OBJECTIVE:This study investigated the effect of salvia miltiorrhiza-asarum ointment(SMAO)plus Chinese medical massage on knee osteoarthritis in a rat ***:Hulth's method was used to establish a Sprague-Dawley rat model of knee osteoarthritis(OA).The levels of matrix metalloproteinase-13(MMP-13),collagen-Ⅱ,aggrecan,interleukin(IL)-1β,tumor necrosis factor-α(TNF-α),and IL-6 were measured by enzyme-linked immunosorbent *** joint space was assessed by a Perlove X-ray *** was examined by hematoxylin-eosin and Safranin O *** mRNA and protein expression levels of Notch1,MMP-13,collagen-Ⅱ,and aggrecan were measured by quantitative reverse transcription-polymerase chain reaction and Western blotting,***:SMAO plus Chinese medical massage significantly decreased the levels of MMP-13,IL-1β,TNF-α,and IL-6,and increased serum collagen-Ⅱ and aggrecan *** injury of the knee joint was improved by SMAO *** and protein expression of Notch1 and MMP-13 was remarkably downregulated,but collagen-Ⅱ and aggrecan levels were significantly upregulated in cartilage ***:SMAO combined with Chinese medical massage effectively relieves OA symptoms,which may involve inhibiting inflammation through the Notch1/MMP-13 signaling pathway.

Danshen,the dried roots and rhizomes of salvia miltiorrhiza Bunge(***),is widely used in the treatment of cardiovascular and cerebrovascular ***,the bioactive compounds from Danshen,exhibit a wide spectrum of pharmacological properties,suggesting their potential for future therapeutic *** biosynthesis is a complex process involving at least six P450 enzymes that have been identified and characterized,most of which belong to the CYP76 and CYP71 *** this study,CYP81C16,a member of the CYP71 clan,was identified in *** in vitro assay revealed that it could catalyze the hydroxylation of four para-quinone-type tanshinones,namely neocryptotanshinone,deoxyneocryptotanshinone,and danshenxinkuns A and ***81C16 emerged as a potential broad-spectrum oxidase targeting the C-18 position of para-quinone-type tanshinones with an impressive relative conversion rate exceeding 90%.Kinetic evaluations and in vivo assays underscored its highest affinity towards neocryptotanshinone among the tested *** overexpression of SmCYP81C16 promoted the accumulation of(iso)tanshinone in hairy root *** characterization of SmCYP81C16 in this study accentuates its potential as a pivotal tool in the biotechnological production of tanshinones,either through microbial or plant metabolic engineering.

The effects of LaCl3 on the growth,photosynthetic gas-exchange characteristics,chlorophyll fluorescence,and the accumulation of tanshinones and salvianolic acids in salvia miltiorrhiza seedlings were investigated. The results showed that the increase in photosynthesis induced by LaCl3 might be attributed to the enhanced stomatal conductance of the leaves and the increased level of the photochemical efficiency of PS II. The accumulation of tanshinone IIA and cryptotanshinone was markedly increased with the application of LaCl3 at 20 and 60 mg/L,while tanshinone I was only slightly increased. The content of salvianolic acid B was,however,decreased with the treatment of LaCl3 at 200 mg/L.

The purpose of this study is to determine the effect of La^3＋ solution on seed germination and seedling growth of salvia miltiorrhiza. Different concentrations of La^3＋ were used on seed by soaking treated to determine which concentration is the most suitable for the growth of S. miltiorrhiza. Results show that the low concentration of La^3＋ displays the promotion effect on S. miltiorrhiza seed germination rate and germination potential, and the promotion effect reaches the highest level when the concentration of La^3＋solution is 30 mg/L, the germination index and vigor index are the highest in 20 mg/L, and it can also increase the contents of soluble sugar, soluble protein and chlorophyll. Meanwhile, the activity of antioxidant enzyme system（CAT and SOD） are improved, thus the photosynthesis and resistance of plant are enhanced. On the contrary, when the La^3＋ concentration is high, La^3＋ can inhibit plant growth. Thus La^3＋ displays the ＂homesis effect＂ on S. miltiorrhiza growth.

Plant U-box(PUB)E3 ubiquitin ligases play important roles in hormone signaling pathways and in response to different abiotic stresses,but little is known about U-box genes in Danshen(root of salvia miltiorrhiza Bunge).Here,we identified and characterized 70 SmPUB genes based on its genome *** analysis of U-box genes from *** and Arabidopsis suggested that they can be clustered into seven subgroups(I–VII).Typical U-box domains were found in all identified SmPUB genes through the analysis of conserved ***,qRT-PCR was applied to analyze the relative expression levels of U-box genes in *** roots and leaves under PEG-induced water deficit and salt *** revealed that the SmPUB genes exhibited stronger response to drought than to salt *** the best of our knowledge,this report is the first to perform genome-wide identification and analysis of the U-box gene family in ***,and the results provide valuable information for better understanding of the function of U-box in ***.

