Ever since its outbreak,the COVID-19 pandemic caused by SARS-CoV-2 has been spreading rapidly causing a global health crisis.Accurate diagnosis and instituting appropriate intervention to relevant individuals are essential not only to slow down the spread of this pandemic but also to manage our resources efficiently.To date,testing for SARS-CoV-2 infection mostly relies on reverse transcription-quantitative polymerase chain reaction(RT-qPCR)on a nasopharyngeal or saliva specimen and remains the gold standard for diagnosis as it exhibits high sensitivity and Specificity.

Recently,genetically targeted cancer therapies have been a topic of great interest.Synthetic lethality provides a new approach for the treatment of mutated genes that were previously considered unable to be targeted in traditional genotype-targeted treatments.The increasing researches and applications in the clinical setting made synthetic lethality a promising anticancer treatment option.However,the current understandings on different conditions of synthetic lethality have not been systematically assessed and the application of synthetic lethality in clinical practice still faces many challenges.Here,we propose a novel and systematic classification of synthetic lethality divided into gene level,pathway level,organelle level,and conditional synthetic lethality,according to the degree of Specificity into its biological mechanism.Multiple preclinical findings of synthetic lethality in recent years will be reviewed and classified under these different categories.Moreover,synthetic lethality targeted drugs in clinical practice will be briefly discussed.Finally,we will explore the essential implications of this classification as well as its prospects in eliminating existing challenges and the future directions of synthetic lethality.

Purpose:The study aimed to compare the effects of a long general warm-up,a long specific warm-up,and a short specific warm-up upon sprint ability in soccer players.Methods:Twelve male soccer players(age:18.3±0.8 years,mean±SD;body mass:76.4±7.2 kg;body height:1.79±0.05 m) conducted 3 types of warm-ups with 1 week in between:a long general warm-up,a long specific warm-up,and a short specific warm-up followed by 3 sprints of 40 m each.The best,average,and total sprinting times together with heart rate and ratings of perceived exertion were measured.Results:The sprint times(best,average,and total time) were significantly better when performing a long specific or short specific warm-up compared with the long general warm-up(all p Specificity is more important in a warm-up routine before sprint performance than the duration of the warm-up.

Background Necrotizing enterocolitis(NEC)is associated with high mortality and morbidity in neonates.For infants with NEC,intestinal perforation is the most serious complication,and confirming perforation and performing radical surgical treatment as early as possible may reduce mortality and sequelae.The aim of this study was to identify the specific imaging characteristics of intestinal perforation after NEC on supine abdominal X-ray for the early diagnosis of intestinal perforation.Methods A retrospective study was conducted at the Children’s Hospital of Chongqing Medical University.Infants admitted to the hospital from 2013 to 2020 with NEC(Bell’s stage≥Ⅱ)were divided into perforation and non-perforation groups.All infants were examined by abdominal X-ray in the erect and supine positions.The sensitivity and Specificity of specific X-ray signs were analyzed.Results A total of 598 infants were included,113 of whom suffered from perforation.On the supine abdominal films,lucency over the liver shadow,the liver falciform ligament sign,the football sign,the Rigler sign,the triangle sign and more than any one of the above signs had sensitivities of 64.60%,45.13%,37.17%,30.97%,15.93%and 86.73%,respectively.None of these signs were found on erect or supine abdominal films in the non-perforation group.The total of accuracy of prediction was 46.76%,and the Specificity of all the signs was 100%.Conclusion Specific signs on supine abdominal X-ray could be used to confirm perforation in neonates with NEC with 86.73%sensitivity and 100%Specificity.

Forkhead box p3^(+)(Foxp3^(+))regulatory T cells(Tregs)are indispensable for immune homeostasis and for maintaining immune tolerance.Several studies have confirmed that injection of a T cell population depleted of Tregs causes autoimmunity,rejection of grafts and inflammatory disorders,whereas reconstitution with Tregs inhibits these pathogenic processes.Over the last two decades,intense efforts have been made to identify Treg subsets,Treg differentiation processes,and the molecular signatures and regulators that determine Treg lineage Specificity and stability.1–6 Several lines of evidence clearly demonstrate that Foxp3^(+)Tregs do not constitute a homogeneous population,and various subsets of Tregs,such as thymic Tregs(tTregs),in vitro-generated Tregs(iTregs),and peripherally induced Tregs(pTregs),have been identified.5 In addition to Foxp3,epigenetic factors and metabolic processes play key roles in maintaining Treg identity and function,and mediate the switch between effector T cells and Tregs.

