Stomata,which consist of paired guard cells,are known to have played crucial roles in the colonization of land by plants. Turgor-driven stomatal movement controls transpiration and gas exchange between plants and the environment. Several key genes and regulatory networks underlying stomatal development have been uncovered by molecular genetic analysis,however,much less is known about how signals involved in stomatal development are transmitted to rna polymerase ⅱ(Pol Ⅱ),which plays a central role in the transcription of m rna coding genes. Herein,we reported a partial loss-offunction mutant of the third largest subunit of nuclear DNA-dependent rna polymerase ⅱ(NRPB3). It exhibited an increased number of stomatal lineage cells and stomatal clusters. Similar stomatal phenotypes were observed in a weak allele of the second largest subunit of nuclear DNA-dependent rna polymerase ⅱ(NRPB2). These results suggested that Pol Ⅱ plays essential roles in stomatal development. Genetic analysis indicated that NRPB3 synergistically interacted with stomatal patterning and differentiation regulators. We also found physical associations of NRPB3 with two b HLH transcription factors,FAMA and INDUCER OF CBF EXPRESSION1(ICE1),indicating that NRPB3 serves as an acceptor for signals from transcription factors involved in stomatal development. Our findings highlight the surprisingly conserved activating mechanisms mediated by the third largest subunit of Pol Ⅱ in eukaryotes.

TBX5, as a member of the T-box-containing transcription factor family, encodes a protein of 518 amino acids and is expressed in the embryonic heart and developing limb tissues.1 The coding region of TBX5 cDNA is 1.5 kb with eight exons including the N-terminal portion, the DNA binding domain and C-terminal region. We reported that the abnormality in transcription level of the TBX5 gene might be the mechanism underlying human simple congenital heart disease in the absence of TBX5 mutations.

Metastatic breast cancer has high rates of relapse and mortality. The transcription factor hypoxia-inducible factor(HIF) plays a crucial role in breast cancer metastasis by inducing hundreds of the downstream target genes. rna polymerase II is essential for HIF-mediated transactivation. However, the mechanism by which rna polymerase ⅱ is loaded to HIF target genes remains unknown. Based on mass spectrometry analysis, we identified epigenetic remodeler CHD4 as a novel HIF regulator. CHD4 is highly expressed in invasive breast tumors and this overexpression is significantly correlated with poor clinical outcomes in patients with breast cancer. CHD4 depletion dramatically attenuates the tumorigenic potential of breast cancer cells in colony formation, migration and invasion in vitro, and suppresses breast tumor growth and metastasis in mice. To elucidate the underlying mechanism, we performed Co-IP, rna-seq, ChIP-qPCR and RT-qPCR assays, and found that CHD4 interacts with HIF complex at the hypoxia inducible elements(HREs) to provoke the expression of HIF target genes LOX, ANGPTL4, NDNF and VEGFA in a HIF-dependent manner. CHD4 interacts with and recruits rna polymerase II to the HREs, thereby promoting the formation of transcription preinitiation complex. Together, CHD4-HIF-rna polymerase ⅱ axis is critical for breast cancer progression and *** summary, CHD4 augments HIF-mediated breast cancer progression and metastasis, and provides a potential prognostic biomarker and therapeutic target for breast cancer.

Mechanism and factors that control HIV-1 transcription and latency activation

After reverse transcription, the HIV-1 proviral DNA is integrated into the host genome and thus subjected to transcription by the host rna polymerase ⅱ （Pol Ⅱ）. With the identification and characterization of human P-TEFb in the late 1990s as a specific host cofactor required for HIV-1 transcription, it is now believed that the elongation stage of Pol Ⅱ transcription plays a particularly important role in regulating HIV-1 gene expression. HIV-1 uses a sophisticated scheme to recruit human P-TEFb and other cofactors to the viral long terminal repeat （LTR） to produce full-length HIV-1 transcripts. In this process, P-TEFb is regulated by the reversible association with various transcription factors/ cofactors to form several multi-subunit complexes （e.g., 7SK snRNP, super elongation complexes （SECs）, and the Brd4-P-TEFb complex） that collectively constitute a P-TEFb network for controlling cellular and HIV-1 transcription. Recent progresses in HIV-1 transcription were reviewed in the paper, with the emphasis on the mechanism and factors that control HIV-1 transcription and latency activation.

Background: Elongator is a multiprotein complex that exhibits acetyltransferase activity. Results: In the subcomplex Elp4-6, Elp6 acts as a bridge to assemble Elp4 and Elp5, and each subunit adopts a similar RecA-ATPase-like fold. Conclusion: Subcomplex Elp4-6 formsa hexameric ring-shaped structure that is important for histone H3 binding. Significance: This finding may shed light on holo-Elongator complex assembly and its substrate recognition.
