Ethylene plays important roles in plant growth, development, and stress responses, and ethylene receptors have been identified and studied extensively in various plant species. Here we report the cloning of four ethylene receptor genes from soybean. i.e. GmETR1, GmERS1, GmETR2 and GmEIN4. Construction of the phylogenic tree showed that GmETR1 and GmERS1 belong to subfamily I whereas GmETR2 and GmEIN4 belong to subfamily II. The four ethylene receptor genes showed different tissue-specific expression patterns in roots, stems, leaves, cotyledons, flowers, pods and seeds of soybean. These genes were differentially regulated by various abiotic stresses and plant hormones. The possible roles of the four genes in soybean.plant were also discussed.

Plant basic-leucine zipper （bZlP） transcription factors play important roles in many biological processes. In the present study, a bZlP gene, GmbZIP132, was cloned from soybean.and its biological function under abiotic stresses was studied. The transcription of GmbZIP132 was induced by drought and high salt treatments. Among all of the organs analyzed, its expression was the highest in cotyUedon and stems. GmbZIP132 could weakly bind to the GCN4-1ike motif （GLM）（5＇-GTGAGTCAT-3＇） in yeast one-hybrid assay. Compared with wild-type （WT） Arabidopsis plants, transgenic plants overexpressing GmbZlP132 showed reduced abscisic acid sensitivity and increased water loss rate. At the stage of germination, transgenic plants were more tolerant to salt treatment than wild-type plants. The expression of some abiotic stress-related genes, such as rd29B, DREB2A, and PSCS, were upregulated in the transgenic plants. These results indicated that GmbZlP132 was an abioUc stress-related gene, and its overexpression could increase the salt tolerance of transgenic Arabidopsis plants during germination, yet no significant difference of tolerance to abiotic stresses was found between transgenic and wild type plants at the seedling stage.

The main aim of this work was to assess the multi-task role of ferritin （Ft） in the oxidative metabolism of soybean.（Glycine max）. soybean.seeds incubated for 24 h yielded 41 4- 5μg Ft/g fresh weight. The rate of in vitro incorporation of iron （Fe） into Ft was tested by supplementing the reaction medium with physiological Fe chelators. The control rate, observed in the presence of 100μM Fe, was not significantly different from the values observed in the presence of 100 μM Fe-his. However, it was significantly higher in the presence of 100 μM Fe-citrate （approximately 4.5-fold） or of 100 μM Fe-ATP （approximately 14-fold）. Moreover, a substantial decrease in the Trp-dependent fluorescence of the Ft protein was determined during Fe uptake from Fe-citrate, as compared with the control. On the other hand, Ft addition to homogenates from soybean.embryonic axes reduced endogenously generated ascorbyl radical, according to its capacity for Fe uptake. The data presented here suggest that Ft could be involved in the generation of free radicals, such as hydroxyl radical, by Fe-catalyzed reactions. Moreover, the scavenging of these radicals by Ft itself could then lead to protein damage. However, Ft could also prevent cellular damage by the uptake of catalytically active Fe.

Osmotic stress caused by dehydration or high-salt conditions poses a major environmental challenge for plant growth and development. Many studies have shown that protein kinases are important regulators during osmotic stress in plants. In the present study, we describe GmAAPK, a soybean.（Glycine max （L.） Merr.） gene coding for a putative serine/threonine protein kinase （abscisic acid （ABA）-activated protein kinase （AAPK））. The cDNA for GmAAPKis 1 409 bp long and contains a single long open reading frame representing a complete coding region of 359 amino acids. GmAAPKwas mapped onto the D1a＋Q linkage group. Its transcripts are expressed in all tissues, but at high levels in the cotyledon. GmAAPKmRNA was induced by polyethylene glycol, ABA, Ca^2＋ and Na^＋, but not cold （4℃） treatments in soybean.leaves. The results suggest that GmAAPK may participate in the regulatory process during osmotic stresses in soybean.

More substances leaked from a higher-vigor seed sample than from a lower-vigor sample. This indicates that, in some cases, electric conductivity does not represent seed vigor level very well, especially for high-vigor seeds. Results from germination, germination index, leachate conductivity, and the ratio of K^＋/Na^＋ from three-seed lots of Chinese cabbage （Brassica pekinensis （Louv.） Rupr） showed that K^＋/Na^＋ correlated well with germination and germination index. The ability of K^＋/Na^＋ to indicate well changes in vigor was further supported by investigation in soybean.（Glycine max （L.） Merr.） seeds and another cultivar of Chinese cabbage seeds. Thus, seed leakage of K^＋/Na^＋ can accurately indicate seed vigor, whereas the conductivity test failed to do so. Furthermore, K^＋/Na^＋ showed up bigger quantitative differences in vigor level than did the conductivity test. This findings provide a more sensitive and accurate index for the assessment of seed vigor. The mechanisms of Na^＋ and K^＋ ion transport are also discussed.

Effects of doubled CO2 and O3 concentration on soybean.were studied in open-top chambers (OTC). Under doubled CO2 concentration, grain yield and biomass increased, the SOD activity, vitamin C (Vc) and carotenoid (Car) content also increased; Superoxide (O2-.) generating rate decreased, relative conductivity and malondialdehyde (MDA) content significantly *** under doubled O3 concentration, the SOD activity, Vc and Car contents declined, resulting in imbalance of activated-oxygen production, enhanced O2-. generating rate and accelerated process of lipid peroxidation and increase in MDA content and ion leakage of leaves. The final result was decreased grain yield and plant biomass. Interactive effects of doubled CO2 and O3 concentrations on soybean.were mostly counteractive. However, the beneficial effects of concentration-doubled CO2 are more than compensate the negative effects imposed by doubled O3, and the latter in its turn partly counteracted the positive effects of the former.
