The Mn/Yb/Er triple-doped CeO_(2) nanozyme with enhanced oxidase-like activity for highly sensitive ratiometric detection of nitrite

Long-term excessive intake of nitrite(NO_(2)^(-))poses a great threat to human health,needing a simple and fast method to detect NO_(2)-in food.Herein,via a simple and feasible strategy,Mn/Yb/Er triple-doped CeO_(2) nanozyme(Mn/Yb/Er/CeO_(2))was synthesized for highly sensitive ratiometric detection of nitrite.By doping Mn,Yb,Er into CeO_(2) lattice structure,Mn/Yb/Er/CeO_(2) nanozyme showed enhanced oxidase-like activity,obtaining a higher density of oxygen vacancy and a higher ratio of Ce^(3+)to Ce~(4+)than that of CeO_(2).The 3,3,5,5-tetramethylbenzidine(TMB)can be effectively oxidized by Mn/Yb/Er/CeO_(2) to produce the oxidized TMB(ox TMB),showing a significant absorption signal at 652 nm.Additionally,nitrite can react with ox TMB to produce yellow diazotized ox TMB,which is accompanied by an elevated absorption signal at 445 nm and a decreased absorption signal at 652 nm.Thus,based on the oxidase-mimetic activity of Mn/Yb/Er/CeO_(2) and the diazotization reaction between NO_(2)^(-)and ox TMB,a ratiometric colorimetric assay was established for NO_(2)^(-)detection in food.Furthermore,by integrating Mn/Yb/Er/CeO_(2) with a smartphone,a colorimetric smartphone-sensing platform was successfully fabricated for visualization and quantitative detection of NO_(2)^(-).Notably,this two-detection mode showed excellent sensitivity,selectivity,reliability and practicability in monitoring the NO_(2)^(-)in real samples,impling its great potential for food safety.

Programmable Analytical Feature of ratiometric Electrochemical Biosensor by Alternating the Binding Site of Ferrocene to DNA Duplex for the detection of Aflatoxin B1

ratiometric biosensor is among the promising strategies for the advanced analysis;its applicability in multiple scenarios,however,is seriously limited for the fixed analytical feature.Here,we reported ratiometric electrochemical biosensors with programmable analytical feature by alternating the binding site of ferrocene(Fc)to DNA duplex for the detection of aflatoxin B1(AFB1).DNA duplex was synthesized with AFB1 aptamer and its complementary DNA(cDNA)with Fc molecular tagged on aptamer or cDNA.For analysis,aptamer-AFB1 complex stripped from the electrode,and free cDNA triggered hybridization chain reaction to produce double-stranded DNA for methylene blue(MB)absorption.In this way,redox currents of Fc(I_(Fc))and absorbed MB(I_(MB))at the electrode were collected to produce the ratiometric signal of I_(MB)/I_(Fc)as a yardstick for AFB1.When Fc was tagged on aptamer,IFc decreased with higher concentration of AFB1(C_(AFB1)),the resultant biosensor allowed detection with an On/Off(I_(MB)/I_(Fc))mode;when Fc was tagged on cDNA,I_(Fc) kept constant as C_(AFB1)varying,offering an On/Constant(I_(MB)/I_(Fc))mode for the detection.Consequently,biosensor with On/Off mode showed a linear range of 0.1—100 pg/mL with a limit of detection(LOD)of 0.0650 pg/mL,while biosensor with On/Constant mode offered a wider linear rang of 1—10000 pg/mL but a relatively poor LOD of 0.330 pg/mL.Our work revealed the vital importance of binding site of probes to DNA,and this principle can endow ratiometric biosensor with programmable analytical feature(e.g.,linear range,LOD),allowing detections in various scenarios.

A mitochondria-targeted fluorescent probe for ratiometric detection of hypochlorite in living cells

Hypochlorous acid（HOCl） plays a vital role in many physiological and pathological processes as one of reactive oxygen species（ROS）. Developing highly sensitive and selective methods for HOCl detection is of significant interest. In this work, we developed a benzothiazole based probe 1 for ratiometric fluorescence detection of hypochlorite in living cells. The probe can detect HOCl with high selectivity, fast response（within 30 s） as well as low detection limit（0.18 mmol/L）. Fluorescence co-localization studies demonstrated that probe 1 was a mitochondria-targeted fluorescent probe. Furthermore, confocal fluorescence images of He La cell indicated that probe 1 could be used for monitoring intracellular HOCl in living cells. Finally, test strips experiment suggests that the probe 1 can detect the hypochlorous acid in tap water accompanied by remarkable color change.

