Exosomes derived from bone marrow mesenchymal stem cells protect the injured spinal cord by inhibiting pericyte pyroptosis

Mesenchymal stem cell(MSC)transplantation is a promising treatment strategy for spinal cord injury,but immunological rejection and possible tumor formation limit its *** therapeutic effects of MSCs mainly depend on their release of soluble paracrine *** are essential for the secretion of these paracrine *** marrow mesenchymal stem cell-derived exosomes(BMSC-EXOs)can be substituted for BMSCs in cell ***,the underlying mechanisms remain *** this study,a rat model of T10 spinal cord injury was established using the impact ***,30 minutes and 1 day after spinal cord injury,the rats were administered 200μL exosomes via the tail vein(200μg/mL;approximately 1×106 BMSCs).Treatment with BMSC-EXOs greatly reduced neuronal cell death,improved myelin arrangement and reduced myelin loss,increased pericyte/endothelial cell coverage on the vascular wall,decreased bloodspinal cord barrier leakage,reduced caspase 1 expression,inhibited interleukin-1βrelease,and accelerated locomotor functional recovery in rats with spinal cord *** the cell culture experiment,pericytes were treated with interferon-γand tumor necrosis factor-α.Then,Lipofectamine 3000 was used to deliver lipopolysaccharide into the cells,and the cells were co-incubated with adenosine triphosphate to simulate injury in ***-treatment with BMSC-EXOs for 8 hours greatly reduced pericyte pyroptosis and increased pericyte survival *** findings suggest that BMSC-EXOs may protect pericytes by inhibiting pyroptosis and by improving blood-spinal cord barrier integrity,thereby promoting the survival of neurons and the extension of nerve fibers,and ultimately improving motor function in rats with spinal cord *** protocols were conducted with the approval of the Animal Ethics Committee of Zhengzhou University on March 16,2019.

Mechanism of retinal pericyte migration through Angiopoietin/Tie-2 signaling pathway on diabetic rats

AIM： To investigate the mechanism of pericyte migration through Angiopoietin-2 （Ang-2）/Tie-2 signaling pathway. METHODS： We divided the rats into 5 groups. Each diabetic rat model groups injected with Tie-2 inhibitor, ERK1/2 inhibitor, Akt/PKB inhibitor, and DMSO intravitreal. Retinal digest preparation was done to examine the retinal vasculature including pericyte： endothelial ratio, and morphology of pericyte migration. Tie-2, ERKI/2 and Akt/PKB phosporylation were analyzed by confocal laser scanning microscopy. RESULTS： There was a correlation between pericyte migration with increasing Ang-2 （P〈0.05）. pericyte number reduced by 40% （1：2.4） after 5wk diabetes on diabetic rats. The pericyte： endothelial ratio on group with Tie-2 inhibitor were 1：1.8. The same result shows on group with Akt/PKB inhibition. ERK1/2 inhibitor group shows the best results of pericyte： endothelial ratio （1：1,7）, Inhibition on Tie-2 receptor decreased the phosphorylation activity of Tie-2, ERK1/2 and Akt/PKB pathway. ERK1/2 inhibition also decreasing the phosphorylation of Tie-2 and Akt/PKB. But on Akt/PKB inhibition, the phosphorylation of Tie-2 and ERK1/2 were relative the same. CONCLUSION： Ang-2 has a role for pericyte migration on diabetic rats through Tie-2 receptor, ERKII2 and Akt/PKB pathways. ERK1/2 is a dominant pathway based on the ability to supress another pathway activity and decreasing pericyte migration on diabetic rats.

Retinal　capilary　pericyte　apoptosis　in　early　human　diabetic　retinopathy

Objective To investigate the nature of retinal capillary pericyte dropout in early human diabetic retinopathy. Methods In the present study, neural retinas of 12 postmortem eyes from 6 patients (3 diabetic and 3 nondiabetic) were first fixed in buffered formaldehyde and then digested with trypsin. The preparations of trypsin digested retinal blood vessels were stained with terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) technique and then with perdiodic acid Schiff technique (PAS), and counter stained with hematoxylin to evaluate DNA fragmentation of cell nuclei and diabetic changes of retinal capillaries. Results TUNEL positive apoptotic pericytes were observed in retinal capillaries in all three diabetic patients in this study. The TUNEL positive pericytes were presented as darker stained cells with PAS hematoxylin staining. A few TUNEL positive retinal capillary endothelial cells were discovered in both eyes of one of the three diabetic patients. Neither TUNEL positive pericytes nor positive endothelial cells were found in the three nondiabetic patients. A variety of typical diabetic microvascular changes were noticeable with PAS hematoxylin staining in trypsin digested retinal preparations from diabetic patients. These changes included thickened capillary walls, microaneurysms, “ghost cell” appearance of pericytes, acellular segments of capillaries, and a decreased ratio of retinal capillary pericytes to endothelial cells. All of these changes are characteristics of diabetic retinal microangiopathy. Conclusions Retinal capillary pericyte apoptosis is a speci fic form of cell death which occurs during the early development of diabetic retinopathy. The significant decrease in the ratio of pericytes to endothelial cells and the predominant TUNEL positive pericytes over endothelial cells suggest that a greater impact of apoptosis related mechanisms occurs in pericytes than in endothelial cells and,

