FTIR Spectroscopy in Cultural Heritage Studies: Non-destructive Analysis of Chinese Handmade Papers

Chinese handmade papers have been the carriers of paper-based documents, paintings and calligraphies, which are of great importance for historical and cultural heritage studies. This research developed the attenuated total reflectance-fourier transform infrared(ATR-FTIR) spectroscopic technique for the analysis of Chinese handmade papers. The test was applied to Chinese handmade papers taken from bark, hemp and bamboo groups, followed by papers from ancient books and arts collected in library and museum. Our study indicated tliat FTIR was an efficient analytical method to identify fiber types of library/museum papers of unknown origin. Moreover, information about degree of crystallinity and state of deterioration of cellulosic fibers was learned from FTIR spectra. This research provides a non-destnictive method to study the molecular structure of cellulosic fibers, and guides tlie selection of appropriate Chinese handmade papers for the restoration of cultural heritage objects.

Collagen from Tendon of Yezo Sika Deer (<i>Cervus nippon yesoensis</i>) as By-Product

Collagen from tendon of Yezo sika deer was prepared by limited pepsin digestion. The yield of collagen was very high;35.7% on the basis of lyophilized dry weight. The secondary structure of this collagen was different from that of porcine skin by ATR-FTIR analysis, although it was the same characteristics, e.g. SDS-PAGE, subunit composition, ther-mal behavior, as porcine collagen. Since taking up a problem of bovine spongiform encephalopathy infection in land animals such as calf or bovine, collagen from aquatic materials has been used in various industries. However, the present study indicates that tendon of Yezo sika deer as by-product of meat industry will have potential as an important collagen source for use in the foods, cosmetics, and medical fields.

Characterization of Acid-Soluble Collagen from Skins of Surf Smelt (Hypomesus pretiosus japonicus Brevoort)

Acid-soluble collagen was extracted from the skins of surf smelt and characterized. The yield of collagen was high about 24.0% on a dry weight basis. By SDS-polyacrylamide gel electrophoresis and CM-Toyopearl 650 M column chromatography, this collagen is a heterotrimer with a chain composition of α1α2α3. The denaturation temperature was 32.5℃, about 4.5℃ lower than that from porcine skin. Attenuated total reflectance-fourier transform infrared analysis showed that the percentage of secondary structural components in this collagen were 11% α-helix, 34% β-sheet, 19% β-turn, and 21% others. It suggests that the triple helical structure is present in the acid-soluble collagen from the skins of the surf smelt in comparison to that from the skin of porcine.
