We report a case of a haemodialysis patient that presented a catheter-related bacteraemia caused by a Coagulase negative staphylococcus. With the utilization of molecular biology techniques the bacterial isolate recovered from catheter was surprisingly identified as S. hominis by sequencing of the 16S ribosomal gene. The S. hominis isolate, which is not often associated with infections in dialysis patients, was resistant to methicillin, being mecA positive, and to daptomycin. The patient was successfully treated with vancomycin together with the catheter retirement.

羟醛缩合反应能形成新的C-C键,在合成有机化学中占有重要地位,在工业中具有很重要的应用价值。本研究旨在利用基因挖矿手段获得新型羟醛缩合酶基因,在大肠杆菌中表达、纯化,并研究它们的催化性能,探索它们在生物催化合成一些重要药物中间体中的应用。通过基因挖矿获得了staphylococcus hominis来源的N-乙酰神经氨酸裂合酶基因(shnal, Accession No. EFS20452.1),将其构建至pET-28a质粒并在大肠杆菌中得到表达,对目的蛋白进行了纯化。通过对该酶的酶学性质研究发现,ShNAL为四聚体,裂解方向的最适反应pH为8.0;合成方向的最适反应pH为7.5,最适反应温度为45℃。在45℃下孵育2 h对ShNAL的活力基本无影响,高于45℃时,活力迅速下降。该酶在pH5.0-10.0的环境中比较稳定,4℃下放置24h酶的残余活力在70%以上. ShNAL对N-乙酰神经氨酸(Neu5Ac)、N-乙酰甘露糖胺(Man)和丙酮酸(Pyr)的Km值分别是(4.0±0.2)mmol/L、(131.7±12.1)mmol/L和(35.1±3.2)mmol/L, kcatlKm值分别为1.9 L/mmol·s、0.08 L/mmols和0.08 L/mmol·s.由ShNAL催化的N-乙酰甘露糖胺与丙酮酸的缩合反应的转化率达到了91.5%,产率达到了89.4%。通过ShNAL和N-乙酰葡萄糖胺异构酶的串联反应实现了从N-乙酰葡萄糖胺到N-乙酰神经氨酸的转化,转化率达到了30%左右。同时也对得到的来源于Blastopirellula marina的N-乙酰神经氨酸裂合酶BmNAL (GenBankAccession No. ZP1092134.1)进行构建和表达,对其酶活性质进行了鉴定。通过基因比对和挖矿技术我们还得到两个苏氨酸醛缩酶,分别来自Deinococcus deserti(DdTA)和Hoeflea phototrophica(HpTA),基因信息来自Genbank (Accession No.分别为YP02785560.1和ZP2165963.1)。通过工程菌的构建和蛋白的表达获得了目的蛋白,对DdTA进行了纯化,并用酶标仪和TLC检测确定两个酶都有催化苏氨酸醛缩酶活性。
