Dendrimer-like DNA nanostructures have attractive properties such as mechanical stability, highly branched nanostructure, customized sizes, and biocompatibility. In this study, we construct programmable DNA dendrimeric nanoparticles as efficient vehicles to deliver immunostimulatory cytosine-phosphate-guanosine（CpG） sequences for activation of the immune response. DNA dendrimers decorated with CpGcontaining hairpin-loops triggered stronger immune response characterized by pro-inflammatory cytokines production, in contrast to DNA dendrimers loading linear CpG. After further modification with TAT peptide, a typical cell-penetrating peptide, on the surface of the nanocarriers, CpG loops-loaded DNA dendrimers showed the enhanced cell internalization and cytokines production. The TAT-DNA dendrimer-CpG loops constructs did not affect the viability of immune cells and no detectable cytotoxicity was observed. Our results demonstrate that the DNA dendrimers can serve as designable and safe vehicles for delivery of immune modulators and anticancer drugs.

Peptide and protein self-assembly is of critical importance as a tool to understand the natural life phenomenon such as liquid-liquid phase separation（LLPS） in cells.Although a number of studies have demonstrated the formation of biomolecular condensates through LLPS in cells,the fundamental molecular events underpinning the early stages of their assembly and subsequent growth have remained challenging to elucidate.Herein,we will present how simple peptides can be used as a model to investigate the LLPS events in supramoleuclar self-assembly.We discover that LLPS is a fundamental step leading to nucleation of supramolecular nanostructures,and the metastable droplets（condensates） can be controlled to direct the final assembled supramolecular structures including nanoparticles,nanofibrils and nanotubes as well as crystals so on.The peptide-rich liquid droplets promote the formation of nucleation sites,allowing the emergence of thermodynamically favorable nanostructures following Ostwald’s step rule.It is found that the LLPS may be triggered mainly by an entropy effect associated with the debonding between peptides and solvent molecules（desolvation）.These results provide insights into the fundamental role of metastable liquid phases（condensates） in supramolecular self-assembly and importantly offer useful reference for comprehending the LLSP events in the complex cells.

＜正＞4,4’-Biphenylene-silicas were prepared using the self-assemblies of a chiral low-molecular-weight amphiphile as templates,which exhibited chirality at both a nano and an angstrom level.Their CD spectra were simulated by the time-dependent density functional theory.It was found that the stacking and the twist of the biphenylene rings can be revealed from the first and second CD signals, respectively.Although chiral silicas have been widely developed,it is hard to know that they are chiral or not at angstrom levels.Based on the result shown here,4,4’-bis（triethoxysilyl）-l,l’-biphenyl was used as a chiral sensor for silicas.Moreover,other UV active compounds with conformational chirality are proposed to be used as the chiral sensors for silicas and the other chiral materials.

self-assembly is a powerful technique for the bottom-up construction of discrete,well-defined nanoscale structures.The self-assemble of high symmetric polynuclear compounds have increased greatly due to their inspiration and application in biochemistry and new technology.One of the most desired filed is the combination with molecular magnets which own the potential application in information storage or sensors.Based on the molecular magnets possess a characteristic flexibility not only in their structures,but also in their electronic/spin states and electric/magnetic correlation.It is an efficient way to modulate the intrinsic spin state through the control of intramolecular charge transfer which may accompanied drastic change of magnetism.To obtain such charge flexible magnets,we focus on the modulation function of different metal ions in the self-assembly process from which the different metal ions owns different redox potential that can affect the intramolecular charge transfer.Herein,we designed a series of polynuclear nanocages,[{Fe(Tp)(CN)}{M(HO)}{M(L)(HO)}]×12HO×8CHOH and Fe,Ni,Mnwere used as the central metal ions specifically.The auxiliary ligand pyridine-4-carboxaldehyde was carefully chosen in view of the better coordination ability of pyridine and the easy modification of aldehyde group.

Various stimuli-responsive hybrid vehicles incorporating organic and inorganic components into a container as drug delivery systems have been seen rapid development in the past few decades[1]. As the most important inorganic constituent of human teeth and bones, hydroxyapatite(Ca10(PO)(OH), HAP) has been widely applied for controlled drug delivery due to its excellent biocompatibility, bioactivity, biodegradability[2]. Unfortunately, hollow HAP(H-HAP) microparticles are often observed rapid initial burst release, which limited the broad applications of H-HAP microparticle in the drug delivery area. In this study, a smart drug delivery platform combining H-HAP, chitosan(CS)/hyaluronic acid(HA) natural polyelectrolyte multilayers and Au nanorods(Au NRs) via the layer-by-layer(LbL) self-assembly technique had been prepared. Herein, H-HAP microparticles were chosen as a drug carrier, polyelectrolyte multilayers assuaged the initial burst release and improved the pH-responsive property of the hybrid microparticles due to the stretch and shrinkage of the polymer chains at various pH values. AuNRs capped on the surface of H-HAP/CS/HA microparticles acted as the "gate-keeper" to potentially obstruct the leakage of and endow the hybrid microparticles with excellent NIR-responsive drug release property. The possible formation process regarding to this study is schematically showed in Fig. 1. The study provides a facile and green route to fabricate hierarchical multi-responsive drug carriers with high drug loading capacity and outstanding biocompatibility, which are highly attractive for remotely controllable drug delivery area.

Platinum(Ⅳ) complexes are prodrugs of cisplatin with multiple potential advantages over platinum(Ⅱ)drugs.From chemical point of view,Platinum(Ⅳ) complexes offers two additional axial ligands in their coordination sphere to fine tune there lipophilicity and redox properties.Lithocholic acid or LCA is a secondary bile acid,gains much attention in recent years as potential chemotherapeutic agent which in vitro can selectively kills cancer cells while sparing normal cells.Besides,it also act as an anti-aging molecule by delaying chronological lifespan of budding yeast cells through altering the metabolic processes such as respiration and reactive oxygen species (ROS) production in the mitochondria.We have designed and synthesized a lithocholic acid conjugated platinum(Ⅳ)complex (LT-Platin) and its successful synthesizes was conformed with electron spray ionization mass spectroscopy and H,C and Pt NMR spectroscopy.The potential of LT-Platin to reduce into cytotoxic platinum (Ⅱ) complex was verified with cyclic voltammetry (CV) and inductively coupled plasma mass spectrometry (ICP-MS).LT-Platin spontaneously self-assembles in aqueous solution to form carrier-free nanoparticles with an average size diameter of 100-150 nm as confirmed by transmission electron microscopy (TEM) and dynamic light scattering (DLS).In addition,it showed a time dependent higher cellular uptake in tested cell lines with possible energy-dependent cellular entrance pathway.Biological assay reveals that the LT-Platin showed selective nature toward human lung cancer cells over normal cells compared to its parental compound cisplatin.Interestingly,it shown to be very sensitive against cisplatin resistant lung and breast cancer cells which is also ascribed with its higher cellular uptake but not to its lipophilicity.Mechanistic studies indicates that the complex causes cell cycle arrest at S phase and showed apoptosis-inducing ability in A549 cells.As a conclusion,our study reveals that conjugation of cisplatin to lithocholic acid forms a promising d
