Evaluation of Fluorescent stains for Viability Assessment of the Potato Cyst Nematodes Globodera pallidaand G. ellingtonae

Potato cyst nematodes (PCNs), Globodera pallida and Globodera rostochiensis, are quarantine pests of potato which cause significant damage to production and farm gate revenue worldwide. Accurately assessing viability of PCN eggs is important for eradication and management programs. The goal of this study was to develop a quick and reliable fluorescent staining method to evaluate viability of G. pallida and Globodera ellingtonae eggs. The staining efficiency of eight fluorescent stains was evaluated using G. pallida eggs compared with the conventional Meldola’s Blue (MB) staining method. The staining efficiency of the fluorescent stains ranged from 80.33 ± 2.99 (Sytox Green) to 100% (Acridine Orange) for non-viable eggs. Two stains were further evaluated for their efficiency in assessing viability of encysted eggs from five different greenhouse-reared G. pallida cyst sources which contained both viable and non-viable eggs. For the G. pallida cyst sources, viability ofencysted eggs were estimated to be 41.02 ± 3.81 to 62.66% ± 3.12% when stained with Acridine Orange (AO) and 79.52% ± 1.54% viability for G. ellingtonae. Both staining time and stain concentration were significant for staining efficiency of released and encysted eggs. Staining time and concentration were optimized for released eggs at 4 h at 10 μg/ml and for encysted eggs at 16 h at 25 μg/ml respectively for AO. Fluorescent stains accurately and rapidly assessed percent egg viability and were determined to be as sensitive as a seven-day incubation with the Meldola’s Blue staining method.

Light-Dependent Properties of Finishing and Leather Defects

The uniformity of the colour in leathers for the furnishing and automotive sectors is a particularly important feature, especially for white or light-coloured articles, intended to the high-end market;on these articles any colour alteration can be very visible and unpleasant, with possible technical and economic consequences for the producers. In a previous study, we analysed the possible causes behind the formation of some peculiar pink/salmon stains on different kinds of white/beige/light-coloured leather furnishing items, where we focalized our attention on TiO2 properties and its possible interaction with some organic-based antioxidants. In recent years, due to many other similar cases of defects that occurred to tanners and to upholstery traders, we decided to enhance the investigations concerning this topic. More in detail we analysed, defective leathers, powder pigments and chemicals, in order to identify the possible role of some substances in this issue, with particular reference to some antioxidants and aluminosilicates, where several diagnostic techniques have been utilised, as ATR-IR (Attenuated Total Reflectance IR Spectroscopy), SEM-EDX (Scanning Electron Microscopy-Energy Dispersive X-ray Analysis), XRF (X-ray Fluorescence spectrometry), GC-MS (Gas Chromatography-Mass Spectrometry), and DSC/TGA (Differential scanning calorimetry/Thermogravimetry) equipment.

Green recycling of chicken feather and sheep wool using the partially purified alkaline protease from Bacillus circulansL.

The previously optimized crude alkaline protease from the haloalkaliphilic Bacillus circulans L. was partially purified using ammonium sulphate fractionation and dialysis. The best specific activity (27.7 U/mg protein) was obtained at 80% saturation. The optimum reaction temperature and reaction pH was 47℃ and 9, respectively. The enzyme activity was enhanced with Ca and K chlorides but suppressed with HgCl2 and EDTA. The partially purified protease showed strong proteolytic activity on sheep wool and chicken feather. Also, the enzyme was compatible with the common detergent Tide and could improve its cleaning power in removing blood stain. These findings support the application of the present alka-line protease in biotechnological industries.
