Effect of exogenous GA3 on flowering quality, endogenous hormones, and hormone-and flowering-associated gene expression in forcing-cultured tree peony（Paeonia suffruticosa）

Gibberellins(GAs)promote flowering in the forcing-cultured tree peony(Paeonia suffruticosa),however,the mechanism of regulating flowering is not fully *** this study,exogenous GA3 was applied to five-year-old Luoyang Hong plants to explore responses in terms of endogenous hormones,flowering quality,and the hormone-and flowering-associated gene *** GA3 application significantly promoted flower bud development and new branch growth,as well as improved flowering *** GA3 application also stimulated the synthesis of endogenous GA3 and indole-3-acetic acid(IAA)but reduced abscisic acid(ABA)*** further elucidate the regulatory mechanism,eight genes for GA biosynthesis and signaling,including PsCPS,PsKS,PsGA3ox,PsGA2ox,PsGID1b,PsGID1c,PsDELLA,and PsGID2 were cloned for the first time,and sequence analysis was also *** results suggested that all the cloned genes have conserved structure as each homologous gene reported in the other *** trees constructed by the each cloned gene showed that the phylogenetic evolutionary relationship of *** was closely related to Vitis *** expression patterns of the above genes,and genes for ABA and IAA biosynthetic and signaling,and the flowering time were also *** of the above genes showed higher expression in the control buds than those in the GA3 treated buds at six developmental stages,whereas the expression levels of PsSOC1 and PsSPL9 were up-regulated by GA3 *** results also showed that the GA-biosynthetic and signaling pathways are conserved in tree peony,and the PsCPS,PsGA3ox,PsGA2ox,PsGID1,PsDELLA,and PsGID2 genes are necessary for feedback regulation of ***,hormone changes promoted PsSOC1 and PsSPL9 expression,and repressed PsSVP expression,which contributed to the improvement flowering quality in tree peony of forcing culture.

Effect of Combined Chilling and GA_3 Treatment on Bud Abortion in Forced ‘Luoyanghong' Tree peony(Paeonia suffruticosa Andr.)

This research investigated the effect of combined chilling and GA_3 treatment on bud abortion of tree peony under greenhouse conditions to determine the dynamics of exogenous hormones and chilling effects on bud abortion. Research of such nature will increase the understanding of the effects of GA_3 and chilling level on flower development thereby, promoting off-season cultivation techniques in tree peonies to increase production that meets market demand. Hormone and sugar levels in the ‘Luoyanghong' tree peony were respectively determined using High-Performance Liquid Chromatography(HPLC) and spectrophotometer. Non-potted chilling with 200 mg·L^(-1) GA_3 treatment reduced bud abortion rate. Flowering rate in non-potted chilling with 200 mg·L^(-1) GA_3 plants was 58%, as against 21% in potted chilling plants with 200 mg·L^(-1) GA_3 treatment. Sugar accumulation in retained or aborted flower buds was conversely related to ABA level. Low ABA and high sugar levels were noted in retained buds whereas the reverse induced bud abortion. ABA level apparently inhibited sucrose and reducing sugar uptake in aborted buds whereas GA_3 enhanced sucrose uptake in retained buds. GA_3 and IAA levels were higher in retained than in aborted buds, which made plants with retained bud more competitive in nutrient uptake(by root)for growth and development. Overall, combined chilling and GA_3 treatment enhanced plant growth and development, particularly bud sprouting, growth and flowering.

Herbaceous peony in warm climate:Modelling stem elongation and growers profit responses to dormancy conditions

We analysed the data collected for herbaceous peony cultivated in a warm climate region and stored in winter under three constant chilling *** used the quadratic regression model to describe the stem elongation responses to winter dormancy conditions,and the logistic function to describe the weekly stems *** predicted maximal stem length from the first model was used as the input parameter for the second *** than 4000 data for various(a)chilling constant temperatures during dormancy,(b)dormancy duration,and(c)germination duration,were *** models were applied to determine the optimal number of chill *** this purpose,two criteria were used in different versions of the model:the maximal stem length and the maximal profit of *** the two chilling temperatures of 2℃ and 6℃,the optimal values of chill units(in the models of a maximal stem length and maximal profit of farmers)are close to one another,and the values of a maximal stem length and maximal profit are significantly *** the case of the third chilling temperature of 10℃,the model failed to determine the optimal number of chill *** method of inverse confidence intervals for testing the significance of the optimal number of chill units was used.

Genetic Diversity Analysis of Morphological Traits among 22 peony Cultivars

[ Objective] This study analyzed genetic diversity of 22 representative peony cultivars to investigate genetic differences between peony cultivars and ex- plore excellent germplasm resources, aiming at providing scientific basis for analysis of genetic relationships between different peony cultivars and germplasm innova- tion. [ Method ] Flower color, flower type, plant height, plant width, flower diameter, leaf length, leaf width, plant type and leaf type of 22 peony cultivars were surveyed. The genetic distances between peony cuhivars were calculated with Euclidean distance method. The genetic diversity of 22 peony cultivars was analyzed by UPGMA cluster analysis. [ Result] Based on the overall average of total genetic distance, UPGMA cluster analysis results were classified. Thus, 22 peony culti- vars were divided into five categories. Category [ consisted of Yinhongqiaodui, Jizhuahong, Babaoxiang, Chongloudiancui, Congzhongxiao, Weizi, Yanlongzi and Qiejiazi ; category II consisted of Huhong; category III consisted of Xiaohuhong, Baiyulan, Yingsuhong, Fengdan, Dajinfeng, Jingyu, Panzhongquguo, Qingshan- guanxue, Mojiatrrong, Dahongyipin and Lanhuakui; category IV consisted of Haihuang; category V consisted of Sihelian. The classification results were consistent with pedigree analysis. peony cuhivars with unknown pedigree were assigned to corresponding heterosis group. [ Conclusion] The diversity analysis of morphologi- cal traits is an effective method for investigating genetic differences between different cultivars, which may lay the theoretic foundation for breeding, industrial devel- opment and application of new peony cultivars.

