sox9 in biliary atresia: New insight for fibrosis progression

Radiotherapy for head and neck cancer can cause serious side effects, including severe damage to the salivary glands, resulting in symptoms such as xerostomia, dental caries, and oral infection. Because of the lack of long-term treatment for the symptoms of xerostomia, current research has focused on finding endogenous stem cells that can differentiate into various cell lineages to replace lost tissues and restore functions. Here, we report that sox9^(+)cells can differentiate into various salivary epithelial cell lineages under homeostatic conditions. After ablating sox9^(+) cells, the salivary glands of irradiated mice showed more severe phenotypes and the reduced proliferative capacity. Analysis of online single-cell RNAsequencing data reveals the enrichment of the Wnt/β-catenin pathway in the sox9^(+) cell ***, treatment with a Wnt/β-catenin inhibitor in irradiated mice inhibits the regenerative capability of sox9^(+) cells. Finally, we show that sox9^(+) cells are capable of forming organoids in vitro and that transplanting these organoids into salivary glands after radiation partially restored salivary gland *** results suggest that regenerative therapy targeting sox9^(+) cells is a promising approach to treat radiation-induced salivary gland injury.

Analysis of intron sequence variability of the conservative HMG-box of sox9 genes in allotetraploids and their original parents

The sox genes of allotetraploids and their original maternal red crucian carp (Carassius caassius red var.) and original paternal common carp (Cyprinus carpio L.) were detected by PCR with the designed primers based on the conserved HMG-box sequence in different species. Sequencing of sox genes indicated that two sox9 genes (Atsox9a and Atsox9b) existed in allotetraploids, while only one sox9 gene existed in red crucian carp (Rcsox9a) and common carp (Ccsox9b) . All of the four sox9 genes contained an intron in the HMG-box. with the sizes of 413 bp, 703 bp, 401 bp and 714 bp, respectively. Moreover, the introns obeyed the rule of "GT-AG" . A high similarity was observed between introns of Atsox9a and Rcsox9a (94.4%), Atsox9b and Ccsox9b (97.8%). Interestingly, the deduced amino acid sequences of their corresponding exons all shared 100% identity. Thus, introns of the HMG-domain of sox9s in allotetraploids and their original parents have not only the length polymorphism but also intron variability. Our results provide significant molecular evidence for the origin and evolution of allotetraploids.

Photobiomodulation inhibits the expression of chondroitin sulfate proteoglycans after spinal cord injury via the sox9 pathway

Characterisation and Expression Analysis of sox9 in the Multiocellated Racerunner, Eremias multiocellata

sox9 is an important member of sox family which is involved in a variety of developmental processes including sex determination and gonadal differentiation. The cDNA of sox9 from multiocellated racerunner E. multiocellata was cloned using reverse transcription-polymerase chain reaction （RT-PCR） and rapid amplification of cDNA ends （RACE）. The sequence contains a 1497 bp open reading frame, which encodes a 498 amino acid protein with a predicted molecular weight of 55.45 kDa. Emsox9 displays high similarity to those of reptiles, and shows an overall amino acid identity of 〉82%. We also investigated the tissue-specific expression of Emsox9 mRNA by realtime quantitative PCR. sox9 mRNA is present in brain, heart, liver, kidney, gonads and muscle tissues of adult E. multiocellata, with the highest expression in brain and testis. The results indicate that sox9 may play important roles in some tissues during E. multiocellata neural and gonadal development.
