rna recombination——INTRODUCING POLY(A) INTO Qβ PHAGE

Poly(A) molecule is inserted between two Qβrna molecules which have been partiallydegraded, one from the 3’-end and another from the 5’-end. E. coli HfrH is infected with thesemosaic rna molecules and a few recombinated phages carrying poly(A) are obtained. Analysisof the rna of 907 phage (one of these recombination phages) shows that the chain lengthof the poly(A) it contained is about 40.

Coleus（Coleus blumei） is an ornamental plant grown worldwide and can be infected by several viroids in the genus Coleviroid,family *** to now,six mainviroids that infect coleus have been reported:Coleus blumei viroid-1 to Coleus blumei viroid-6（CbVd-1～CbVd-6）,which size of references sequences range from 248～364 nucleotides（nts）.Coleus blumei viroids（CbVds） are good examples to illustrate the roles of recombination in the genetic variability and evolution of viroids considering that they are prone to frequent genome-wide *** instance,CbVd-2 and CbVd-6 are viroid chimeras—the former is made up of the right half of CbVd-1 and the left half of CbVd-3 and the latter is made up of the right half of CbVd-5 and the left half of ***-4 is an in vitro-generated ’inverse’ chimera of CbVd-2 which has been proved to be infectious after inoculated to coleus *** thus CbVd-3 is the most active viroid in the recombination event occurred among CbVds so far,and further study on CbVd-3 would be beneficial to elucidate the recombination mechanism of ***,as members of the family Pospiviroidae,it is noteworthy that all of the known coleus viroids share a common central conserved region（CCR）,which is known to be an important structural element for replication of viroids in the family Pospiviroidae（i.e.,processing and ligation）,and appears to be essential for the preservation of the secondary structure and maintenance of viroid *** addition,two complementary viroid-derived small rnas（vd-srnas） of 22 nts that were generated from the CCR of CbVds was identified through high-throughput sequencing analysis of small rnas derived from CbVds,suggesting an important role of CCR in vd-srna *** detection methods of Sap-direct RT-PCR and hybridization method using 8 CCR-probe,which were established both based on the CCR sequences of CbVds,would be helpful to find new viroid chimeras in ***,just like other viroi
