BACKGROUND The pathological complete response(ypCR)rate following neoadjuvant chemotherapy for advanced gastric cancer remains low and lacks a universally accepted treatment protocol.Immunotherapy has achieved breakthrough progress.CASE SUMMARY We report two female patients with gastric cancer defined as clinical stage cT4N1-2M0.Detection of mismatch repair protein showed mismatch repair function defect,and perioperative treatment with programmed death protein 1 inhibitor combined with S-1+oxaliplatin achieved ypCR.Surprisingly,the patients underwent clinical observation after surgery but developed different degrees of metastasis at~6 mo after surgery.CONCLUSION PD-1 inhibitor combined with chemotherapy provides a more strategic choice for comprehensive perioperative treatment of gastric cancer.

The SRY-related high mobility group(HMG)box(sox)transcription factors participate in many physiological processes of animal growth,development,and reproduction and are related to spermatogenesis in many species.However,the relationship between sox and spermatogenesis in Eriocheir sinensis is rarely reported.Here,we studied the role of Es-sox8 in the spermatogenesis of E.sinensis and its possible regulation mechanism.Immunofluorescence results demonstrated Es-sox8 signal in both cytoplasm and nucleus of spermatogonia,spermatocytes as well as spermatids,but not in mature spermatozoa.Hematoxylin and eosin staining showed a significant increase in the number of spermatozoa with abnormal nuclear morphology in vivo,described as prominent edges and corners,after the knockdown of Es-sox8 through RNA interference.This indicated a possible role of Es-sox8 in the nuclear deformation process in the spermatogenesis of E.sinensis.Analysis of the mRNA levels of Es-bone morphogenetic protein 2(BMP2)in the Es-sox8 knocked-down testis tissue revealed significantly decreased transcription of Es-BMP2.Chromatin immunoprecipitation results showed the binding of Es-sox8 protein to the promoter region of Es-BMP2.Thus,Es-sox8 can directly regulate the transcription of Es-BMP2 by activating the promoter of Es-BMP2 and thus affects the sperm nucleus deformation of E.sinensis.

RBPjk-dependent Notch signaling regulates both the onset of chondrocyte hypertrophy and the progression to terminal chondrocyte maturation during endochondral ossification. It has been suggested that Notch signaling can regulate sox9 transcription, although how this occurs at the molecular level in chondrocytes and whether this transcriptional regulation mediates Notch control of chondrocyte hypertrophy and cartilage development is unknown or controversial. Here we have provided conclusive genetic evidence linking RBPjk-dependent Notch signaling to the regulation of sox9 expression and chondrocyte hypertrophy by examining tissuespecific Rbpjk mutant（Prx1Cre;Rbpjkf/f）, Rbpjk mutant/sox9 haploinsufficient（Prx1Cre;Rbpjkf/f;sox9f/1）,and control embryos for alterations in sox9 expression and chondrocyte hypertrophy during cartilage development. These studies demonstrate that Notch signaling regulates the onset of chondrocyte maturation in a sox9-dependent manner, while Notch-mediated regulation of terminal chondrocyte maturation likely functions independently of sox9. Furthermore, our in vitro molecular analyses of the sox9 promoter and Notch-mediated regulation of sox9 gene expression in chondrogenic cells identified the ability of Notch to induce sox9 expression directly in the acute setting, but suppresses sox9 transcription with prolonged Notch signaling that requires protein synthesis of secondary effectors.

Objective： To compare the effect of neoadjuvant chemotherapy （NACT） with adjuvant chemotherapy （ACT） using oxaliplatin plus S-1 （sex） or capecitabine （CapeOX） on gastric cancer patients with D2 lymphadenectomy. Methods： This was a two-by-two factorial randomized phase Ⅱ-Ⅲ trial, and registered on ISRCTN registry （No. ISRCTN12206108）. Locally advanced gastric cancer patients were randomized to neoadjuvant sex, neoadjuvant CapeOX, adjuvant sex, or adjuvant CapeOX arms. Primary analysis was performed on an intention- to-treat （ITT） basis using overall survival （OS） as primary endpoint. Results： This trial started in September 2011 and closed in December 2012 with 100 patients enrolled. Treatment completion rate was 56%, 52%, 38% and 30% in the four arms, respectively. NACT group had fewer dropouts due to unacceptable toxicity （P=0.042）. Surgical complication rate did not differ by the four groups （P=0.986）. No survival signifcant difference was found comparing NACT with ACT （P=0.664; 5-year-OS： 70% vs. 74% respectively）, nor between the sex and CapeOX groups （P=0.252; 5-year-OS： 78% vs. 66% respectively）. Subgroup analysis showed sex significantly improved survival in patents with diffuse type （P=0.048）. Conclusions： No significant survival difference was found between NACT and ACT. sex and CapeOX had good safety and efficacy as neoadjuvant regimens. Diffuse type patients may survive longer due to sex.

