Nucleic acid enzymes are single-stranded nucleic acid molecules that catalyze specific chemical *** rnas found in nature such as group I intron and hammerhead ribozyme catalyze important biochemical reactions and are essential for cell ***,in vitro selections in the laboratory have produced artificial ribozymes and deoxyribozymes with various catalytic activities. Among them,rna-cleaving nucleic acid enzymes have been developed as novel molecular tools for biosensing and gene silencing ***,the intrinsic low biological stability of DNA and rna ultimately limit their functions in vivo. Here we describe the evolution of an rna-cleaving enzyme made of threose nucleic acid(TNA),which is resistant to nuclease degradation and considered an rna progenitor during early evolution of life on *** show that this TNA enzyme can specifically cleave the model mrna sequence of EGFR T790 M mutant gene while leaving wild-type substrate *** results demonstrate the capability of TNA to fold into a three-dimensional structure with catalytic property and to perform biologically useful functions.
