Inheritance of Angular Leaf Spot [phaeoisariopsis griseola （Sacc.） Ferr] Resistance in Common Bean （Phaseolus vulgaris L,） Population Developed from Kablanketi x Mexico 54

The genetic resistance to angular leaf spot （ALS） caused by phaeoisariopsis griseola in the common bean cultivar Mexico 54 was investigated on disease reactions in parental, F1, F2 and backcross generations derived from crosses between a resistant cultivar Mexico 54 and a susceptible cultivar Kablanketi under screen house conditions. The heritability （h2） estimate was as high as 0.719 indicating a successful transfer of ALS resistance among progenies and thus selection can be performed in early generation. High heritability coupled with high expected genetic advance of 39.5% is considered to be more useful in predicting the outcome of selecting the best individuals. Chi-square values were computed to determine whether the observed ratios for disease reactions deviated from expected Mendelian ratios for a single, dominant gene controlling resistance to angular leaf spot in common bean. Based on the resistance of the F2, and the backcross generation to the resistant parent, a 3 resistant： 1 susceptible segregation ratio in the F2 and a 1 resistant： 1 susceptible segregation ratio in the backcross generation to the susceptible parent was obtained implying that resistance to the isolate ofphaeoisariopsis griseola is governed by a single, dominant gene.

Angular leaf spots of green bean(Phaseolus vulgaris L.) were observed in organic greenhouses in *** lesions were gray,irregular to angular,Lesions on petioles were *** in vivo were curved cylindrical to obclavate with one to four sepeta and measured 46.2-50.8μm×7.6-8.0μ*** conidiophores were 219-222μm high and clustered together to form synnemata measuring 41.4 to 44.7μm in *** pathogen was isolated by placing conidia from diseased leaves onto potato dextrose agar(PDA).Colonies measuring 11-12 mm in diameter composed of dense,dark olive mycelium developed after incubation in the dark at 25℃for 25 *** causal agent was tentatively identified as phaeoisariopsis griseola(Sacc.) Ferraris,the cause of angular leaf spot of bean,and molecular identification were done by *** of the isolate was tested on Phaseolus vulgaris and Vigna unguiculata with conidia obtained from the second subculture of 14-day-old colonies on *** suspensions of 2×10 conidia per ml were sprayed onto the upper and lower surfaces of the first trifoliolate leaves of six runner bean plants,18 days after *** and control plants (sprayed with distilled water) were placed in a growth chamber with a 12-h photoperiod at 24±2℃and 95 to 100%relative humidity and 48 h later moved to the *** symptoms were evaluated 18 days after *** control plants were healthy,all inoculated plants showed symptoms similar to those observed in the *** fungus that was consistently reisolated from lesions in the inoculated plants was identified as phaeoisariopsis griseola on the basis of fungal morphology,symptoms produced on *** our knowledge,even thousgh there are many report of this disease in other provinces of China,this is the first report of angular leaf spot occurring on *** in Shandong,and its pathogenicity on Vigna unguiculata indicated that phaeoisariopsis griseola is a potential threat to Leguminosa
