Beyond boundaries:Neuroprotective effects of steroids and ecdysteroids in sh-sy5y cells-A systematic review

Steroids and ecdysteroids have been shown to exhibit a range of biological effects,including anti-inflammatory,anticancer,and *** systematic review aims to highlight the evidence-based neuroprotective and antioxidant effects of steroids and ecdysteroids in sh-sy5y neuroblastoma cells.A comprehensive literature search was conducted on May 11,2023,without publication source restrictions,using various electronic databases,including PubMed,Web of Science(WoS),Scopus,and Cumulative Index toNursing and Allied Health *** 103articles,only20 studies were included for investigating the neuroprotective effects of steroids and ecdysteroids in sh-sy5y cells exposed to oxidative stress or neurotoxic *** risk of bias and quality assessment of the included studies were evaluated in accordance with the Nature Publication Quality Improvement Project specific *** selected studies reported the antioxidant effects of the tested compounds on sh-sy5y cells and demonstrated their ability to scavenge free radicals and prevent lipid *** findings suggest that neurosteroids have potential therapeutic applications for the prevention and treatment of neurodegenerative diseases characterized by oxidative stress and neuronal *** studies have investigated the molecular mechanisms underlying the neuroprotective and antioxidant effects of steroids and ecdysteroids in sh-sy5y *** mechanisms include the activation of antioxidant enzymes,such as superoxide dismutase and glutathione peroxidase,and the modulation of various signaling pathways,including the phosphoinositide 3-kinase/protein kinase B and mitogen-activated protein kinase/extracellular signal-regulated kinase *** review provides evidence that the tested compounds have remarkable neuroprotective and antioxidant effects in human neuroblastoma sh-sy5y cells.

Overexpression of cytoglobin gene inhibits hypoxic injury to sh-sy5y neuroblastoma cells

A plasmid for cytoglobin expression, pAcGFP1-Cl-cytoglobin, was transfected into sh-sy5y cells. Cobalt chloride was used to establish a model of hypoxia. Western blotting indicated that cytoglobin was overexpressed and there was low expression of hypoxia-inducible factor-la in sh-sy5y cells after transfection. Following cobalt chloride-induced hypoxia, cytoglobin and hypoxia-inducible fac- tor-la expression gradually increased in sh-sy5y cells. Flow cytometry showed that with increas- ing duration of hypoxia, the proportion of normal cells significantly diminished in the transfected and non-transfected groups. The proportion of cells in the early stages of apoptosis increased. However, the proportion of apoptotic cells was significantly lower in the transfected group compared with the non-transfected group. These results demonstrate that cytoglobin and hypoxia-inducible factor-la are strongly up-regulated by hypoxia, and that there is a strong relationship between hy- poxia-inducible factor-la and cytoglobin during hypoxic injury.

Vanadyl complexes work with cinnamaldehyde in promoting cell viability under the β-amyloid burden in sh-sy5y neural cells

The Alzheimer＇s disease （AD） is one of the common cognitive disorders in the elderly. AD shares some similar pathological characters with diabetes mellitus （DM）, suggesting potential application of anti-diabetic agents, such as vanadyl complexes, in therapeutic treatment of AD. In the present work, we studied the effects of vanadyl acetylacetonate （VO（acac）2） and cinnamaldehyde （CA） on an AD model based on sh-sy5y neural cells. The experimental results showed that VO（acac）2 at sub-micromolar concentrations could improve the viability of neural cells with or without increased β-amyloid （Aβ） burden; and the combination of VO（acac）2 and CA showed an additive cell protection effects. Further investigation revealed that for sh-sy5y neural cells, VO（acac）2 could activate PPART-AMPK signal transduction and inhibit GSK 3β, one of the major kinases for Tau hyperphosphorylation. Meanwhile, CA could correct the abnormal mitochondrial morphology due to Aβ-induced excessive mitochondrial fission, thus restoring/enhancing the mitochondrial function. In addition, both VO（acac）2 and CA decreased intracellular reactive oxygen species （ROS） level and inhibited formation of toxic Aβ oligomers. Overall, VO（acac）2 might work with CA in improving the neural cell viability under the Aβ burden, suggesting application of vanadium metallodrugs in AD treatment.