Objective: Suppression of transcytosis is required for blood brain barrier(BBB) maintenance, and pericyte is potential regulating the BBB by inhibiting transcytosis. Therefore, Clarify and intervention to transcytosis induced BBB leakage after subarachnoid hemorrhage(SAH) are of prime interest. Our main aim was to study the potential role of pericyte foxf2 in transcytosis and consequent BBB disruption after SAH. Approach and Results: In an endovascular perforation SAH model, we characterize the SAH-induced BBB leakage in wide type(WT) and pericyte knockout(p) mice and subsequent brain edema on a cellular, molecular, and functional level. pericyte paucity aggravated the reduction of capillary foxf2 and docosahexaenoic acid(DHA) levels, endothelial transcytosis and BBB leakage after SAH;Using both retroviral and adenovirus associated viral approaches, we show that genetic inhibition of foxf2 significantly reduced the production of DHA from cultured pericyte;In contrast, overexpression of foxf2 enriched capillary DHA concentration, reduce cav1 and transcytosis, and eventually alleviated BBB disruption, brain edema and neurological defect at 2d after SAH. Conclusions: SAH causes pericyte reduce the production of DHA thereby enhances transcytosis and BBB disruption. Foxf2 plays an important role in those pathologies and could be a therapeutic target for SAH-induced BBB leakage.

Vascular pericytes,important mural cells that protect vascular integrity in healthy tissues,are often associated with tumor development in pathological conditions,but their functions in cancer inva sion remain elusive Recent studies have shown that pericytes retain progenitor cell properties,and upon proper stimulations differentiate into some other cells,such as adipocytes,osteoblasts,and skeletal muscle ***,by culturing pericytes on different polyac rylamide(PA) gels that mimic stiffness from healthy to pathological tissues,we found pericytes are matrix stiffness-responsive and different mechanical stimuli from PA gels induce the occurrence of pericyte-fibroblast transition(PFT).Soft PA gels that mimic the stiffness of healthy lung tissues retain the identity and behavior of pericytes,whereas stiff PA eels that reflect the stiffness of tumorous lung tissues help to accomplish PFT and increase their mobility and *** stiffness-induced PFT depends on the activation of YAP(Yes-associated protein),an transcription factor which upon receiving mechanical signals from integrinα1 and integrin a5 transfers from cytosol to nucleus to mediate transcription *** results reve al a mechanism through which vascular pericytes convert to fibroblasts and migrate away from vasculatures to help tumor development,and thus targeting matrix stiffness-induced PFT may offer a new perspective to the treatment of cancer.

Nitration of TRPM2 as A Molecular Switch Induces Autophagy During Brain pericyte Injury

Aims:Dysfunction of neurovascular pericytes underlies breakdown of the bloodbrain barrier,but the molecular mechanisms are largely unknown. In this study,we evaluated the role of the transient receptor potential melastatin-related 2(TRPM2) channel and autophagy during brain pericyte injury both in vitro and in vivo. Results:A rapid induction in autophagy in human brain vascular pericytes,in the zinc oxide nanoparticles(ZnONP)-induced cell stress model,was paralleled with an increase in the expression of the TRPM2-S truncated isoform,which was abolished by treatment with a nitric oxide synthase inhibitor and a peroxynitrite scavenger. Furthermore,Y1485 in the C-terminus of the TRPM2 protein was identified as the tyrosine nitration substrate by mass spectrometry. Overexpression of the Y1485 S TRPM2 mutant reduced LC3-Ⅱ accumulation and pericyte injury induced by ZnO-NP. Consistently,LC3-Ⅱ accumulation was reduced and pericytes were better preserved in intact brain microvessels of the TRPM2 knockout mice after ZnO-NP-induced vascular injury. Innovation and Conclusions:Our present study has revealed a novel mechanism of autophagy disturbance secondary to nitrosative stress-induced tyrosine nitration of TRPM2 during pericyte injury.

Vascular regeneration is a challenging topic in tissue repair. As one of the important components of the neurovascular unit(NVU),pericytes play an essential role in the maintenance of the vascular network of the spinal cord. To date, subtypes of pericytes have been identified by various markers, namely the PDGFR-β, Desmin, CD146, and NG2, each of which is involved with spinal cord injury(SCI) repair. In addition, pericytes may act as a stem cell source that is important for bone development and regeneration, whilst specific subtypes of pericyte could facilitate bone fracture and defect repair. One of the major challenges of pericyte biology is to determine the specific markers that would clearly distinguish the different subtypes of pericytes, and to develop efficient approaches to isolate and propagate pericytes. In this review, we discuss the biology and roles of pericytes, their markers for identification, and cell differentiation capacity with a focus on the potential application in the treatment of SCI and bone diseases in orthopedics.

TNFSF15 facilitates the differentiation of CD11b^(+) myeloid cells into vascular pericytes in tumors
