Dear Editor,Multiple myeloma(MM)is characterized by the excessive production of monoclonal immunoglobin.As a lot of novel agents were approved during the last two decades,deeper responses and longer survival have been achieved in MM patients,such as minimal residual disease(MRD)measured via flow cytometry(FCM).l The development of oligoclonal bands(OB)and immunoglobin isotype(Ig)switch predicted favorable outcomes after high-dose therapy followed by autologous stem cell transplantation(ASCT)in newly diagnosed MM(NDMM)patients.2 However,even if the appearance of OB is linked to a better response,MRD negativity requires further discussion.

The effect of a pulsed electric current on the residual stress evolution of metal materials has been investigated.It was found that the surface and internal residual stresses in the as-quenched samples were reduced dramatically by electropulsing.A large number of experimental data show that the residual stress reduction is proportional to the initial residual stress and related to the material properties and electropulsing parameters.Under the combined actions of drift electrons,Joule heating,and residual stress,the dislocation mobility was enhanced,resulting in plastic strain and the decrease in residual stress.Drift electrons played a unique role in the electropulsing treatment,acting as an additional force pushing dislocations forward.The dislocations ultimately accumulated at a grain boundary,forming a parallel arrangement.Finally,the phenomenological equation of the residual stress evolution under electropulsing was derived from the experimental data.

A formula was brought forward to correlate residual strain with hardness measured by nanoindentation method. Using the formula, residual strains in prestrained aluminum alloy samples were evaluated. In order to compare with the calculated strain, such values were also measured by strain gauge method. The results show that the differences of the residual strain obtained by nanoindentation and strain gauge method are not more than 8%, which shows that the formula is suitable for characterizing residual strain of aluminum alloy.

Acrylates have been widely used in the synthesis of pharmaceutical polymers. The quantitation of residual acrylate monomers is vital as they are strong irritants and allergens, but after polymerization, are relatively inert, causing no irritation and allergies. Poly(ethylene oxide)(PEO) hydrogels were prepared using pentaerythritol tetra-acrylate(PETRA) as UV crosslinking agent. A simple, accurate, and robust quantitation method was developed based on gas chromatographic techniques(GC), which is suitable for routine analysis of residual PETRA monomers in these hydrogels. Unreacted PETRA was initially identified using gas chromatography–mass spectrometry(GC–MS). The quantitation of analyte was performed and validated using gas chromatography equipped with a flame ionization detector(GC–FID). A linear relationship was obtained over the range of 0.0002%–0.0450%(m/m) with a correlation coefficient(r2)greater than 0.99. The recovery( 4 90%), intra-day precision(%RSD o 0.67), inter-day precision(%RSD o2.5%), and robustness(%RSD o1.62%) of the method were within the acceptable values. The limit of detection(LOD) and limit of quantitation(LOQ) were 0.0001%(m/m) and 0.0002%(m/m), respectively.This assay provides a simple and quick way of screening for residual acrylate monomer in hydrogels.

For researching the residual stress distribution characteristics of the different large thickness titanium alloy joints by electron beam welding,under heat treatment or non-heat treatment conditions,the changes in residual stress distribution characteristics of electron beam welding joints,which thickness is 50 mm,were measured using blind-hole method with local layer-by-layer removal.Effect of post-welding heat treatment on the residual stress distribution also studied.The results show that blind-hole method with local layer-by-layer removal is suitable to measure residual stresses in thick plate,and good to reflect the welded residual stress distribution of large gradient.residual stress of the test samples in the weld and heat affected zone has high stress level,and internal stress dramatic changes with thickness in this areas.After heat treatment,the samples' stress distributions were significantly reduced.The samples' transverse and longitudinal residual stress are basically the same.Homogenization is obvious.