Performance with Respect to Flue Gas Composition of a Combined Desulfurization/Denitration Process Using Powder-Particle Fluidized Bed

A new combined desulfarizatinn/denitraticon (Desox/DeNOx) procees was teeted in this study. The procees uses the so-called powder-partlcle fluidized bed (PPFB) as the major reactor in which a coarse DeNOx catalyst, several hundrsd micrometers in size, is fluidized by flue gas as the fluidization medium particles while a contlnuogsly supplied fine Desox sorbent, several to tens of micrometers in dianteter, is entrained with the flue gas. Ammonin for NOx reduction is fed to the bottom of the bed, thus, sox and NOx are simultaneously removed in the single reactor.By adopting a model gas, SO2-NO-HaO-N2-air, to simulate actual flue gas in a laboratory-scale PPFB, simultaneous SO2 and NO removals were explored with respect to various gas components of flue gas. It was found that the vaxlations of SO2 removal with concentrations (fractions) of oxygen, water vapor, SO2 and NO in flue gas are little affected by the simultaneous NOx reduction. However,the dependencles of NO removal upon such gas components are clveely related to the inter-actions between Desox sorbent and DeNOx catalyst.

Gastric carcinoma is a heterogeneous malignant disease involving genetic factors. To identify predictive markers for gastric cancer treatment in Chinese patients, we evaluated the association between polymorphisms of the gene encoding cytochrome P450 2A6 （CYP2A6） and outcomes of S-1 plus oxaliplatin （sox） chemotherapy treatment. Clinical data on 60 consecutive gastric cancer patients receiving sox regimen were collected prospectively. We sequenced all exons of CYP2A6 and a total of 22 different polymorphisms were detected in the present study. Comprehensive analyses of these genetic polymorphisms were performed to determine their association with both safety and efficacy of sox regimen. Our results showcd that polymorphisms of （＇ YP2A6 wcrc associated with the safety and efficacy of sox treatment. Among them, missense mutations CYP2A6 rs60823196 and rs138978736 could be possible risk 6actors （P 〈 0.05） for severe diarrhea induced by sox, whereas CYP2A6 rs138978736 could be a conceiwtble predictor for overall survival of patients treated with sox adjuvant chemotherapy. Further large-scale randomized prospective studies are warranted to confirm these findings.

Human induced pluripotent stem cells （hiPSCs） can self-renew indefinitely and have the potential to differentiate into all cell types in the human body, thus hold great promise in regen- erative medicine, drug screening and developmental biology studies. However, integrating retroviral （Takahashi et al., 2007） or lentiviral （Hockemeyer et al., 2008） gene delivery systems and the exogenous oncogene c-Myc （Takahashi et al., 2007; Hockemeyer et al., 2008） have been typically used in the reprogramming process, which could increase risks of insertional mutations or epigenetic abnormalities and conse- quently pose biosecurity concerns for basic research and clinical applications of hiPSCs.

Comparison of Extraction Techniques with Different Solvents for DDT Analogues in Sediments

Four techniques, soxhlet extraction (sox), ultrasonic extraction (USE), fluidized-bed extraction (FBE) and acce-lerated solvent extraction (ASE) with different solvents (methanol, hexane/acetone and acetonitrile) were used for theextraction of DDT analogues in sediments. Results revealed that the four extraction techniques had high recoveries (>86.0%) with low standard deviations (soxhlet extraction using hexane/acetone (1:1). Considering solvent- and time-consumption, level of automation,and environmental risk, accelerated solvent extraction with hexane/acetone (1:1) was better than the other extractiontechniques.
