Diversified Application of Barcoded plato(plato-BC) Platform for Identification of Protein Interactions

Proteins usually associate with other molecules physically to execute their functions.Identifying these interactions is important for the functional analysis of proteins.Previously,we reported the parallel analysis of translated ORFs(plato)to couple ribosome display of full-length ORFs with affinity enrichment of mRNA/protein/ribosome complexes for the “bait”molecules,followed by the deep sequencing analysis of mRNA.However,the sample processing,from extraction of precipitated mRNA to generation of DNA libraries,includes numerous steps,which is tedious and may cause the loss of materials.Barcoded plato(plato-BC),an improved platform was further developed to test its application for protein interaction discovery.In this report,we tested the antisera-antigen interaction using serum samples from patients with inclusion body myositis(IBM).Tripartite motif containing 21(TRIM21)was identified as a potentially new IBM autoantigen.We also expanded the application of plato-BC to identify protein interactions for JQ1,single ubiquitin peptide,and NS5 protein of Zika virus.From plato-BC analyses,we identified new protein interactions for these “bait”molecules.We demonstrate that Ewing sarcoma breakpoint region 1(EWSR1)binds to JQ1 and their interactions may interrupt the EWSR1 association with acetylated histone H4.RIO kinase 3(RIOK3),a newly identified ubiquitin-binding protein,is preferentially associated with K63-ubiquitin chain.We also find that Zika NS5 protein interacts with two previously unreported host proteins,par-3 family cell polarity regulator(PARD3)and chromosome 19 open reading frame 53(C19orf53),whose attenuated expression benefits the replication of Zika virus.These results further demonstrate that plato-BC is capable of identifying novel protein interactions for various types of “bait”molecules.

Not Athenian or a Stranger:The Veiled Critique of Aristotle in plato’s Laws

plato’s last dialogue,the Laws,occupies an anomalous position within his larger body of work.An individual identified as the“Athenian stranger”replaces Socrates and reverses key Socratic teachings,most notably by endorsing tyranny.Scholars conclude that plato abandoned his earlier political recommendations in favor of a more pragmatic vision.In that case,the Laws should be treated as plato’s definitive work,the ultimate statement of his thought,when in fact,much more attention is paid to earlier dialogues,particularly the Republic.The problem is resolved and the true significance of the Laws revealed when the text is read as plato’s ironic critique of his brilliant-but-rebellious student,Aristotle.Reasoning from Aristotelian premises,the Athenian stranger arrives at conclusions that platonists and Aristotelians alike would find unpalatable or absurd.The alleged rupture between plato’s earlier and later work disappears.The esoteric writings that are thought to have been the product of Aristotle’s later career are shown to have emerged from ideas that plato himself was familiar with and rejected.

plato the Democrat？ Some Thoughts on the Politics of the Laws

In the Laws （which plato calls his ＂second-best society＂） plato asserts that the best attainable form of society will combine the better features of autocracy and democracy. The democracy will be one where aidos （＂respect＂） will be a prominent feature, as will be the rule of laws underpinned by the belief that God, not man, is the measure of all things. Unlike in the Republic, the accumulation of wealth and property will be the right of all citizens, including rulers. But it will operate under strict limits： a maximum of four quanta of property will be allowed by any citizen, while a minimum of one （which will provide a good life though perhaps not a rich one） will be guaranteed. The affinity of such a view with that of John Rawls is striking. The article ends with a brief interview between the reader and plato, in which some of the above issues are discussed.

plato on Necessity and Disorder

In the Timaeus, plato makes a distinction between reason and necessity. This distinction is often accounted for as a distinction between two types of causation： purpose oriented causation and mechanistic causation. While reason is associated with the soul and taken to bring about its effects with the good and the beautiful as the end, necessity is understood in terms of a set of natural laws pertaining to material things. In this paper I shall suggest that there are reasons to reconsider the latter part of this account and argue for a non-mechanistic understanding of necessity. I will first outline how the notion of necessity is introduced in the dialogue. Next I will show how a mechanistic account of necessity fails to capture plato＇s purpose of treating it as a causal factor; and, finally, I will argue that this purpose is better understood as an attempt, on plato＇s part, to account for the causal origin of disorder and irrationality, an origin articulated in terms of a pre-cosmic situation and the notoriously difficult notion of the third kind.

