subgroup analysis were similar to the overall results.Conclusions This meta-analysis shows that rs3025039 was not associated with susceptibility to BA in the Chinese population.Further validation may entail additional research.PROSPERO registration number CRD42020203812.

Zika virus disease is caused by Zika virus infection,as transmitted by Aedes spp.mosquitoes.Many of the Zika virus strains isolated from patients display different pathogenicities toward humans.The vector mosquitoes for Zika virus are mainly of the Aedes genus,especially Aedes aegypti and Aedes albopictus.However,susceptibility and interactions between Aedes spp.mosquitoes and Zika viruses remain unclear.In this study,we chose two Zika virus strains(FSS 13025 and PRVABC59)with different abilities to infect the primary vector mosquitoes Ae.aegypti and Ae.albopictus.The transcriptomes and small RNA profiles of infected and uninfected mosquitoes were comparatively analyzed,and differentially expressed genes were functionally examined using RNA interference.According to the results,the susceptibility of PRVABC59 was higher than that of FSS13025 in Aedes vector mosquitoes,and Ae.aegypti was more susceptible to Zika virus than was Ae.albopictus.For PRVABC59 infection,specific differential expression profiles correlated with Ae.aegypti and Ae.albopictus,and susceptibility was significantly affected when three targeted genes were successfully knocked down.Compared with PRVABC59,infection of Ae.albopictus with FSS 13025 generated more 21-nt virus small interference RNA.It can be concluded that the susceptibility of vector Aedes spp.mosquitoes to Zika viruses varies and that the interactions between mosquitoes and Zika virus correlate with susceptibility.

The susceptibility of the northern corn rootworm Diabrotica barberi （Smith ＆ Lawrence） to mCry3A and eCry3.1Ab proteins derived from Bacillus thuringiensis （Bt） was determined using a diet bioassay. Northern corn rootworm neonates were exposed to different concentrations of mCry3A and eCry3.1Ab, incorporated into artificial diet. Larval mortality was evaluated after 7 d. The mCry3A and eCry3.1 Ab proteins were found to be toxic to the northern corn rootworm larvae. The LCs0 and LC99 values for mCry3A were 5.13 and 2482.31 μg/mL, respectively. For eCry3.1Ab, the LC50 and LC99 values were 0.49 and 213.01 μg/mL. Based on the estimated lethal concentrations, eCry3, lab protein was more efficacious to northern corn rootworm larvae than mCry3A. These lethal concentration values will be used as diagnostic doses for routine annual monitoring for change in susceptibility of field collected northern corn rootworm to mCry3A, and eCry3.1Ab toxins.

Background:Glioma is one of the central nervous system(CNS)tumors in children,accounting for 80%of malignant brain tumors.Nucleotide excision repair(NER)is a vital pathway during DNA damage repair progression.Xeroderma pigmentosum group D(XPD)or excision repair cross‐complementing group 2(ERCC2)is a critical factor in the NER pathway,playing an indispensable role in the DNA repair process.Therefore,the genetic variants in XPD may be associated with carcinogenesis induced by defects in DNA repair.Methods:We are the first to conduct a multi‐center case‐control study to investigate the correlation between XPD gene polymorphisms and pediatric glioma risk.We chose three single nucleotide polymorphisms and genotyped them using the TaqMan assay.Results:Although there is no significant association of these genetic variations with glioma susceptibility,the stratified analysis revealed that in the subtype of astrocytic tumors,the rs13181 TG/GG genotype enhanced glioma risk than the TT genotype,and carriers with two to three genotypes also elevated the tumor risk than 0‐1 genotypes.Conclusion:In conclusion,our findings provided an insight into the impact of XPD genetic variants on glioma risk.

This study investigates the susceptibility of hot cracking and weldment heat treatment of Haynes 230 superalloy. The Varestriant test was conducted to evaluate this susceptibility. Welding was performed by gas tungsten arc welding （GTAW） and plasma arc welding （PAW） with stress relief heat treatment and solid solution heat treatment. A tensile test is then performed to measure the changes in the mechanical properties of the heattreated material. The results indicate that the number of thermal cycles does not affect the susceptibility of Haynes 230 superalloy to hot cracking. However, it does increase the strain. In weldment of heat treatment, stress relief annealing increases the yield strength and tensile strength of the welded parts. The section of the tensile specimens shows fibrous fractures on the welded parts, regardless of whether they are heat-treated.

