Effects of Different Broth Enrichment upon Phage Magnetoelastic Biosensor for Fast Detecting Low <i>Salmonella</i>Counts on Problematic produce

According to the FDA Bacteriological Analytical Manual (BAM) for Salmonella identification in produce, two pre-enrichment steps with 48 hours of incubation are the golden procedures. Lactose broth is recommended for the first pre-enrichment step medium for leafy greens, and the universal pre-enrichment (UP) broth is for tomatoes. However, the suggested broths were evaluated to have the maximum performance using the culture-dependent methods, and may not be applied to other methods, such as biosensor detection platform. A wireless bacteriophage magnetoelastic (ME) biosensor has been recently developed for real-time or rapid detection of food-borne pathogens in various foods. This affinity-based biosensor utilizes a phage oligonucleotide as the probe to capture target bacteria. In this study, the efficiencies of different pre-enrichment media for early detection of low Salmonella on spinach leaves and tomatoes use ME biosensors to shorten detection time. Four broths of modified peptone water, Lennox broth (LB), lactose broth, and UP broth were selected in this study. Various pre-enrichment times for ME biosensor detection were investigated. After spiking 4 cfu/g Salmonella on the tomatoes surfaces, the phage biosensor was able to detect Salmonella within 5 hours of pre-enrichment comparing to 24 hours in the FDA procedures. For Salmonella spiked spinach leaves, the same medium showed Salmonella positive within 7 hours. This study demonstrated that LB broth is the best medium to shorten pre-enrichment time to pass Salmonella number detection thresholds for ME biosensor detection in spinach and tomatoes when comparing to FDA procedures.

Objectives:Foodborne disease outbreaks linked to fruits and vegetables have been increasing in occurrence worldwide;therefore,the aim of this study was to identify the reported foodborne outbreaks associated with fruit and vegetable consumption in Brazil from 2008 to *** And Limitations:Thirty produce related outbreaks resulted in 2926 illnesses,347 hospitalizations,and no *** bacterial pathogens were identified as etiological *** these,Salmonella was the most frequent(30 per cent of outbreaks)followed by Staphylococcus aureus(23.3 per cent),Escherichia coli(10 per cent),Bacillus cereus(6.6 per cent),and thermotolerant coliforms(3.3 per cent),whereas etiological agents could not be determined for 26.6 per cent of *** most common food vehicles implicated in outbreaks were generically named as fruits and vegetables(46.6 per cent of outbreaks).The term salad was used generically and specifically like salads(two outbreaks),raw/cooked salads(four outbreaks),vegetable salad,tropical salad,Caesar salad,and raw salad of cabbage and *** one outbreak was related exclusively to fruit(fruit pulp),whereas other outbreaks were related to cooked carrot,lettuce,cucumber,watermelon/cabbage,and chard/*** sources and issues related to the future control of produce-related foodborne disease outbreaks are discussed.

4D products are basic mode of digitized products that are made by combining model remote sensing with GIS technology, and have already brought everybody’s high recognition. Coal mining makes great destroy to land of coal mining area, and ecological environment becomes worse, therefore land reclamation of mining area has already turned into investigated emphases. 4D meaning is briefly introduced, and its six characteristics are expounded in this paper. Production technique and application status of 4D are summarized, and existed problems are analyzed in developed aspect. Land reclamation of mining area is a complex system engineering, and application of 4D products can be impenetrated in whole process. Otherwise 4D has broad application scope in land management of mining area and mine information construction. Compound of remote sensing and DTM can be apply to solve mine survey, monitor of surface and marginal slope and so on.

The objective of this study was to assess the condition of the road network within the district with a view to find out if and how they affect the transportation costs. Four sets of primary and secondary data on the district’s road surface types, road condition mix, cost of transportation of farm produce and humans on the road network were collected, collated and subjected to statistical analysis using a Completely Randomized Design. The results indicated that the road conditions had high significant effects on the transportation costs of both human and agricultural produce. The road network of the district consists of 21 roads with a total length of 176.6 kilometers out of which 8 were classified as poor, 7 as fair and only 6 as good. The highest agricultural produce transportation cost of 1.46 per tonne-kilometer was obtained from 3 of the poor roads while the lowest cost of 0.86 per tonne-kilometer was obtained from only 1 of the 6 good roads. The highest passenger transportation cost of 0.3 per passenger kilometer was obtained for 1 of the 8 poor roads while the lowest cost of 0.1 per passenger kilometer was obtained for all the 6 good roads and 4 of the fair roads. In conclusion, transportation cost of passengers on the poor and fair roads was 2 - 3 times as high as the cost of transportation on the good roads. Transportation cost of agricultural produce on the poor roads was 70% higher than it was on the good roads.

This research examined small farmers’ operational choice in converting to organic farming in the Southern region of the United States. A logistic model fitted to survey data from responses of 456 produce growers found the key determinants of the conversion choices of farmers and offered quantitative impacts of the major influential factors retained in the fitted model. Among them, barriers to the conversion were linked to farmer’s age, risk aversion, years of farming, and the lack of education as well as low yield of organic farming;nevertheless, the model also revealed the existence of stimuli reflecting on access to farmers markets, peer exemplars, and operational scale. The confluence of variables retained partially explained a slow but steady conversion to organic farming in the United States. Besides the broad implication for policy making in the agricultural sector, this study found that organic farming was closely tied to small farms and organic farming could be a back road to the future of small US farms.
