CD8^(+) T lymphocytes are pivotal cells in the host response to antitumor *** microenvironments provide the conditions necessary for regulating infiltrating CD8^(+) T cells in favor of tumor survival,including weakening CD8^(+) T cell activation,driving tumor cells to impair immune attack,and recruiting other cells to reprogram the immune *** in tumor microenvironment,stromal cells exert immunosuppressive skills to avoid CD8^(+) T cell *** this review,we explore the universal function and fate decision of infiltrated CD8^(+) T cells and highlight their antitumor response within various stromal architectures in the process of confronting neoantigen-specific tumor ***,this review provides a foundation for the development of antitumor therapy based on CD8^(+)T lymphocyte manipulation.

The differential effects of prostate stromal cells derived from different zones on prostate cancer epithelial cells under the action of sex hormones

It is well known that prostate cancer （PCa） occurs predominantly in the peripheral zone （PZ）, whereas benign prostatic hyperplasia （BPH） typically develops in the transition zone. To identify possible mechanisms underlying zonal differences, we compared the effects of prostate stromal cells derived from the peripheral zone （PZsc） and the transition zone （TZsc） on a PCa epithelial cell line （PC3） in the presence of sex hormones. First, we observed that androgen receptor （AR） mRNA was more highly expressed in PZsc than TZsc when the cells were treated with dihydrotestosterone （DHT） and β-oestradiol （E2）（P〈0.05）. By ELISA, we looked for differences in the secretion of peptide growth factors from PZsc and TZsc. We found that keratinocyte growth factor （KGF） secretion increased with increasing concentrations of DHT （P〉0.01） and was higher in PZsc than TZsc. Under treatment with DHT plus E2, PZsc secreted more transforming growth factor-β1 （TGF-β1） than TZsc, but this pattern was reversed when the cells were treated with E2 only. With increasing concentrations of DHT, insulin-like growth factor-1 （IGF-1） secretion increased in PZsc but decreased in TZsc. To further characterize the effects of PZsc and TZsc on PC3 cells, we developed a coculture model and performed MTT assays, Western blot analysis and real-time RT-PCR. We found that PZsc promoted PC3 cell proliferation and progression better than TZsc, particularly when treated with 10 nmoll^- 1 DHT plus 10 nmoll^-1 E2. In conclusion, our data suggest that PZsc may have a greater capacity to induce PCa development and progression than TZsc via growth factors regulated by sex hormones. These findings provide possible mechanisms underlying zonal differences in prostate diseases, which may aid the search for novel therapeutic targets for PCa.

Irradiation Injury Temporarily Induces Enhancement of APN/CD13 Peptidase Activity on Aorta-gonads-mesonephros (AGM)-derived stromal cells

This study was designed to investigate the expression of aminopeptidase N （APN）/CD13 on intraembryonic AGM stromal cells, and the change of its enzymatic activity after irradiation injury. The expression of APN/CD13 on AGM stromal cells was assayed by RT-PCR and immunihistochemistry. After the stromal cells in AGM region were irradiated with 8.0 Gy of ^60Co T-rays, APN/CD13 enzymatic activity was measured by spectrophotometer at different time points. The result showed that AGM stromal cells strongly expressed APN/CD13. The enzymatic activity of APN/CD13 decreased temporarily after irradiation injury, then increased to higher level 4 h after irradiation, and it returned to the pre-irradiation level 24 to 48 h after the irradiation. The enzymatic activity of APN/CD13 was temporarily enhanced after irradiation injury, which might be one of the compensatory mechanisms that promote the hematopoietic recovery after irradiation.

The cellular niche for intestinal stem cells: ateam effort

The rapidly self-renewing epithelium in the mammalian intestine is maintained by multipotent intestinal stem cells(ISCs) located at the bottom of the intestinal crypt that are interspersed with Paneth cells in the small intestine andPaneth-like cells in the colon. The ISC compartment is also closely associated with a sub-epithelial compartmentthat contains multiple types of mesenchymal stromal cells. With the advances in single cell and gene editingtechnologies, rapid progress has been made for the identification and characterization of the cellular componentsof the niche microenvironment that is essential for self-renewal and differentiation of ISCs. It has becomeincreasingly clear that a heterogeneous population of mesenchymal cells as well as the Paneth cells collectivelyprovide multiple secreted niche signals to promote ISC self-renewal. Here we review and summarize recentadvances in the regulation of ISCs with a main focus on the definition of niche cells that sustain ISCs.

Pay more attention to the immunologically comparative evaluation of HIV-1 DNA vaccine in combination with adjuvant cytokines： a long way to go

To the Editor： A recent article entitled ＂HIV-1 DNA vaccine with adjuvant cytokines induces specific immune responses against HIV-1 infection in mice＂ was published in the Chinese Medical Journal. The authors descriptively analyzed the adjuvant potential of an interleukin-12 （IL-12）, interleukin-18 （IL-18）, regulated on activation of normal T-cell expressed and secreted factor （RANTES）, macrophage inflammatory protein 1 alpha （MIP-1α or stromal cell derived factor 1 （SDF-1） plasmid when coimmunized with an HIV Gag gene-based DNA vaccine. Consequently the authors believed that IL-12, IL-18, or SDF-1 plasmid DNA potentially serves as immune adjuvants in combination with therapeutic HIV-1 DNA vaccine. The RANTES, MIP-1α plasmid DNA potentially serves as Immune adjuvants in combination with HIV-1 prophylactic vaccine. Although the experiment allows us to consider such a coinoculation strategy as one of the means against HIV-1 infection, some special attentions should be highlighted.

Isolation, Cuture and Biological Characteristic Analysis of stromal and Glandular Epithelial cells of Buffalo (Bubalus bubalis) Endometrium

[Objective] The experiment aimed to set up a method for isolating and culturing endometrial stromal cells (BESC) and endometrial glandular epithelial cells(BEGEC) of buffalo as well as laid foundation for studying biological mechanism of embryo implantation and uterine diseases. [Method] The enzymatic digestion method, scraping method, serial filtration and differential velocity adherent technique were used to isolate BESC and BEGEC, then immunocytochemical method and TRYPAN-Blue assay were used to determine the purity and survival rate of isolated cells. [Result] The BESC and BEGEC were successfully isolated and cultured while immunocytochemical method and cell count method demonstrated that the purity was over 90%. The result of TRYPAN-Blue assay shown that survival rate of BESC and BEGEC was 91% and 78% respectively. [Conclusion] The enzymatic digestion method, scraping method, serial filtration and differential velocity adherent technique could isolate BESC and BEGEC with high purity.
