Luminal acidification in the epididymis is an important process for the regulation of male fertility. Low pH and low bicarbonate concentration are among key factors that keep spermatozoa in a dormant state while they mature and are stored in this organ. Although significant bicarbonate reabsorption is achieved by principal cells in the proximal regions of the epididymis, clear and narrow cells are specialized for net proton secretion. Clear cells express very high levels of the vacuolar proton pumping ATPase （V-ATPase） in their apical membrane and are responsible for the bulk of proton secretion. In the present paper, selected aspects of V-ATPase regulation in clear cells are described and potential pathologies associated with mutations of some of the V-ATPase subunits are discussed. （Asian J Androl 2007 July; 9： 476-482）

Background Distal renal tubular acidosis(dRTA)is a kidney tubulopathy that causes a state of normal anion gap metabolic acidosis due to impairment of urine *** review aims to summarize the etiology,pathophysiology,clinical findings,diagnosis and therapeutic approach of dRTA,with emphasis on genetic causes of *** sources Literature reviews and original research articles from databases,including PubMed and Google *** searching was performed to identify additional studies about *** dRTA is characterized as the dysfunction of the distal urinary acidification,leading to metabolic *** pediatric patients,the most frequent etiology of dRTA is the genetic alteration of genes responsible for the codification of distal tubule channels,whereas,in adult patients,dRTA is more commonly secondary to autoimmune diseases,use of medications and *** with dRTA exhibit failure to thrive and important laboratory alterations,which are used to define the *** oral alkali and potassium supplementation can correct the biochemical defects,improve clinical manifestations and avoid nephrolithiasis and *** dRTA is a multifactorial disease leading to several clinical *** and laboratory alterations can be corrected by alkali replacement therapy.

Organic acid content is one of the most important factors influencing fruit *** predominant organic acid in most pear cultivars is malic acid,but the mechanism controlling its accumulation remains *** this study,by comparing gene expression levels and organic acid contents,we found that the expression of PbPH5,which encodes a P_(3A)-ATPase,is highly correlated with malic acid accumulation in four different pear species,with correlation coefficients of 0.932**,0.656*,0.900**,and 0.518*(*,P<0.05;**,P<0.01)for Pyrus bretschneideri Rehd.,*** Linn.,*** Nakai.,and *** Maxim.,***,the overexpression of PbPH5 in pear significantly increased the malic acid *** contrast,silencing PbPH5 via RNA interference significantly decreased both its transcript level and the pear fruit malic acid content.A subcellular localization analysis indicated that PbPH5 is located in the ***,a phylogenetic analysis indicated that PbPH5 is a PH5 homolog gene that is clustered with the Petunia hybrida,Malus domestica,and Citrus reticulata *** together,these findings suggest that PbPH5 is a functionally conserved ***,the accumulation of malic acid in pear fruit is at least partly related to changes in the PbPH5 transcription levels.

AIM:To test whether the expression and activity of H,K-ATPase in parietal cells would be affected by cigarette smoke ***: Extracts of cigarette smoke were administered into mice by gastric gavage (5 mg/kg body weight/day) for 3 d or in drinking water for 7 or 14 d. For the latter, each day a mouse consumed 5 mL water containing extracts of two cigarettes, on average. Control littermate mice received only vehicle. To compare the amount of H,K-ATPase in control and smoke-treated mice, the stomach was processed for Western blotting and immunohistochemical analysis using monoclonal antibodies specific for α- or β-subunits of H,K-ATPase. The p-nitrophenylphospatase activity assay was used as a measurement for K-dependent H,K-ATPase ***: Probed transblots showed an increase in the amount of H,K-ATPase in smoke-treated mice which was confirmed by immunohistochemistry and was found to be due to increased amounts of protein per parietal cell rather than an increased parietal cell number. The increase in the amount of H,K-ATPase was associated with an enhancement of its enzymatic activity. K-dependent activity in control and smoke-treated mice was significantly different (respectively, 0.12 μmol/mg vs 0.27 μmol/mg per minute, P<0.05).CONCLUSION: Administration of cigarette smoke extract is associated with an increase in the amount and activity of H,K-ATPase and hence, smokers are susceptible to development of peptic ulcer.
