Random mutagenesis is commonly used to study gene function. The screening of mutants exhibiting specific pheno- types assists in the identification of phenotype-related genes. In the current study, we isolated Antarctic bacteria, and developed a transposon Tn5 mutagenesis system. A total of 26 strains were isolated from seawater and freshwater near Antarctic King Sejong Research Station, King George Island. Six psychrobacter strains were identified as psychrophilic, with optimal growth tempera- tures of 10~C or 15~C psychrobacter cryohalolentis PAMC 21807 with a high growth rate at 4~C was selected for transposon mutagenesis. Tri-parental conjugation with a plasmid containing Tn5 produced 13 putative recombinants containing the selectable marker. Genomic Southern hybridization confirmed Tn5 existed as episomes for seven recombinants, and for a single recombinant, Tn5 was integrated into the genome of psychrobacter cryohalolentis PAMC 21807. The result indicates that the mutagenesis method, although successful, has a relatively low rate. The psychrophilic bacteria isolated in this study may be a useful resource for studying cold adaptation mechanisms, and the mutagenesis method can be applied to genetic analysis.

Abstract Heat shock proteins （Hsps）, produced by organisms under high temperature stimulation, play important roles in protein folding, translocation, and refolding/degradati0n. In this study, we investigated the expression level of the GrpE Hsp gene Hsp845 of psychrobacter sp. G under different temperature and salinity stresses by quantitative real-time PCR and western blotting, respectively. At both transcriptional and translational levels, Hsp845 gene expression was induced by high temperature （30~C） and inhibited by low temperatures （0~C and 10~C）. Hsp845 expression was also induced both by the absence of salt （0%0） and high salinity （90%0 and 120%o） at the transcriptional level, but was only induced by high salinity （90%0 and 120%o） at the translational level. In a combined stress treatment, Hsp845 was more sensitive to high temperature than to salinity at both transcriptional and translational levels. The increase in the translational-level expression of Hsp845 lagged behind that at the transcriptional level, and Hsp845 maximum expression was also higher at the transcriptional than at the translational level. In the absence of salt, transcriptional- and translational-level expressions exhibited opposite patterns, suggesting that the underlying mechanism requires further study.

根据南极适冷菌psychrobactersp.G的基因组完成图,对其假定蛋白基因PSYCG_10180全长进行了基因克隆、序列分析与异源表达,并利用qRT-PCR和western blotting技术对其在不同温度/盐度胁迫条件下的表达特征进行了研究。序列分析表明,与蛋白PSYCG_10180相似性较高的均为假定蛋白;相似性最高的已知功能蛋白为蓝光传感蛋白(WP_010201745.1),二者的相似性仅有37%。在mRNA水平上,基因PSYCG_10180的表达被低温(0℃)显著诱导,在2,12h均被不同盐度显著抑制;在温度/盐度协同胁迫下,2h内低温均能诱导其表达,而高温低盐(30℃,15)则抑制其表达。在蛋白水平上,不同温度胁迫下,蛋白PSYCG_10180的表达量在2h变化不大,12h时均被诱导;在不同盐度胁迫下,该蛋白的表达在12h均被诱导;在温/盐协同胁迫下,除在2h被高温低盐(30℃,15)诱导外,其余条件下该蛋白的表达均被抑制。比较分析表明,在mRNA水平上,基因PSYCG_10180的表达更容易受温度的影响;在蛋白水平上,盐度对该基因表达的影响则要大于温度;在温盐协同胁迫时,盐度起主导作用。冻融试验表明,表达该假定蛋白可显著提高重组菌株的冻融存活率。本研究表明,假定蛋白基因PSYCG_10180可能在菌株psychrobacter sp.G的低温等环境适应性中起着重要作用。

以南极适冷菌psychrobacter sp.G为研究对象,利用qRT-PCR和Western blot技术从转录水平和翻译水平对冷激蛋白基因Csp2039在不同温度/盐度胁迫条件下的表达特征进行了研究。在转录水平上,qRT-PCR分析表明,冷激蛋白基因Csp2039的表达于6h时显著被低温(0℃,10℃)和高温(30℃)诱导;低盐(0)胁迫下,基因Csp2039的表达先被诱导后被抑制,在盐度为15的胁迫下,基因Csp2039的表达均被诱导,高盐(90,120)时基因Csp2039的表达在6h时显著被诱导;在温度和盐度协同胁迫条件下,温度为0℃时,无论盐度高低,基因Csp2039的表达均显著上升,而高温(30℃)时,除2h被低盐诱导以外,其余条件下均显著被抑制。在翻译水平上,Western blot分析表明,冷激蛋白Csp2039的表达被低温(0℃,10℃)显著诱导,但高温(30℃)对其抑制并不明显;在低盐(0,15)胁迫下,Csp2039蛋白的表达均被诱导,高盐(90)胁迫下,Csp2039蛋白的表达先被抑制,然后逐渐升高;在温度和盐度协同胁迫条件下,低温(0℃)时,当盐度为15时,Csp2039蛋白的表达被诱导,盐度为90时,Csp2039蛋白的表达量被显著抑制;而高温(30℃)时,该蛋白在盐度15时6h的表达量最大,盐度90时于6h其表达被显著抑制。比较分析表明,在转录水平上基因Csp2039表达量的变化更容易受温度影响,而在翻译水平上盐度对Csp2039蛋白的影响作用大于温度;基因Csp2039在翻译水平上对温度/盐度胁迫的应答时间要迟于转录水平的。

目的对来自深海沉积物中的海洋细菌psychrobacter submarinus 1A01998的次级代谢产物进行研究。方法采用大孔吸附树脂、硅胶、ODS和Sephadex LH-20凝胶等柱层析色谱分离方法,对海洋细菌psychrobacter submarinus 1A01998进行次级代谢产物的分离纯化,根据1H NMR、13C NMR和MS等波谱数据分析并结合相关文献对化学结构进行鉴定。结果从深海细菌psychrobacter submarinus 1A01998的发酵粗提物中分离得到4个化合物,分别鉴定为环（L-脯氨酸-L-酪氨酸）（1）、环（D-脯氨酸-L-酪氨酸）（2）、3-吲哚甲醛（3）、2＇-O-甲氧基尿嘧啶核苷（4）。结论化合物1~4均为首次从该属中分离得到,其中化合物1、2为环二肽类化合物。

[目的]研究南极菌B-3对黄瓜的诱导抗病作用。[方法]以黄瓜幼苗为材料,研究南极菌B-3胞外寡聚糖的分子量、单糖组成,及对黄瓜叶中防御酶的诱导作用与黄瓜白粉病的诱导抗病性。[结果]B-3胞外寡聚糖经高效凝胶渗透色谱测定其分子量为2112D,气相色谱分析由甘露糖和葡萄糖2种单糖组成;0.3%、0.5%和0.8%的B-3胞外多糖分别对黄瓜幼苗喷施不同时间后,黄瓜体内的几丁质酶,β-1,3-葡聚糖酶,苯丙氨酸解氨酶(PAL),过氧化物酶(POD)的活性均较对照有不同程度的提高,且与0.5%的壳聚糖具有相似的诱导效果;同时,0.5%的B-3多糖能够显著地降低黄瓜白粉病的病情指数,防效达24.49%。[结论]B-3寡聚糖有望开发为新型抗病诱导子。
