Effects of Tualang honey in modulating nociceptive RESPONSES at the spinal cord in offspring of prenatally stressed rats

Objective: This study was done to determine whether Tualang honey could prevent the altered nociceptive behaviour, with its associated changes of oxidative stress markers and morphology of the spinal cord,among the offspring of prenatally stressed rats.Methods: Pregnant rats were divided into three groups: control, stress, and stress treated with Tualang honey. The stress and stress treated with Tualang honey groups were subjected to restraint stress from day 11 of pregnancy until delivery. Ten week old male offspring(n = 9 from each group) were given formalin injection and their nociceptive behaviours were recorded. After 2 h, the rats were sacrificed, and their spinal cords were removed to assess oxidative stress activity and morphology. Nociceptive behaviour was analysed using repeated measures analysis of variance(ANOVA), while the levels of oxidative stress parameters and number of Nissl-stained neurons were analysed using a one-way ANOVA.Results: This study demonstrated that prenatal stress was associated with increased nociceptive behaviour, changes in the oxidative stress parameters and morphology of the spinal cord of offspring exposed to prenatal stress; administration of Tualang honey reduced the alteration of these parameters.Conclusion: This study provides a preliminary understanding of the beneficial effects of Tualang honey against the changes in oxidative stress and neuronal damage in the spinal cord of the offspring of prenatally stressed rats.

Common mycorrhizal networks activate salicylic acid defense RESPONSES of trifoliate orange(Poncirus trifoliata)

Citrus canker, caused by Xanthomonas axonopo-dis pv. citri ('Xac'), is an important quarantine disease in citrus crops. Arbuscular mycorrhizal fungi (AMF) form symbiotic interactions with host plants and further affect their disease resistance, possibly by modulating the activity of salicylic acid (SA), a key phytohormone in disease resistance. Common mycorrhizal networks (CMNs) can interconnect plants, but it is not yet clear whether CMNs promote resistance to citrus canker and, if so, whether SA signaling is involved in this process. To test this possibility, we used a two-chambered rootbox to establish CMNs between trifoliate orange (Poncirus trifoliata) seedlings in chambers inoculated (treated) or not (neighboring) with the AMF, Paraglomus occultum. A subset of the AMF-inoculated seedlings were also inoculated with Xac (+AMF+Xac). At 2 d post-inoculation (dpi), compared with the +AMF-Xac treatment, neighboring seedlings in +AMF+Xac treatment had lower expression levels of the SA biosynthetic genes, PtPAL, PtEPS1, and PtPBS3, but higher SA levels, which attributed to the upregulation of PtPAL and PtPBS3 in treated seedlings and the transfer of SA, via CMNs, to the neighboring seedlings. At 4 dpi, the pathogenesis-related (PR) protein genes, PtPR1, PtPR4, and PtPR5, and the transcriptional regulatory factor gene, PtNPR1, were activated in neighboring seedlings of+AMF+Xac treatment. At 9 dpi, root phenylalanine ammo-nia-lyase activity and total soluble phenol and lignin concentrations increased in neighboring seedlings of+AMF+Xac treatment, likely due to the linkage and signal transfer, via CMNs. These findings support the hypothesis that CMNs transfer the SA signal from infected to neighboring healthy seedlings, to activate defense RESPONSES and affording protection to neighboring plants against citrus canker infection.

Enhancing the bake-hardening RESPONSES of a pre-aged Al-Mg-Si alloy by trace Sn additions

Effects of additions of trace Sn on the bake-hardening RESPONSES of a pre-aged Al-0.85 Mg-0.85 Si(in wt%)alloy were investigated through mechanical tests,differential scanning calorimetry,electrical resistivity and transmission electron microscopy.Results indicate that trace Sn additions reduced the number density of pre-aging clusters by inhibiting the formation of unstable counterpart during pre-aging treatment,leading to low strength and high supersaturation of solute atoms.In a subsequent paint-bake treatment,the presence of highly supersaturated solute atoms and high concentrated free vacancies moderated the activation energy barrier ofβ’’phase and thus kinetically accelerated the formation ofβ’’.Consequently,the trace Sn additions enhanced the bake-hardening RESPONSES of the pre-aged alloys significantly.The Sn-containing pre-aged Al-Mg-Si alloys with low strength and great bake-hardening RESPONSES hold promising potential for automotive body skin applications.

Surface energy-mediated fibronectin adsorption and osteoblast RESPONSES on nanostructured diamond

Nanostructured diamond have potential applications in many biomedical related fields and demonstrated extraordinary capacity to influence cellular RESPONSES. Studying the surface property of nanodiamond and its influence to protein adsorption and subsequent cellular RESPONSES along with the mechanism behind such capacity becomes more important. Here the role of surface energy associated with nanostructured diamond in modulating fibronectin and osteoblast(OB, bone forming cells) RESPONSES was investigated. Nanocrystalline diamond(NCD) and submicron crystalline diamond(SMCD) films with controllable surface energy were prepared by microwave-enhanced plasma chemical vapor deposition(MPCVD) techniques. Fibronectin adsorption on the diamond films with varied surface energy values was measured via the enzyme-linked immunosorbent assay(ELISA) and the relationship between the surface energy and fibronectin adsorption was studied. The result indicated that fibronectin adsorption on nanostructured surfaces was closely related to both surface energy and material microstructures. The spreading and migration of OB aggregates(each containing 30–50 cells) on the NCD with varied surface energy values were also studied. The result indicates a correlation between the cell spreading and migration on nanodiamond and the surface energy of nanostructured surface.