A comparison of pharmacokinetic interaction of paeoniflorin, albiflorin and total peony glucosides

Aim Paeoniflorin （PF） and albiflorin （AF） are the main components of total peony glucosides （TPG）, which has been widely used as the valuable monoterpene glycosides in Paeouia lactiflora Pall and have many biological activities such as anti-inflammatory, anti-hypertension and antioxidation effects. In this study, the pharmacokinetic interaction and possible interaction mechanism among Pie and A1e and TGP were conducted and in- vestigated. A sensitive and specific ultra high-performance liquid chromatography - tandem mass spectrometry method （UPLC-MS） was successfully developed and validated for simultaneous quantification of Pie and AF in rat plasma after intragastric administration and the comparative study of pharmacokinetics for PF and A1e in different combinations. The combinations of Pie （ 80 mg· kg^- 1 ）, Ale （ 14 mg ·kg^- 1 ）, co-administration of Pie （ 80 mg · kg^-1） and Ale （14 mg· kg^-1）, TPG （Pie 80 rag. kg^-1） and TPG （Alel4 mg.·1kg^-1） were orally administrated to rats respectively. Chromatographic separation was performed on a ZORBAX Eclipse plus Cls column using 0.2% formic acid-methanol （79：21, V/V） as mobile phase. The calibration curves were linear in ranges of 0.5 - 1000 μg · L^-1. The results indicated that the main pharmacokinetic parameters including AUC, MRT and tl/2 between the single ingredient （ Pie and Ale） and TPG had significant difference （ P 〈 0.05 or P 〈 0.01 ）. The different phar- macokinetics data implied that the multiple active components of TPG resulting some potential drug-drug interac- tions, which may promote the absorption and decease the elimination of Pie and Ale. Our results could be helpful for further investigation of the interaction mechanism of Pie and Ale.

Study on Nutritional Characteristics of Dianjiang peony at Different Growth Stages

[Objective] This study aimed to investigate the nutritional characteristics of Dianjiang peony at different growth stages.[Method] The yield,nutrient content and nutrient uptake of Dianjiang peony at different growth stages and available nutrient content in soil were investigated by field survey and indoor chemical analysis.[Resuit] The results showed that biomass,leaf weight,tender stem weight and cortex moutan weight （yield） of 1-4 years old peony exhibited a significant quadratic regression relationship with the cultivation *** average content and uptake of 12 nutrients in leaves exhibited an overall order of Ca＞N＞K＞Mg＞S＞P＞Fe＞Mn＞ Zn＞Cu,which was consistent to that in tender ***,the average content and uptake of various nutrient elements in cortex moutan exhibited an overall order of N＞Ca＞K＞Mg＞P＞S＞Fe＞Zn＞Cu＞Mn,which was different from that in leaves and tender *** addition,the average content and uptake of various nutrient elements in petioles exhibited an overall order of Ca＞K＞N＞Mg＞S＞P＞Fe＞Mn＞Zn＞*** content of organic matter in soil of Dianjiang peony at different growth stages varied greatly; to be specific,contents of N,S,Mn and Zn were higher.[Conclusion] This study provided theoretical basis for the production of high-quality and high-yield peony.

Tissue Culture of Dianjiang peony Variety Taipinghong

[Objective] This study was to raise the reproductive efficiency of a Dianjiang peony variety Taipinghong. [Method] The Taipinghong was studied on the best selection and collection time of explant, the induction in the early generation and the subculture multiplication. [Result] The best explant of Taipinghong was bulbil, and the best collecting time was March. The best induction recipe in early generation was modified MS medium （ammonium nitrate rate was reduced by half; calcium ion concentration doubled）＋arrageenan 6 g/L＋succharcose 30 g/L＋NAA 0.2 mg/L＋6-BA 2.0 mg/L＋GA3 0.5 mg/L＋AC 100 mg/L. [Conclusion] In the solution to browning phenomenon, both dark and AC （activated charcoal） treatments could obviously decrease the browning rate of explant.

Effects of Water Stress on Photosynthesis and Fluorescence Characteristics in peony

[ Objective] Study on the photosynthesis and fluorescence characteristics in peony under water stress. [ Method ] The two peony varieties Huhong and Luoyanghong were treated by different water stress for determining the photosynthesis and fluorescence characteristics. [ Result ] With the aggravation of water stress, the net photosynthetic rate （Pn） and stomatal conductance （Gs） decreased, while the intercellular CO2 concentration （Ci） increased. Drought could decrease Pn, constrain gas exchange and change the daily photosynthesis. Fo of peony leaf increased and Fv/Fm decreased under water stress especially water logging, causing the inactivation of the PS II reaction center, and the chlorophyll fluorescence characters gradually recovered until afternoon. [ Conclusion] The 75% soil relative water content （SRWC） is the best condition for growth of peony. Compared with drought, water logging is more unfit for the growth of peony. For the two varieties, Huhong assumed more tolerance to drought, accordingly more adaptability.