Hydrogen peroxide(H2O2)plays a significant role in regulating a variety of biological processes.Dysregulation of H2O2 can lead to various diseases.Although numerous fluorescent imaging probes for H2O2 have been reported,the development of H2O2 ratiometric fluorescent probe with large Stokes shift remains rather limited.Such probes have shown distinct advantages,such as minimized interference from environment and improved signal-to noise ratio.In this work,we reported a new pyrene-based compound Py-VPB as H2O2 fluorescent probe in vitro.The probe demonstrated ratiometric detection behavior,large Stokes shift and large emission shift.In addition,the probe showed high sensitivity and selectivity towards H2O2 in vitro.Based on these excellent properties,we successfully applied Py-VPB to the visualization of exogenous and endogenous H2O2 in living cells.Cell imaging study also showed that our probe was localized in the mitochondria.We envision that the probe can provide a useful tool for unmasking the biological roles of mitochondrial H2O2 in living systems.

The fluorescent chemosensors for detecting single anion have been largely synthesized. However, the simultaneous detection and degradation of multiple anions remain a major challenge. Hypochlorite ion（ClO-） and thiocyanate ion（SCN-） are commonly used in a range of industries and the daily life. Although ClO- and SCN- play critical roles in many processes, an excess level of ClO- and SCN- can lead to a wide range of diseases. Therefore, development of a method for accurate, simple, and rapid detection and degradation of SCN- and ClO- is significant in environmental, medicine and the life sciences. ratiometric fluorescent probes allow the measurement of emission intensities at two different wavelengths, which provides a built-in correction for the environmental effects. At present, only a few ratiometric fluorescent sensors have been reported for detection of ClO- with moderate success, and all of them can not work in pure water solution, which limit their potential application in environment and bio-logical systems. Moreover, there is not report ratiometric fluorescent sensor for detection SCN- or simultaneous detection of Cl O- and SCN- in water. Herein, a white emission nanoprobe was synthesized by Coumarin-Rhodamine CR1-Eu complex coordinated with dipicolinic acid（dpa）-PEG-Fe3O4 nanoparticles for the selective detection of Cl O- and SCN- ions on the controlling by a logic gate. This nanoprobe shows a marked rose red to violet emission color change in response to ClO-, a reversible violet to rose red emission color change in response to SCN-, high Cl O- and SCN- selectivity and sensitivity with a detection limit of 0.037 and 0.250 nM, respectively. Furthermore, the SCN- and ClO- can degrade simultaneously through the redox reaction between ClO-and SCN-. Due to exhibiting superparamagnetism at room temperature, this nanoprobe can be easily separated from the detected solution. The development of new methods for rapid and effective detection of biosulfite(HSO) is of great significance because of the key roles of HSOto human health. In this work, a new quinoline-based probe, WY, was designed and synthesized according to Knoevenagel condensation reaction. Due to the nucleophilic addition reaction between WY and HSOand the resulting broken conjugated structure, WY exhibits excellent specificity, high sensitivity and a rapid response towards biosulfite with "naked-eye" recognition and this superb recognition cannot be affected by the coexistence of other common ions. Notably, the demonstration of WY, as a ON-OFF fluorescence probe for the detection of exogenous biosulfite, was achieved in living animals including adult zebrafish and nude mice.

Naphthalimide-rhodamine based fluorescent probe for ratiometric sensing of cellular pH

The pH values of lysosomes in cancer cells is slightly lower than that in normal cells, which can be used to distinguish cancer cells from normal cells. According to this, a naphthalimide-rhodamine based fluorescent probe（hereafter referred to as RBN） with a pK_a of 4.20 was designed and synthesized for ratiometric sensing of cellular pH via fluorescence resonance energy transfer（FRET）, which can respond to different pH precisely through ratiometric fluorescence intensity（Ⅰ_（577）/Ⅰ_（540））. RBN can be employed to distinguish cancer cells from normal cells on the basis of different fluorescent response, in particular, RBN showed excellent water solubility and low cell toxicity, all these are quite significant for potential application in cancer diagnose and therapy.