To the Editor: Irritable bowel syndrome (IBS) is a prevalent functional gastrointestinal disorder that presents as abdominal pain with altered bowel habits. The pathophysiologic mechanism of IBS is not well understood, although many hypotheses have been proposed, including visceral hypersensitivity, gastrointestinal dysmotility, lowgrade inflammation of the intestinal mucosa, and dysfunction of the brain-gut interaction. Dysfunction of the braingut interaction is thought to be involved in IBS because a considerable proportion of patients with IBS have some form of psychologic comorbidity, such as depression or anxiety. In addition, mental and psychologic problems increase the risk for IBS and the symptom severity of IBS. Recently, changes in the gut microbiota have also been suggested to contribute to both IBS and depression. Our previous study demonstrated significant altered gut microbiota profiles in patients with diarrhea-predominant IBS (IBS-D) and depression.

Epstein-Barr virus (EBV) infection is closely associated with nasopharyngeal carcinoma (NPC)and considered one of the major risk factors[1]. A limited number of viral proteins, such as latent membrane proteins (LMP1, LMP2) and EB nuclear antigen1 (EBNA1), are expressed in NPC cells[2].Recognition epitopes for CD8+and CD4+T cells were included in the LMP2 antigen and EBNA1 C-terminal region (amino acid No. 380-641), respectively[3,4].Both CD4+and CD8+memory T-cell RESPONSES were efficiently reactivated by EBNA1 and LMP2[5;6].

Centrifuge model test and numerical interpretation of seismic RESPONSES of a partially submerged deposit slope

Partially submerged deposit slopes are o ften encountered in practical engineering applications.Howeve r,studies on evaluating their stability under seismic loading are still rare.In order to understand the seismic behavior of partially submerged deposit slopes,centrifuge shaking table model tests(50 g) were employed.The RESPONSES of horizontal accelerations,accumulated excess pore pressures,deformation mode,and failure mode of the partially submerged deposit slope model were analyzed.In dynamic centrifuge model tests,EQ5 shaking event was applied numerically.The results indicated that in the saturated zone of the deposit slope,liquefaction did not occur,and the measured horizontal accelerations near the water table were amplified as a layer-magnification effect.It was also shown that the liquefaction-resistance of the deposit slope increased under multiple sequential ground motions,and the deformation depth of the deposit slope induced by earthquake increased gradually with increasing dynamic Ioad amplitude.Except for the excessive crest settlement generated by strong shaking,an additional vertical permanent displacement was initiated at the slope crest due to the dissipation of excess pore pressure under seismic loading.The result of particle image velocimetry(PIV) analysis showed that an obvious internal arc-slip was generated around the water table of the partially submerged deposit slope under seismic loading.

PXR variants:the impact on drug metabolism and therapeutic RESPONSES

The pregnane X receptor(PXR) plays an important and diverse role in mediating xenobiotic induction of drug-metabolizing enzymes and transporters.Several protein isoforms of PXR exist,and they have differential transcriptional activity upon target genes; transcript variants 3(PXR3) and 4(PXR4) do not induce target gene expression,whereas transcript variants 1(PXR1) and 2(PXR2) respond to agonist by activating target gene expression.PXR protein variants also display differences in protein–protein interactions; PXR1 interacts with p53,whereas PXR3 does not.Furthermore,the transcript variants of PXR that encode these protein isoforms are differentially regulated by methylation and deletions in the respective promoters of the variants,and their expression differs in various human cancers and also in cancerous tissue compared to adjacent normal tissues.PXR1 and PXR4 m RNA are downregulated by methylation in cancerous tissue and have divergent effects on cellular proliferation when ectopically overexpressed.Additional detailed and comparative mechanistic studies are required to predict the effect of PXR transcript variant expression on carcinogenesis,therapeutic response,and the development of toxicity.

Transcriptomic RESPONSES of Acinetobacter harbinensis HITLi 7~T at Low Temperatures

Acinetobacter harbinensis HITLi 7^T, a novel species in the genus Acinetobacter, was isolated from Songhua River in Harbin, China. It is a psychrotolerant heterotrophic nitrification bacterium that grows in temperatures from 2 ℃ to 35 ℃ with an optimal growth temperature of 8 ℃- 20 ℃[1]. The strain plays an important role in ammonium removal from source water and water sanitation improvement in cold areas. Biologically enhanced activated carbon (BEAC) filters immobilized with HITLi 7^T are used to treat source water in winter, exhibiting stronger ammonium removal capability and higher microbial stability during BEAC treatment at low temperatures[2]. However, the low temperature adaptation mechanism of HITLi 7^T has not been clearly elucidated yet.

In situ[2+3] cycloaddition synthesis,crystal structures,strong SHG RESPONSES and fluorescence properties of three novel Zn coordination polymers

We report on the synthesis of two acentric and one centrosymmetric Zn metal-organic complexes with 3-tetrazolenethyl pyridine spacers obtained in situ by hydrothermal routes.X-ray diffraction structural analysis reveals that they have the same equivalent nodes but with dissimilar topologies.The two acentric frameworks[Zn(Tzmp)Cl]n(1)and([Zn(Tzmp)Br]n(2),HTzmp=3-tetrazolemethyl pyridine)are isomorphism which exhibit an acentric 3D framework with(10,3)-b net called"ths",while the centro symmetric complex([Zn(Tzmp)N3]n(3))features a distinctive 2D sheet with Shubnikov hexagonal plane net.Photo-luminescent studies suggest the ligand-field strength of coordinated negative ions(Cl^->Br^->N3^-)has ordered adjusting effects on the emission redshift.The second harmonic generation(SHG)measurement shows that compounds 1 and 2 are nonlinear optically active,with SHG RESPONSES being 2/3 and half of the standard potassium dihvdrogen phosphate(KDP),respectively.