Effects of salvia miltiorrhiza Bunge extract and its single components on monosodium urate-induced pain in vivo and lipopolysaccharide-induced inflammation in vitro

OBJECTIVE:To investigate the possible antinociceptive effects of salvia(S.)miltiorrhiza Bunge and its single components in monosodium urate(MSU)-induced pain model in mice and lipopolysaccharide(LPS)-induced inflammation model in RAW264.7 ***:Pretreatment of *** Bunge extract(from 1 to 50μg/mL)concentration-dependently attenuated LPS-induced nitric oxide(NO)*** extract of *** Bunge(50 or 100 mg/kg)also caused reversals of decreased threshold for pain in the MSU-treated group as measured by Von-Frey ***,we assessed the antinociceptive and anti-inflammatory properties of the active single components from *** Bunge such as 15,16-dihydrotanshinoneⅠ,tanshinoneⅠ,cryptotanshinone,miltirone,tanshinoneⅡA,and salvianolic acid *** of them showed an anti-inflammatory effect in LPS-induced NO release model and an antinociceptive effect in MSU-treated pain ***:Our results suggest that *** Bunge extract may exert anti-inflammatory effect by reducing LPS-induced NO release and an antinociceptive property in MSU-treated pain ***,tanshinoneⅡA,miltirone,cryptotanshinone,and 15,16-dihydrotanshinoneⅠ,not only appear to be responsible for LPS-induced NO release induced by *** Bunge,but also in the production of *** Bunge extract-induced antinociception in MSU-treated pain ***:Therefore,the analgesic and anti-inflammatory property of *** Bunge indicate it as a therapeutic potential candidate for the treatment of pain and inflammation.

The demonstration of beneficial effects of salvia miltiorrhiza (DanShen) on ischemic diseases has revolutionized the management of angina pectoris, myocardial infarction (MI) or stroke in Chinese society. Experimental studies have shown that DanShen dilated coronary arteries, increased coronary blood flow, and scavenged free radicals in ischemic diseases, so that it reduced the cellular damage from ischemia and improved heart functions. Clinical trials also indicated that DanShen was an effective medicine for angina pectoris, MI, and stroke. This review will focus on DanShen’s effects in angina pectoris, MI and stroke.

Objective:To investigate the protective effects and mechanisms of action of dexamethasone and salvia miltiorrhiza on multiple organs in rats with severe acute pancreatitis (SAP). Methods:The rats were divided into sham-operated, model control, dexamethasone treated, and salvia miltiorrhiza treated groups. At 3, 6, and 12 h after operation, the mortality rate of different groups, pathological changes, Bcl-2-associated X protein (Bax) and nuclear factor-κB (NF-κB) protein expression levels in multiple organs (the pancreas, liver, kidneys, and lungs), toll-like receptor 4 (TLR-4) protein levels (only in the liver), intercellular adhesion molecule 1 (ICAM-1) protein levels (only in the lung), and terminal deoxynucleotidy transferase mediated deoxyuridine triphosphate (dUTP) nick end labeling (TUNEL) staining expression levels, as well as the serum contents of amylase, glutamate-pyruvate transaminase (GPT), glutamic-oxaloacetic transaminase (GOT), blood urea nitrogen (BUN), and creatinine (CREA) were observed. Results:The mortality rate of the dexamethasone treated group was significantly lower than that of the model control group (Psalvia miltiorrhiza treated group (Psalvia miltiorrhiza treated group were significantly lower than those in the dexamethasone treated group (P<0.05). Objective To investigate the effects of salvia miltiorrhiza (SM) extracts on the expression of heme oxygenase-1 (HO-1) and nitric oxide synthase (NOS) in small pulmonary arteries (SPAs) of rats with chronic hypoxia. Methods After two weeks of hypoxia, rats were treated with diltiazem, and small, median, and large doses of SM extracts. The lungs were tested for the expression and distribution of HO-1, endothelial NOS (eNOS) and inducible NOS (iNOS) by using immunohistochemistry and Western blot method. Results Median and large doses of SM extracts significantly reduced hypoxia-induced media thickening in the SPAs (similar with diltiazem), recovered repaired ultrastructure injury, decreased HO-1 and iNOS levels, and increased eNOS expression in the SPAs. Conclusions Median and large doses of SM extracts play significant roles in inhibiting structural remodeling in rats with hypoxic pulmonary hypertension. These effects might attribute to the suppression of HO-1 and iNOS, and the promotion of eNOS expression under the conditions of hypoxia.

Tanshinones are a class of bioactive components in the traditional Chinese medicine salvia miltiorrhiza, and their biosynthesis and regulation have been widely studied. Current studies show that basic leucine zipper(bZIP) proteins regulate plant secondary metabolism, growth and developmental processes. However, the b ZIP transcription factors involved in tanshinone biosynthesis are ***, we conducted the first genome-wide survey of the b ZIP gene family and analyzed the phylogeny,gene structure, additional conserved motifs and alternative splicing events in S. miltiorrhiza. A total of 70 Smb ZIP transcription factors were identified and categorized into 11 subgroups based on their phylogenetic relationships with those in Arabidopsis. Moreover, seventeen Smb ZIP genes underwent alternative splicing events. According to the transcriptomic data, the Smb ZIP genes that were highly expressed in the Danshen root and periderm were selected. Based on the prediction of b ZIP binding sites in the promoters and the co-expression analysis and co-induction patterns in response to Ag^+ treatment via quantitative real-time polymerase chain reaction(qRT-PCR), we concluded that Smb ZIP7 and Smb ZIP20 potentially participate in the regulation of tanshinone biosynthesis. These results provide a foundation for further functional characterization of the candidate Smb ZIP genes, which have the potential to increase tanshinone production.
