本研究对实验感染食蟹猴疟原虫后9、10和14天的斯氏按蚊阳性蚊胃,进行扫描电镜观察。晚期(分化的)卵囊表面呈现凹凸不平皱褶。成孢子细胞体常呈圆球形或椭圆形,表面光滑,子孢子芽从其表面长出。子孢子体细长,稍弯曲,体表光滑;虫体前部较细,顶端稍平,后部稍粗,末端钝圆。本文对成孢子细胞不同发育阶段的形态予以较详细描述,对子孢子出囊方式做了初步讨论。

A Protocol for a Highly Consistent, High Level Production <i>in Vivo</i>of <i>Plasmodium falciparum</i>Oocysts and sporozoites

Investigation of the intimate relationship between the human malaria parasite Plasmodium falciparum and its Anopheles vector requires the reliable production and isolation of successive sexual stages of the parasite from infected mosquitoes. Such an advance in propagation would benefit a range of molecular, cellular, immunochemical and transmission-blocking research studies. Parasite cultivation, mosquito rearing, infection and subsequent dissection of mosquitoes are all highly technical procedures that require both skill and experience to perform with competence. Furthermore, to produce mosquitoes of an appropriate age to infect during the short period in which parasites are viable for infection demands precise planning in order to coordinate the interacting life cycles of the parasite and vector. Here, a protocol is described for the complete development of P. falciparum within Anopheles stephensi. A very consistent, high level production in vivo of P. falciparum oocysts and sporozoites is demonstrable by dissection of the mosquito midgut and salivary glands, respectively.

Objective:To find out which TLRs can be activatied by sporozoite and discuss its effort in development of malaria ***:Firstly,we acquired purified sporozoites through DE52 *** we amplified and cloned the full-coding sequence of mice TLR1,TLR3,TLR5,TLR6,TLR7 and TLR9,and established a TLRs agonist screening platform,then systemically screened the possible TLRs agnoists contained in sporozoites lysate using NF-κB or IFN-βreporter assay;Secondly,we tested whether sporozoites lysate could induce peritoneal macrophage to release inflammatory cytokines dependent on both TLR2;Finally,the inhibitory role of TLR2 in the development of pre-erythrocytic stage was determined according to the level of both liver parasite load and parasitemia in the wide-type and TLR2 knockout mice after challenged with wild-type ***:Here,we firstly found purified sporozoites lysate could only activate TLR2,but not other known *** study shown either TLR2 homodimization,or TLR2heterodimization TLR2/1,TLR2/6 could recognize sporozoite lysate,but the ability to activate TLR2 of sporozoites was almost lost after boiling water bath for 20 *** sporozoite lysate could also induce the macrophages to release pro-inflammatory cytokines,including IL-6 and TNF-α,in a TLR2-dependent ***,TLR2 signaling pathway was critical for the host immune response against th esporozoite challenge,as the levels of both liver parasite load and subsequent parasitemia were significantly elevated in either TLR2-or MyD88-deficient mice,compared to those of the wild-type *** of TLR2 by agonists could inhibit the development of parasite in the *** observed higher level of parasite burden in TLR2-/-mice was demonstrated closely associated with the reduced production of pro-inflammatory cytokines(IFN-γ,IL-6 and TNF-α)in the liver and recombinant mouse IFN-γ,IL-6 and TNF-αcan significantly reduced liver-stage parasite burden in TLR2-/-***

Objective:To explore the effect of hepatocyte growth factor signaling pathway activation on Plasmodium berghei ***:In this study,hepatocyte growth factor was detected by ELISA and Western blotting *** injury was detected by FITC-dextran absorption assay,and hepatocyte growth factor expression was shown to be expressed in the same injury cells by immunofluorescence against hepatocyte growth *** addition,Activation of hepatocyte growth factor and its receptor signaling pathway was detected with immunoprecipitation and detection of phosphorylation ***:It was found that injury of hepatocytes by sporozoite migration induced the secretion of hepatocyte growth factor and it was hepatocyte growth factor that rendered hepatocytes susceptible to Plasmodium sporozoite *** addition,hepatocyte infections depended on activation of the hepatocyte growth factor and its receptor signaling ***:Our results indicate that hepatocyte growth factor and its receptor may possibly be potential targets for new approaches to malaria treatment.
