Relationship between phenotypes of cell-function differentiation and pathobiological behavior of gastric carcinomas

AIM: To reveal the correlation between the functional differentiation phenotypes of gastric carcinoma cells and the invasion and metastasis by a new way of cell-function ***:Surgically resected specimens of 361 gastric carcinomas(GC) were investigated with enzyme-, mucin-, and tumor-related marker immunohistochemistry. According to the direction of cell-function differentiation, stomach carcinomas were divided into five functionally differentiated types. RESULTS: (1) Absorptive function differentiation type (AFDT): there were 82 (22.7%) patients including 76 (92.7%) aged 45 years. Sixty-nine (84.1%) cases belonged to the intestinal type. Thirty-eight (46.3%) expressed CD44v6 and 9 (13.6%) of 66 male patients developed liver *** 5-year survival rate of patients in this group (58.5%) was higher than those with the other types (P estrogen receptors (ER) (129, 81.7%) than other types (P<0.01). Ovary metastasis was found in 12 (19.4%) out of 62 female subjects. The patients with this type GC had the lowest 5-year survival rate (24.7%) among all types. (4) Specific function differentiation type (SFDT): 13 (3.6%) cases. Nine (69.2%) tumors of this type derived from APUD system, the other 4 (30.7%) were of different histological differentiation. Sixty per cent of the patients survived at least five years. (5) Non-function differentiation type (NFDT): 32 (8.9%) cases. Mast cell density and the context of clinicopathological parameters and expression of p185,estrogen receptor,and proliferating cell nuclear antigen in gastric carcinoma

AIM:To investigate the relationship between the mast cell density(MCD)and the context of clinicopathological parameters and expression of p185,estrogen receptor(ER), and proliferating cell nuclear antigen(PCNA)in gastric carcinoma. METHODS:Mast cell,p185,ER,and PCNA were detected using immunohistochemical S-P labeling *** cell was counted in tissue of gastric carcinoma and regional lymph nodes respectively,and involved lymph nodes(ILN)were examined as usual. RESULTS:MCD was significantly related to both age and depth of penetration(x^2=4.688,P21/0.03 mm^2 and MCD≤21/0.03 mm^2 in 100 patients;MCD in 1-6 ILN group patients was significantly higher than that in 7-15 ILN or>15 ILN group patients(u=6.881,8.055,P21/ 0.03 mm^2 and MCD≤21/0.03 mm^2 in 100 patients. CONCLUSION:Mast cell may have effect on inhibiting invasive growth of tumor,especially in the aged patients; The number of mast cells,in certain degree,may predicate the number of involved lymph nodes,which is valuable for assessment of prognosis;MCD was related to the expression of p185,ER,and PCNA in gastric *** suggests that mast cell accumulation may inhibit the proliferation and the dissemination of the gastric carcinoma. Comparison of hyaluronidase expression, invasiveness and tubule formation promotion in ER （-） and ER （＋） breast cancer cell lines in vitro

Background Hyaluronidase （Hyase） is an enzyme which hydrolyses hyaluronan （HA）, a large nonsulfated glycosaminoglycan. Several genes have been identified to code for hyaluronidases in humans. Its role has only recently been underlined in the invasion of prostate cancer, colonic cancer, and breast cancer. Moreover, the findings were in agreement with some experimental results which showed that HA-derived oligosaccharides had angiogenesis-promoting activity. All these findings prompted us to investigate factors that had been characterized as putative invasive factors in different human breast cancer-derived cell lines. Methods We selected two series of human breast cancer-derived cell lines whose expression of estrogen receptors （ER） was previously published. Hyaluronidase secretion in culture medium and expression of matrix metallo-proteinase （MMP）-9, cathepsin-D （cath-D） and vascular endothelial growth factor （VEGF） by cells were determined. We also investigated cell invasiveness in the Matrigel invasion assay, and studied the capability of cancer cells to promote in vitro formation of tubules by endothelial cells. Results ER（-） cells secreted significantly more hyaluronidase （P 〈0.001） and expressed significantly more VEGF （P 〈0.01）, MMP-9 （P 〈0.05） and cath-D （P 〈0.0001） than ER（＋） cells. Invasion through Matrigel by ER（-） Hyase（＋） cells was significantly higher than that by ER（＋） Hyase（-） cells （P 〈0.05）. In both cases, invasion was decreased by heparin （P 〈0.05）. When ECV-304 endothelial cells were co-cultivated in millicell chambers with cancer cells, ECV-304 cells were induced to form tubules. Tubule formation was demonstrated to be more prominent with ER（-） Hyase（＋） cells than with ER（＋） Hyase（-） cells （P 〈0.05）. Aim: To determine the expression of estrogen receptor-α (ERα) and estrogen receptor-β (ERβ) in prostate cancer (PCa) and to investigate the relationship between expression of ER and clinico-pathologic features of prostaric carcinoma. Methods: Thirty-two cases of PCa, 12 cases of normal prostate tissue (NOR) and 32 cases of benign prostate hyperplasia (BPH) were analyzed for the expression of ERα and ERβ by using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), and the products were sequenced. Results: The comparison of the normal, hyperplastic and tumor prostate tissues indicated an overexpression of ERa in tumor specimens (P<0.01) . On the contrary, the expression of ERβ was significantly reduced in tumor tissues compared with that in normal and hyperplastic specimens (P<0.01), suggesting that severe clinico-pathologic features of PCa were associated with lower ERβ expression. Spearmen analysis showed a significant negative correlation between ERβ expression and tumor stage and grade (P<0.05 ), and a significant positive correlation between ERα expression and tumor stage, grade, respectively (P<0.01) . Our analysis also showed that hormone-refractory PCa (HRPC), compare to hormone-dependent PCa (HDPC), displayed a decreased expression of ERβ (P<0.01) and an increased expression of ERα. Conclusions: The results suggest that ERα and ERβ may play an important role in the development of PCa. The loss or decrease of ERβ expression is associated with higher Gleason grade tumors and PCa with higher metastatic potential. Our data show that the loss of ERβ could be one of the key elements leading to uncontrolled growth of prostate epithelial cells.
