Caryophyllene-type sesquiterpenoids and α-furanones from the plant endophytic fungus pestalotiopsis theae

Two new caryophyllene-type sesquiterpenoids, pestathenols A(1) and B(2) and one new α-furanone, pestatheranone A(6), along with five known compounds(3–5, 7 and 8) have been isolated from the crude extract of the plant endophytic fungus pestalotiopsis theae. Their structures were unambiguously established by extensive spectroscopic analyses. The absolute configuration of the 5,6-diol moiety in 1 was assigned using Snatzke’s method. Compounds 1 and 2 showed weak cytotoxicity against HeLa cell line.

pestalotiopsis theae grown in different solid-substrate fermented cultures can produce structurally diverse and biologically active secondary metabolites. Pupukeanane chlorides existing in this fungus featured by complex structures and Diels-Alder biosynthetic routes. In current study,two new diphenyl ethers,pestalotethers E-F(1-2),two new pestalolides(3-4),together with three known compounds(5-7) were isolated from the fungus pestalotiopsis *** structures were established by spectroscopic *** 1-2 are the rare fungal metabolites of diphenyl *** 4 showed cytotoxic activity against the human tumor lines,including HL60 and HEP-3 B,with IC values of 0.9 and 4.29 μM,respectively.

Tea gray blight disease is an important disease of the tea tree,which can harm tea seedlings,as well as the shoots,fresh leaves and mature leaves of trees for various growth *** disease can cause the tea plants weakened and severethe production loss of tea *** gray blight disease is one of the most important diseases in Guizhou tea *** this work,the pathogenic fungus was isolated,purified from the diseased tea leaves,and confirmed the pathogenicity of the pathogen according to Koch’s *** isolated strain of DX-1 was further identified by the morphology and molecular *** study indicated that DX-1 represents the features as following;on PDA medium,the colonyis obviously wheel round like,margin regular of the colonies;Reverse represents light yellow colony with white or colorless *** observation using the optical microscope and SEM indicated that theconidia represent fusi form with 4 diaphragms and 2 thin ends,and 2-3 appendages and1 appendage exist onthe top and the tail end of the cell,*** with（16.07±1.24） μm×（4.23±0.20） μm is colorless in the early stage,and the 3 septum near the center change brown in the late *** genes of EF-1α,β-tubulin and LSU were further amplified,purified and sequenced,and then the phylogenetic trees were constructed using MEGA6.0 software according to Neighbor-joining *** results indicated that DX-1 strain was closely related to pestalotiopsis ***1（KJ623232.1） and YN1 B4（KU252565.1） against two phylogenetic treesof EF-1α and β-tubulin;and then clustered together with *** strain 3.919 2（JN940840.1） in a branch,and closely related to *** stain 3.909 8（JN940825.1） for the phylogenetic tree of LSU.
