Angiogenin(ANG) is an angiogenic factor which also promotes tumor cell proliferation ***, the underlying molecular mechanisms remain ***-protein interactions play crucial roles in biological processes,therefore,we reason that identifying ANG-ineracting proteins is a feasible way to elucidate ANG’s biological functions and mechanism of action. We have identified ANG-interacting proteins in HeLa cells using co-immunoprecipitation(Co-IP) coupled with mass spectrum(MS) *** results showed 14 potential ANG-binding partners,two of which had been *** annotation revealed non-muscle myosin heavy chain 9(MYH9),a-actinin 4 (ACTN4),and P-actin(ACTB) to be involved in actin cytoskeleton regulations and cell *** interactions between these three proteins and ANG were confirmed by immunoprecipitation assay,and suggest the possibility that ANG is involved in the *** the coordination of cytoskeleton dynamics and cell-matrix adhesion is particularly important for cell migration,potential ANG functions in the two processes may explain how the protein promotes cell *** verify this hypothesis,we first identified the co-localization of ANG with actin cytoskeleton proteins and found that ANG bond to actin cytoskeleton proteins at the leading edges of migrating ***,we stained F-actin with TRITC-conjugated phalloidin,focal adhesion with paxillin antibody,and detected phosphorylated-FAK levels using Western-blot when ANG was *** showed that the formation of stress fibres was enhanced,focal adhesions were enlarged,and *** was inhibited in ANG down-regulated *** last,we used wound healing assay and time-lapse videomicroscopy to determine the migration speed,and found that migration was retarded in ANG down-regulated ***,our data suggested that ANG might regulate actin cytoskeleton dynamics and cell adhesion through interacting with cytoskeleton proteins,thus promote cell

Ezrin Promotes the Proliferation, Migration, and Invasion of Ovarian Cancer Cells

Objective The underlying mechanism of Ezrin in ovarian cancer(OVCA) is far from being ***, this study aimed to assess the role of Ezrin in OVCA cells(SKOV3 and Ca OV3) and investigate the associated molecular *** We performed Western blotting, reverse transcription-quantitative polymerase chain reaction, MTT, cell colony, cell wound healing, transwell migration and invasion, Rho A and Rac active pull down assays, and confocal immunofluorescence experiments to evaluate the functions and molecular mechanisms of Ezrin overexpression or knockdown in the proliferation and metastasis of OVCA *** The ectopic expression of Ezrin significantly increased cell proliferation, invasiveness, and epithelial–mesenchymal transition(EMT) in OVCA cells. By contrast, the knockdown of endogenous Ezrin prevented OVCA cell proliferation, invasiveness, and EMT. Lastly, we observed that Ezrin can positively regulate the active forms of Rho A rather than Rac-1 in OVCA cells, thereby promoting robust stress fiber *** Our results indicated that Ezrin regulates OVCA cell proliferation and invasiveness by modulating EMT and induces actin stress fiber formation by regulating Rho-GTPase activity, which provides novel insights into the treatment of the OVCA.

Dear Editor, Streptococcus suis （S. suis） is a Gram-positive, facultative anaerobic coccus and an important emerging pathogen. It is associated with bacterial meningitis in adults, especially in southeastern Asia （Wertheim et al., 2009）. A striking feature of S. suis infection is the resulting complications, which may include deafness and vestibular dysfunction. Complications of some kind affect 50% of S. suis patients in Europe and 73% of those in Asia （Gottschalk et al., 2010）. Some S. suis surface-associated factors, such as the capsule and Fhb, may affect the pathogenesis of meningitis. Of these secreted factors, suilysin was found to be the most important.