In drug discovery research, residual solvent measurement is an integral part of purity analysis for synthesis of a drug candidate before it is used for toxicity testing. This is usually carried out using gas chromatography(GC)with direct injection sample introduction. This method requires testing compounds to be soluble at high concentrations( > 50 mg/mL, usually in DMSO) to achieve acceptable sensitivity, a hurdle which is not always achievable for some samples such as cyclic peptides and oligonucleotides. To overcome the limitation associated with the direct injection approach, a new method using the Chromatoprobe thermal extraction device was developed for quantifying residual solvents of drug discovery compounds. This method not only consumes significantly less material(less than 1 mg), but also shows higher sensitivity than the direct injection approach.In addition, because no diluent is required with the Chromatoprobe thermal extraction, all residual solvents can be detected and measured without further method optimization. In our study, we compared data from GC residual solvent analysis using the Chromatoprobe solid sample introduction to those of the direct injection method for seven in-house samples. Our results showed a good agreement between the data from these two sample introduction methods. Thus, the Chromatoprobe sample introduction method provided a samplesparing alternative to the direct injection method for the measurement of residual solvents in drug discovery.This method can be particularly useful for residual solvent analysis in samples that are available only in limited amounts, poorly soluble, and/or unstable in the diluents used for the direct injection method.

Background: Several studies have shown that detection of minimal residual disease (MRD) in acute myeloid leukemia (AML) is an independent prognostic factor. This study aimed to evaluate the significance of dynamic MRD pretransplantation on outcome of AML patients receiving allogeneic hematopoietic stem cell transplantation (allo?HSCT). Methods: We retrospectively analyzed 145 consecutive AML patients undergoing allo?HSCT in complete remission status between June 2013 and June 2016. MRD was determined with multiparameter flow cytometry after the first and second courses of chemotherapy and pre?HSCT. Results: In matched sibling donor transplantation (MSDT) settings, patients with positive MRD had higher cumulative incidence of relapse (CIR) than those without MRD after the first (32.3 ± 9.7% vs. 7.7 ± 3.1%, χ2 = 3.661, P = 0.055) or second course of chemotherapy (57.1 ± 3.6% vs. 12.5 ± 2.7%, χ2 = 8.759, P = 0.003) or pre?HSCT (50.0 ± 9.7% vs. 23.0 ± 3.2%, χ2 = 5.547, P = 0.019). In haploidentical SCT (haplo?SCT) settings, the MRD status at those timepoints had no significant impact on clinical outcomes. However, patients with persistent positive MRD from chemotherapy to pre?HSCT had higher CIR than those without persistent positive MRD both in MSDT and haplo?SCT settings. Patients with persistent positive MRD underwent MSDT had the highest relapse incidence, followed by those with persistent positive MRD underwent haplo?SCT, those without persistent MRD underwent haplo?SCT, and those without persistent MRD underwent MSDT (66.7 ± 9.2% vs. 38.5 ± 6.0% vs. 18.8 ± 8.7% vs. 12.0 ± 1.0%, χ2 = 20.763, P < 0.001). Multivariate analysis showed that persistent positive MRD before transplantation was associated with higher CIR (hazard ratio [HR] = 1.69, 95% confidence interval [CI]: 1.200–2.382, P = 0.003), worse leukemia?free survival (HR = 1.812, 95% CI: 1.168–2.812, P = 0.008), and overall survival (HR = 2.354, 95% CI: 1.528–3.627, P < 0.001). A novel process monitoring method based on convolutional neural network(CNN)is proposed and applied to detect faults in industrial process.By utilizing the CNN algorithm,cross-correlation and autocorrelation among variables are captured to establish a prediction model for each process variable to approximate the first-principle of physical/chemical relationships among different variables under normal operating conditions.When the process is operated under pre-set operating conditions,prediction residuals can be assumed as noise if a proper model is employed.Once process faults occur,the residuals will increase due to the changes of correlation among variables.A principal component analysis(PCA)model based on the residuals is established to realize process monitoring.By monitoring the changes in main feature of prediction residuals,the faults can be promptly detected.Case studies on a numerical nonlinear example and data from two industrial processes are presented to validate the performance of process monitoring based on CNN.
