Background rhodococcus equi （R. equl） infection commonly occurs in grazing areas, especially in patients with AIDS or with T-lymphocyte immuno-deficiencies. Literature reviews revealed that cases radiologically and pathologically diagnosed of AIDS complicated by R. equi infection are rare. This study aimed to investigate the imaging features and Datholoqical basis of AIDS complicated by pulmonary R. equi infection.

Procalcitonin—A Useful Biomarker for Pneumonia Associated with rhodococcus equi?

Background: Procalcitonin, a precursor protein of the hormone calcitonin, is a sensitive marker for sepsis in human medicine, which is used for diagnosis of bacterial pneumonia in adults and neonates to initiate antibiotic therapy. Objectives: In this study, procalcitonin was evaluated as a potential biomarker for rhodococcus equi associated pneumonia. Methods: In four foals procalcitonin was measured at four time-points (day 0 before antibiotic therapy, day 1, 3 and 5/6 during therapy) in plasma using an equine specific ELISA. Inclusion criteria for the study were a positive sepsis score, ultrasonographic evidence of pulmonary abscesses in addition >8 cm, a positive microbiology out of tracheobronchial secretion and positive response to antibiotic treatment (azithromycin 10 mg/kg BDW 24q PO and rifampicin 10 mg/kg BDW 12q PO) within a weak including improvement of clinical status and reduction of ultrasonographic score rhodococcus equi.

几丁质脱乙酰基酶(chitin deacetylase,CDA)是一种将几丁质中的乙酰基团去除而生产壳聚糖及其衍生物的酶。所生产的壳聚糖及其衍生物可以应用于生物医药、食品加工等领域,具有广阔的发展前景。与热碱化学法相比,利用CDA来制备壳聚糖具有节约成本、产品质量均一等特点。本文以从土壤中筛选出的马红球菌rhodococcus equi所高产的CDA(ReCDA)为研究对象,对其进行异源表达、产酶条件优化和蛋白纯化,并对ReCDA的酶学性质及底物偏好性展开初步分析。利用基因工程技术将ReCDA分别在无CDA活性的*** BL21(DE3),*** Rosetta,Saccharomyces cerevisiae BY4741,Pichiapastoris GS 1 1 5 菌株中进行异源表达,并对每个异源表达菌株的产酶条件进行优化。本研究发现,在大肠杆菌异源表达菌株 *** BL21/pET28a(+)-ReCDA-His(6)和 *** Rosetta/pET28a(+)-ReCDA-His(6)中的最佳产酶条件为细胞初始OD600值为0.6～0.7,添加0.15 mM IPTG在22℃诱导48 h以上,酶活力可达到119.58 U/mL。在酿酒酵母异源表达菌株Saccharomyces cerevisiae BY4741/pYES2.0-ReCDA-His(6)中,最佳产酶条件为细胞初始OD660值约为1.0,在30℃条件下,0.2%的半乳糖添加量,以220 rpm震荡培养,每隔12 h补加等量的半乳糖诱导48 h以上,酶活力可达到17.25 U/mL。在毕赤酵母异源表达菌株Pichiapastoris GS115/pPIC3.5K-ReCDA-His(6)和 Picha pastoris GS115/pPIC9K-ReCDA-GST 中的最佳产酶条件为细胞初始的OD660值为1.0～2.0,2%甲醇添加量,在30℃,220 rpm条件下,每隔12h补加等量的甲醇诱导表达90h以上,酶活力达到75.85～85.68 U/mL。探究了 Ni-NTA亲和层析,阴离子交换层析和GST-谷胱甘肽亲和层析对ReCDA分离纯化的影响。优化了 ReCDA的分离纯化条件,最终纯化出ReCDA蛋白,相对分子质量约为33.8 kDa。对纯化的ReCDA进行酶学性质分析其最适pH为7.5,最适温度为35℃。此外,1 mM Zn2+对酶活力表现出明显的抑制作用。进一步分析了 ReCDA的底物偏好性,其中ReCDA在胶体几丁质、乙二醇几丁质、几丁质寡糖、壳聚糖(DAs,85%)和一些带有乙酰基的氨基酸中表现出明显的脱乙酰活性。本课题研究首次通过异源表达及蛋白纯化方式对新型ReCDA的理化性质进行分析,丰富了对几丁质脱乙酰基酶家族蛋白的研究,为生物酶法生产壳聚糖及衍生物的工业生产应用提供了实践基础。