The neurotoxic effects and influence of beta-amyloid peptide (Aβ)on membrane lipids and nicotinic acetylcholine receptors (nAChRs)in human sh-sy5y neuroblastoma cells were investigated in *** of the cultured cells to varying concentrations of Aβe- voked a significantly decrease in cellular reduction of MTT(3-(4,5-dimethylthiazol-2-yl) -2,5,diphenyl tetrazolium bromide),together with enhanced lipid peroxidation and protein oxi- *** reductions in the total contents of phospholipid and unbiquinone-10,as well as in the levels of the ? 3 and ? 7 subunit proteins of nAChRs were detected in cells exposed to Aβ.In contrast,such treatment had no effect on the total cellular content of *** these alterations,increased lipid peroxidation and decreased levels of cellular phospholipids were most sensitive to Aβ,occurring at lower *** addition,when sh-sy5y cells were pretreated with the antioxidant vitamin E,prior to the addition of Aβ,these alterations in neurotoxicity,oxidative stress,composition of membrane lipids and expression of nAChRs were par- tially *** findings suggest that stimulation of lipid peroxidation by Aβ may be involved in eliciting the alterations in membrane lipid composition and the reduced expression of AChRs asso- ciated with the pathogenesis of AD.

sh-sy5y human neuroblastoma cell line： in vitro cell model of dopaminergic neurons in Parkinson＇s disease

Objective To evaluate the human neuroblastoma sh-sy5y cell line as an in vitro model of dopaminergic （DAergic） neurons for Parkinson＇s disease （PD） research and to determine the effect of differentiation on this cell model. Date sources The data of this review were selected from the original reports and reviews related to sh-sy5y cells published in Chinese and foreign journals （Pubmed 1973 to 2009）. Study selection After searching the literature, 60 articles were selected to address this review. Results The sh-sy5y cell line has become a popular cell model for PD research because this cell line posses many characteristics of DAergic neurons. For example, these cells express tyrosine hydroxylase and dopamine-13-hydroxylase, as well as the dopamine transporter. Moreover, this cell line can be differentiated into a functionally mature neuronal phenotype in the presence of various agents. Upon differentiation, sh-sy5y cells stop proliferating and a constant cell number is subsequently maintained. However, different differentiating agents induce different neuronal phenotypes and biochemical changes. For example, retinoic acid induces differentiation toward a cholinergic neuronal phenotype and increases the susceptibility of sh-sy5y cells to neurotoxins and neuroprotective agents, whereas treatment with retinoic acid followed by phorbol ester 12-O-tetradecanoylphorbol-13-acetate results in a DAergic neuronal phenotype and decreases the susceptibility of cells to neurotoxins and neuroprotective agents. Some differentiating agents also alter kinetics of 1-methyl-4-phenyl-pyridinium （MPP~） uptake, making sh-sy5y cells more similar to primary mesencephalic neurons. Conclusions Differentiated and undifferentiated sh-sy5y cells have been widely used as a cell model of DAergic neurons for PD research. Some differentiating agents afford sh-sy5y cells with more potential for studying neurotoxiclty and neuroprotection and are thus more relevant to experimental PD research.

(±)-Crataegusnorin A(la/1b) and B(2a/2b),two pairs of rare 8,9’-epoxy-type norlignan enantiomers featuring a y-butyrolactone ring,were isolated from the fruit of Crataegus *** structures were determined via extensive spectroscopic ***-independent atomic orbital(GIAO) NMR chemical shift calculations,combined with the advanced statistical method DP4+were employed to establish the relative configurations of four ***,chiral separation was accomplished by chiral chromatographic column and the absolute configurations of the four compounds were unambiguously assigned by comparison between their experimental electronic circular dichroism curves with the quantum-mechanically calculated curves based on time-dependent density functional theory(TDDFT).All the isolates were evaluated fo r their neuroprotective activities against H2O2-induced cell injury in human neuroblastoma sh-sy5y *** results showed that two pairs of enantiomers 1a/1b and 2a/2b displayed diff;erent effect on neuroprotective *** them,compound 2a displayed the most potent neuroprotective effect Further flow cytometry analysis indicated that 2a could protect sh-sy5y cells from oxidative damage through inhibiting cell apoptosis.
