cDNA encoding 4 coumarate: CoA ligase (4CL) was isolated from the secondary developing xylem of Chinese white aspen ( Populus tomentosa ) by rt pcr for the first time, which was 1619 bp in length. The coding sequence and putative amino acid sequence showed 97.53% and 97.00% identity to that of Pt4CL1 in quaking aspen ( P. tremuloids ), respectively. Molecular analysis indicated that 4CL was encoded by multiple genes in *** , its mRNA was highly accumulated in xylem and the expression of 4CL revealed a biphasic pattern in one growing season, almost in phase with the expression of other related enzymes in lignin biosynthesis. Transgenic research showed that expression of antisense 4 CL cDNA led to the decreasing of lignin content in transgenic tobaccos, among which the average reduction was 10.3% and the highest could be up to 18.9%. These data suggested that 4 CL gene was a potential gene used in altering lignin biosynthesis by biotechnology for producing new materials of papermaking.

In order to study the molecular background and nematode-trapping mechanism of the *** was selected and extraction method for mycelia proteins of the fungus was optimized;Then proteomes of different predation periods were analyzed to determine the changes of the mycelium at the levels of protein by using i TRAQ technology,and verified by q *** main research results were as follows:i TRAQ technology was carried out for qualitative and quantitative *** results showed that 4244 proteins were *** the control group,compared with group of 12 h,the number of differential proteins in group of 72 h was the highest,with a total of 572;In the experimental group,there were 474 differential proteins identified between group of 12 h and 0 *** the same time point,compared with the control group,there were 296 differential proteins identified in group of 48 h,and 104 proteins were identified in 72 *** GO function of the differential proteins in each treatment group was *** results showed that the differential proteins in the fungus related catalytic activity,molecular binding function,transport activity,metabolic process,biological regulation,stress response,biolocation,carbon utilization,and substance synthesis showed significant *** KEGG enrichment analysis,it was found that the differential protein mainly involved in ubiquitin-mediated proteolysis,endoplasmic reticulum protein processing,sphingolipid metabolism,adhesion process,MAPK signaling pathway,AMPK signaling pathway,energy metabolism and carbon metabolism,peroxidase,oxidative phosphorylation and other eight *** pathways were related to the process of the fungus-feeding *** the above analysis,it was predicted that such as neutral ceramidase,sphingomyelin phosphodiesterase,tyrosinase,might play key role in the predation *** findings would supply the foundations for revealing the biological processes of the fungus,fungal acti