To the Editor:Cervical cancer(CC)is one of the leading causes of cancer deaths in women around the world.[1]In clinical practice,screening methods such as human papillomavirus testing and cytology-based detection have the disadvantages of having a high false rate,being hysteretic or ineffective.[2]Novel non-invasive biomarkers that can help diagnose CC with higher Specificity and sensitivity are in great demand.MicroRNA(miRNA)is a type of non-coding small RNA of 18 to 25 nucleotides in length.Circulating miRNAs show the great potential of being cancer biomarkers for the stable existence in peripheral serum or plasma.[3]In this study,we performed a four-stage experiment using a total of 108 CC and 108 normal control(NC)serum samples to explore the diagnostic potential of serum miRNAs in CC.The schematic diagram of the experimental procedure was shown in Supplementary Figure 1,http://***/CM9/A429.

BACKGROUND The prognosis of hepatocellular carcinoma(HCC)patients remains poor despite advances in treatment modalities and diagnosis.It is important to identify useful markers for the early detection of HCC in patients.Preneoplastic antigen(PNA),originally reported in a rat carcinogenesis model,is increased in the tissues and serum of HCC patients.AIM To determine the diagnostic value of PNA for discriminating HCC and to characterize PNA-positive HCC.METHODS Patients with hepatitis C virus(HCV)-related hepatic disorders were prospectively enrolled in this study,which included patients with hepatitis,with cirrhosis,and with HCC.A novel enzyme-linked immunosorbent assay was developed to measure serum PNA concentrations in patients.RESULTS Serum PNA concentrations were measured in 89 controls and 141 patients with HCV infections(50 hepatitis,44 cirrhosis,and 47 HCC).Compared with control and non-HCC patients,PNA was increased in HCC.On receiver operating characteristic curve analysis,the sensitivity of PNA was similar to the HCC markers des-γ-carboxy-prothrombin(DCP)andα-fetoprotein(AFP),but the Specificity of PNA was lower.There was no correlation between PNA and AFP and a significant but weak correlation between PNA and DCP in HCC patients.Importantly,the correlations with biochemical markers were completely different for PNA,AFP,and DCP;glutamyl transpeptidase was highly correlated with PNA,but not with AFP or DCP,and was significantly higher in PNA-high patients than in PNA-low patients with HCV-related HCC.CONCLUSION PNA may have the potential to diagnose a novel type of HCC in which glutamyl transpeptidase is positively expressed but AFP or DCP is weakly or negatively expressed.

A Sensitive Material for Specifically Treating Tetracycline in Water Environment

There are currently many materials for treating residual antibiotics in the environment, but none of them can specifically remove antibiotics. In addition, these materials are not sensitive enough to low concentrations of antibiotics. Here we show that a sensitive and specific material was developed by the preparation of magnetic Fe_(3)O_(4)-PAMAM-antibody complexes for treating tetracycline. The prepared antibody complexes can specifically treat tetracycline from aqueous solutions and the tetracycline removal ability by adsorption was also investigated. Controlled experiments were carried out with the effects of solution pH, temperature, and initial concentration of the tetracycline. The tetracycline was completely removed within 35 min at room temperature 30 ℃ with the maximum removal rate of almost 100%. Therefore, this material for specifically combining antigen and antibody to treat tetracycline indicated good application prospects for the waste water treatment.

Liquid biopsy,in particular circulating tumor DNA(ctDNA)analysis,has paved the way for a new noninvasive approach to cancer diagnosis,treatment selection and follow-up.As a crucial step in the analysis,the extraction of the genetic material from a complex matrix needs to meet specific requirements such as high Specificity and low loss of target.Here,we developed a new generation of microfluidic fluidized beds(FBs)that enable the efficient extraction and preconcentration of specific ctDNA sequences from human serum with flow rates up to 15µL/min.We first demonstrated that implementation of a vibration system inducing flow rate fluctuations combined with a mixture of different bead sizes significantly enhanced bead homogeneity,thereby increasing capture efficiency.Taking advantage of this new generation of high-throughput magnetic FBs,we then developed a new method to selectively capture a double-stranded(dsDNA)BRAF mutated DNA sequence in complex matrices such as patient serum.Finally,as proof of concept,ligation chain reaction(LCR)assays were performed to specifically amplify a mutated BRAF sequence,allowing the detection of concentrations as low as 6×104 copies/µL of the mutated DNA sequence in serum.
