genetic diversity and population genetic structure of Python bivittatus in China

The Burmese python （Python bivittatus） has recently suffered large population declines in the wild in China due to illegal capture, overexploitation, deforestation and the loss of its natural habitat. Greater knowledge of the genetic diversity and structure of wild P. bivittatus populations is needed to help ensure its effective management. In this study, we investigated the genetic diversity and population genetic structure of wild P. bivittatus in China in detail. 109 P. bivittatus individuals from five distribution areas in Guangdong （GD）, Guangxi （GX）, Hainan （HN）, Fujian （FJ） and Yunnan （YN） province of China were collected, and their genetic structure and diversity were analyzed. Eight highly polymorphic microsatellite loci were utilized to reveal high levels of genetic diversity in the P. bivittatus population. genetic diversity was highest in GX, and lowest in GD. All geographic populations demonstrated a bottleneck effect indicating recent population decline. Fst and AMOVA analyses revealed that there was moderate genetic differentiation among the five populations, and that only 10.59 % of total genetic diversity occurred among populations. Fst values between pop- ulations were positively correlated with their geographical distances. genetic structure analyses revealed that the HN, GX and GD populations, which were geographically closest, were assigned to a genetic cluster, while the YN and FJ populations constituted a single cluster, respectively.

BACKGROUND Colorectal cancer(CRC)ranks third in terms of incidence and second in mortality *** CRC,the silencing of mismatch repair genes,including the mutL homolog 1(hMLH1)has been linked to microsatellite instability(MSI),the lengthening or shortening of microsatellite *** limited data have been presented so far on the link of hMLH1 methylation and MSI in Southeast Asia populations with sporadic CRC,and on its clinical *** To investigate the significance of the MSI status and hMLH1 methylation in CRC Filipino *** Fifty-four sporadic CRC patients with complete clinical data were included in this *** DNA from CRC tumor biopsies and their normal tissue counterparts were profiled for MSI by high resolution melting(HRM)analysis using the Bethesda Panel of Markers(BAT25,BAT26,D2S123,D5S346,and D17S250).hMLH1 methylation screening was performed using bisulfite conversion and methylation specific polymerase chain *** analysis was conducted to calculate their associations to clinicopathological characteristics and survival relevance(Kaplan-Meier curves and the log-rank test).RESULTS hMLH1 methylation was observed in 9%and 35%of CRC and normal samples,*** incidence of consistently methylated hMLH1 found in both normal and CRC was noticed for relation to location of tumor(P<0.05).As for MSI status,D2S123 the most common unstable microsatellite and MSI-high(MSIH)was the most common MSI profile,counted for 46%and 50%of normal and CRC tissues,*** presence of MSI-low(MSI-L)and microsatellite stable(MSS)was 43%and 11%for normal,and 31%and 19%for CRC *** mean month of patients’survival was shorter in patients whose normal and tumor tissues had methylated compared to those with unmethylated hMLH1 and with MSI-H compared to those with MSI-L/MSS(P<0.05).This was supported by significant difference in Kaplan-Meier with log-rank *** data indicated that hMLH1 methylation

A harmonisation approach to traditional Chinese medicine registration in Asian countries

The traditional Chinese medicine(TCM)industry is critical to not only for public health but also for economic *** to the European Union(EU)directives,under the EU framework for(traditional)herbal medicinal products,herbal medicines with long history of use can be registered in ***,there is a condition in this directive in which only those that have at least 10e15 years in the EU market are accepted for registration and/or marketing *** author opinion,the condition of 10-15 years of use in EU countries set within the EU regulatory framework is with consideration of the genetic differences which can result in variation in adverse drug responses among different world *** this concept in mind,it is reasonable to project the principal of the EU directive to the Asian countries where TCM is *** like China,Singapore,Japan and South Korea that have well established drug registration framework are in best position in executing the best practice and facilitate harmonization of registration for TCM within the ***,the registration process itself allows more safety and efficacy data to be collected systemically before and after product registration/marketing *** are valuable information for future drug *** therapeutic value of TCM is limitless,it has been left out in the EU regulatory framework,and the opportunity for it to be expanded and carried forward in modern medicines is shadowed by the limited number of TCM that are qualified to be registered under the EU regulatory *** early establishment of a harmonized risk-based registration process for TCM in Asian countries is *** will strengthen the database to substantiate the history of safe use and further preserving and expanding the therapeutic values of TCM within and beyond the Asian region.

Glomerella Leaf Spot(GLS) has been in an outbreak since 2011 and severely affects apple production(Malus domestica Borkh.) in *** pathogen that causes GLS is Glomerella cingulata(Stoneman) Spauld.& ***,which is epidemic in regions with humid climates from May to October each *** spreads more than 200 km per year,and most apple-producing areas have been invaded in the five years since *** investigate the origin,evolution and diversity of *** for better GLSmanagement,we performed whole genome sequencing of *** isolate 030206 and developed 25 simple sequence repeat marker(SSR) markers according to the SSR motifs identified in the genome *** the 25 SSRmarkers,we analyzed thegenetic diversity and population genetic structure of 96 *** isolates collected from eight prevalent GLS areas;Chiping,Feicheng,Haiyang,Muping,Qixia,Yishui,Yiyuan,and Zhanhua in Shandong province,*** expected heterozygosities(H) for the populations were 0.351,0.240,0.471,0.559,0.556,0.077,0.452,and 0.036,respectively,with a mean of 0.343,and the Shannon indices(I) were 0.560,0.385,0.811,1.100,1.032,0.140,0.662,and 0.051,respectively,with a mean of *** gene flow index(Nm) was *** 96 *** isolates were divided into three subpopulations,denoted GC1,GC2 and ***1 contained 32 isolates,most of which were from the western areas of Shandong province,whereas GC2 contained 18 isolates and GC3 contained 46 isolates,most of which were from the eastern areas of Shandong *** addition,the western genetic population,GC1,contained a few isolates from eastern collection sites,such as Haiyang,Muping and Qixia,whereas few isolates contained in GC2 and GC3 were from western *** results indirectly show that the GLS pathogens in western areas were originally from eastern areas of Shandong province.

Human needs on protein, especially those kinds protein derived from animal, are increasing day by day along with the increasing of human population on earth. Various aspect on livestock production requires more attention from the farmer. Nowadays due to advance biotechnology many genetically control aspects of livestock can be improved based on interest. Previous researches investigated that Beta-defensin have an important function on animal host defense system. However, there is limited study on Beta-defensin gene especially in goat, and the role of Single Nucleotide Polymorphism (SNP) as a factor corelated to genetic diseases or phenotypic traits in animal. Thus, on this bases that we carried out this research.Twelve (12) Chinese goat breeds were used on a series of gene amplification. SNP confirmation test was done and a single nucleotide mutation was found from T to C, between 1349-1363 bp (exon 2) within the goat's Beta-defensin sequence of the population used. population genetics analysis for each breed revealed that C allele as mutant allele common with samples from Sichuan province. This allele occurred more frequent in three of six Sichuan populations. F-statistic analysis on each breed resulted to negative FIS value per population ranging from -0.029 in BR population to -1.000 in LZ population, and there were only 2 populations having positive FIS value 0.202 and 0.235 NJ and CM population respectively. Nm value resulted from this research was 2.0022. Gene diversity value (He) ranged from 0.510 highest in JT population to 0.054 lowest in BR population. All of this condition might likely be caused by inbreeding and or small size of the researched population.An in-depth research needs to be done to investigate the effect of inbreeding, native blood deterioration, and to find which factors are responsible for disequilibrium within the said population.
