The effects of steroid hormones are believed to be mediated by their nuclear receptors(NRs).The p160 coactivator family,including steroid receptor coactivator-1(SRC-1),2 and 3,has been shown to physically interact with NRs to enhance their transactivational *** which SRC-1 has been predominantly localized in the central nervous system including brain and spinal *** is not only localized in neurons but also detectable in neuroglial cells(mainly localized in the nuclei but also detectable in the extra-nuclear components).Although the expression of SRC-1 is regulated by many steroids,it is also regulated by some non-steroidal factors such as injury,sound and ***,SRC-1 has been implied in normal function such as development and ageing,learning and memory,central regulation on reproductive behaviors,motor and food ***,SRC-1 may play a role in the regulation of neuropsychiatric disorders(including stress,depression,anxiety,and autism spectrum disorder),metabolite homeostasis and obesity as well as *** most conditions,the related mechanisms are far from elucidation;although it may regulate spatial memory through Rictor/mTORC2-actin polymerization related synaptic *** inhibitors and stimulator of SRC-1 have shown anti-cancer potentials,but whether these small molecules could be used to modulate ageing and central disorder related neuropathology remain ***,to elucidate when and how SRC-1 is turned on and off under different stimuli is very interesting and great challenge for neuroscientists.

Comprehensive Summary Sepositoside A(1)is the major and first cyclic steroid glycoside isolated from starfishes,which features a strained 16-membered ring formed by tethering C3 and C6 of the steroid aglycone with a linear *** conceived a convergent synthetic plan in which sepositoside A was disconnected into a disaccharide and a steroid-glucose hybrid *** desired 3β,6β-dihydroxy-Δ7,8-23-one aglcyone was readily prepared from dehydroepiandrosterone in 12 *** etherification of the 6β-hydroxy-Δ7,8-steroid with a variety of hexopyranoside-C6-electrophiles and CO_(2)-extrusion of a steroid-pyranose hybrid carbonate were extensively studied;however,formation of the desired steroid-pyranose etherate linkage has not been *** studies on theβ-selective installation of the(1→2)-linked disaccharide onto the steroidal C3β-OH with the corresponding disaccharideα-imidates were also examined,however,formation of theα-glycosides *** present results force us to explore alternative approaches to construct the steroid-pyranose etherate linkage and stepwise assembly of the glycans en route to the starfish cyclic steroid glycosides,which will be presented in two articles that follow.

steroid-bismetalloporphyrin as enzyme model of cytochrome P-450 monooxygenase

The model compounds of androgenic steroid-bismetalloporphyrins 3a—3d and androgenic monometalloporphyrins 2a—2d have been synthesized. Catalytic study in two-phase condition on epoxidation of styrene under the catalysis of model catalysts shows that the catalytic performances of steroid-bismetalloporphyrin is superior to steroid-metalloporphyrin and the corresponding simple metalloporphyrin. The excellent catalytic property of steriod-bismetalloporphyrin can be rationalized as the hydrophobic action of steroid framework and the cooperative action of two metalloporphyrins in catalysis.

steroid elimination within 24 hours after orthotopic liver transplantation:effectiveness and tolerability

BACKGROUND:steroids have been the mainstay of immunosuppressive regimen in liver ***,the use of steroids is associated with various post-transplant *** study evaluated the efficacy and safety of reduced immunosuppressive regimen with steroids(steroid elimination within 24 hours post-transplant) in a cohort of Chinese liver transplant ***:Seventy-six patients in line with the selection criteria were enrolled in this prospective *** patients received anti-IL-2 receptor antibody induction and tacrolimusbased maintenance *** recipients were divided into two groups according to the duration of steroid use:40 transplant in a 3-month withdrawal group and the remaining 36 in a 24-hour elimination *** survival,postoperative infections,biopsy-proven acute rejection and steroidresistant acute rejection,non-healing wound,recurrence of hepatitis B virus(HBV) and hepatocellular carcinoma(HCC),de novo diabetes,hyperlipidemia and hypertension were assessed in the two ***:There was no significant difference in patient survival,incidence of acute rejection episodes and hyperlipidemia,and recurrence of HBV and HCC between the two ***,the incidence rates of post-transplant infection,non-healing wound,de novo diabetes and hypertension were significantly lower in the 24-hour elimination group than in the 3-month withdrawal group(all P values steroid elimination within 24 hours post-transplant is associated with reduced steroidrelated complications without increasing the risk of rejection.

Open label trial of granulocyte apheresis suggests therapeutic efficacy in chronically active steroid refractory ulcerative colitis

AIM： To study the efficacy, safety, and feasibility of a granulocyte adsorptive type apheresis system for the treatment of patients with chronically active ulcerative colitis despite standard therapy. METHODS： An open label multicenter study was carried out in 39 patients with active ulcerative colitis （CAI 6-8） despite continuous use of steroids （a minimum total dose of 400 mg prednisone within the last 4 wk）. Patients received a total of five aphereses using a granulocyte adsorptive technique （Adacolumn, Otsuka Pharmaceutical Europe, UK）. Assessments at wk 6 and during follow-up until 4 mo comprised clinical （CAI） and endoscopic （EI） activity index, histology, quality of life （IBDQ）, and laboratory tests. RESULTS： Thirty-five out of thirty-nine patients were qualified for intent-to-treat analysis. After the apheresis treatment at wk 6, 13/35 （37.1%） patients achieved clinical remission and 10/35 （28.6%） patients had endoscopic remission （CAI〈4, EI〈4）. Quality of life （IBDQ） increased significantly （24 points, P〈0.01） at wk 6. Apheresis could be performed in all but one patient. Aphereses were well tolerated, only one patient experienced anemia. CONCLUSION： In patients with steroid refractory ulcerative colitis, five aphereses with a granulocyte/ monocyte depleting filter show potential short-term efficacy. Tolerability and technical feasibility of the procedure are excellent.