Can Thought Become Sensible? Reflections on the Aesthetic Theories of plato and Hegel

Hegel was highly influenced by plato’s philosophy,as appears in his Lectures on the History of Philosophy.The German philosopher rightly attributed to plato an“aesthetic”theory meant as a search for the meaning of Beauty(the idea of the Beautiful).For Kant the Sublime played the same role that Beauty did for the Greek philosopher,leading the human soul to grasp the super-sensible,as we read in his Critique of the Judgment of Taste.Contrary to what happened in plato,in Hegel’s own aesthetic theory,the investigation was not focused on Beauty in general,but on artistic Beauty,the characters of the beautiful work of art.If plato and Hegel highlight the dialectical-metaphysical function of Beauty,the judgment that they express on art is very different.However,they share an important conception:Beauty is as an intermediate and connecting reality,that functions as a link between thought and sensitivity.

plato＇s Attempts at Defining Sophistry

In the Sophist, plato offers seven definitions of a sophist. In this paper I will consider the relationship between these definitions, and examine how the first six definitions contribute to the final definition, and how the final definition, in turn, incorporates elements of the first six. Through this examination, we can also have a better grasp of plato＇s method of division, and plato＇s aporetic approach to philosophy.

The Republic of plato is a philosophical treatise describing the ideal city-state,a self-sufficient civil entity in the world of Ideas(Forms)that enables both individuals and the society to thrive.Elements of the Republic can be tracedwoka in modern policymaking.plato’s ethical and political philosophy consists of a standpoint of reflection towards contemporary challenges,one of whom is admittedly COVID-19 vaccination.Few countries have made vaccination obligatory for the whole of the population,while many countries consider making vaccination mandatory for particular high-risk groups and front line workers.In response to the growing debate on the necessity and the ethical integrity of mandatory vaccination against COVID-19,this paper discusses the matter in the context of plato’s Republic and provides relevant recommendations.

Socrates' discussions in plato's early dialogues are predominantly concerned with value-concepts. The discussions often start with the social roles which Socrates5 partners play and the virtues associated with those roles. They react to a widespread uncertainty concerning the decline of moral as well as a reassessment of values, all of which took place in the course of the fifth century. plato's Socrates tried to prove that one's life should be guided by standards such as true justice. In this context, he also referred to Hesiod, who had advanced a new concept of justice in reaction to a private problem, but also in an effort to contribute to a change of values in the society of his time (the 8th century). Thus, to some extant Hesiod can be regarded as a forerunner of those like plato who tried to establish fixed norms which are separated from personal interests and should be observed in order to live a good life. I shall argue that Hesiod's position itself is based on a transformation of an ancient concept and influenced later poets like Aeschylus, Sophocles and, despite his criticism, even plato.

Principle, Knowledge, and Personality： Some Reflections on ＂the Good＂ according to plato

This paper focuses on plato＇s ＂Form of the Good,＂ or ＂the Good,＂ with an interest in plato＇s riddle that ＂the Good is the One.＂ Unlike the traditional approach to explaining the Good in the Republic as ＂rational order＂ or a unity of Forms, this ]paper argues that the Good is the unique transcendent principle, like the apex of a hierarchy, but does not encompass the whole structure. According to its Ontological position, its multiple facets （functions） include the Ontological foundation of uniting ＂to be＂ and ＂ought to be,＂ the ultimate source of knowledge, and the Ideal goal of uniting the common good and individual goods. The practical dimension of the Good is highlighted in exploring the lifelong study of the Philosopher-Kings and their political personality. It is also pointed out that ＂sudden enlightenment＂ plays an important role in their path toward the Good. Finally, this paper proposes that the Good should be the a priori beginning of education and the end of the practice of virtues in the community.

plato Revealed: Alexander of Aphrodisias and His Philosophical Historiography

Alexander of Aphrodisias’s Commentary on Aristotle’s Metaphysics is an important testimony to understand plato’s philosophy.In fact,Alexander uses some lost Aristotelian books,especially a work On the Good,from which we learn that plato’s metaphysics is adialectical metaphysics,founded on an original opposition of two principles that shapes the whole reality—these principles being the One and the indefinite Dyad.Sensible things participate in ideas(they receive their being from ideas)and the intermediate mathematical entities lie between these two realities.However,ideas can be traced back to ideal numbers and the principles of ideal numbers are the One and the indefinite Dyad.Thus,these principles constitute their metaphysical foundation of ideas and,through the ideas,of the whole reality.