Objective:To investigate the susceptibility to temephos.permethrin and deltamethrin of Aedes aegypti Ae.aegypti),collected from areas with high incidence of dengue hemorrhagic fever cases in Phitsanulok Province.Thailand.Methods:The F1 progenies of Ae.aegypti colony,originated from five sub-districts including Aranyik.Hua Ro,Nai Muang.Ban Krang and Tha Pho,were used in the bioassays following the procedures of World Heath Organization.For larval bioassay.the late third or early fourth-instar lanae were tested with different concentrations of temephos.For adult bioassay.the females were exposed to 0.75%permethrin or 0.05%deltamethrin.LC_(50)value and mortality rate were analyzed to compare the insecticide susceptibility of the larvae and the adults in each area,respectively.Results:The LC_(50) value of temephos for the larvae from Aranyik.Hua Ro.Nai Muang,Ban Krang and Tha Pho sub-districts was 0.***.0.***.0.***.0.***.and 0.113 ppm,respectively.For permethrin,the highest morlalitv rate(86.84%) was found in the mosquitoes from Aranyik but the others were more resistant with the lower morlality rates(16.00-42.67%).The adult morlality rates after exposing to dellaiiielhrin were higher(82.34-98.67%) in all areas.Conclusions:Ae.acgypti larvae were still susceptible to temephos.Conversely,most tested adults tended to resist the penmethrin and deltamethrin.

Sugarcane borer, Diatraea saccharalis （E）, is a major target of transgenic maize expressing Bacillus thuringiensis （Bt） proteins in South America and the US mid- south region. Resistance development in target pest populations is a major threat to the sustainable use of Bt crops. In our field trials in 2009, a significant number of live borers and plant injury fi＇om D. saccharalis were observed in an experimental SmartStaxTM maize line. The objective of this study was to assess the relative susceptibility of two field popula- tions ofD. saccharalis collected from non-Bt and Bt maize plants containing SmartStaxTM traits to five individual Cry proteins. The five Bt proteins included two proteins （Cry 1A. 105 and Cry2Ab2） that were expressed in SmartStaxTM maize plants and three other common Bt proteins （Cry 1Aa, Cry 1Ab and Cry l Ac） that were not produced in SmartStaxTM. Larval mortality and growth inhibition on Bt diet of the fourth generation after field collections were evaluated 7 days after release of neonates on the diet surface. The laboratory bioassays showed that 50% lethal concentration （LCs0） values for CrylA. 105 and Cry2Ab2 for the population originated from Bt plants were 3.55- and 1.34-fold greater, respectively, than those of the population collected from non-Bt plants. In contrast, relative to the popula- tion from non-Bt plants, the LCs0 of the population sampled from Bt plants were 3.85-, 2.5- and 1.64-fold more sensitive to CrylAa, CrylAb and CrylAc, respectively. The re- sults did not provide clear evidence to conclude that the observed field survival of D. saccharalis on Bt plants was associated with increased levels of resistance.

The fall armyworm,Spodoptera frugiperda,which destroys many economic crops such as rice and maize,has recently invaded China.Insect viruses as biological control agents play important roles in killing pests.One potential viral insecticide is the environmentally highly infective and virulent densovirus.We successfully rescued Junonia coenia densovirus(JcDV)using its infectious clone in different insect cell lines and larvae of three insect species.Results showed that the lysate of cultured insect cells transfected by the JcDV infectious clone killed the 2 nd instar S.frugiperda.The LD50 of homogenate from JcDV-infected Spodoptera litura to the 2 nd instar S.frugiperda(1.76×10^(8)viral genome copies per larva during 10 d post infection)was higher than that of the 2 nd instar S.litura(7.39×10^(7)Jc DV genome copies)or Helicoverpa armigera larvae(9.71×10^(7)JcDV genome copies).The LT50 of the S.litura homogenate(2.60×10^(9)viral genome copies each larva)to the 2 nd instar S.frugiperda was 6.96 d,longer than that of the S.litura(6.18 d)or the 2 nd instar H.armigera(5.94 d).JcDV could infect the fat body of H.armigera,but not S.frugiperda or S.litura.Although JcDV can infect all three lepidopteran species,their susceptibility to the virus differs.JcDV has great potential as a biological control agent against pests such as S.frugiperda.
