Background:pedunculoside(PE)is a pentacyclic triterpenoid saponin found in the dried barks of the Ilex rotunda *** traditional Chinese medicine,the juice extracted from various Ilex species is commonly used for treating inflammatory ***:This study aims to investigate the potential of PE in promoting the healing of oral ulcers in mice and to explore its possible *** and Methods:A mouse model of oral ulcer was established to evaluate the healing promotion effects of PE,and an in-vitro inflammatory cell model was established by using primary fibroblasts isolated from mouse oral mucosal *** transcription and protein expression related to cell proliferation,migration,and collagen synthesis were ***:PE showed the highest cure rate of 85.7%for oral ulcers in mice within 7-days *** significantly promoted cell proliferation and collagen secretion in fibroblast in-vitro *** also upregulated the gene transcription of Smad3,Col1a1 and ***,PE antagonized the inhibitory effect of WP1066 on STAT3activation,but did not affect cell ***:This study demonstrates that PE has potential therapeutic effects in promoting oral ulcer healing in *** improvement was achieved by the increasing collagen synthesis through the upregulation of STAT3 and Smad3 in mucosa *** results provide a preliminary pharmacological work for developing PE as a potential treatment agent for oral ulcers.

Extraction and Crystal Structure of pedunculoside

The title compound of pedunculoside （C36H58O10）, a main active triterpenoid saponin of Ilicis Rotundae Cortex, was isolated from the dried bark of Ilex rotunda Thunb., and characterized by X-ray diffraction analysis. It crystallizes in the orthorhombic system, space group P212121 with C36H58O10, a = 13.046（3）, b = 21.038（4）, c = 28.413（6） A, V = 7798.28（3） A3, Z = 8, Dc= 1.109 g/cm3, Mr = 650.82, F（000） = 2832, andμ = 0.080 mm-1. The final R = 0.0765 and wR = 0.1736 for 15117 observed reflections （I 〉 2σ（I））. The molecular crystal structure of pedunculoside shows relative stereochemistry of 3β, 19α,23-trihydroxy-urs- 12-en-28-oic acid-28-O-β-D-glucopy- rainosyl ester. The molecule is composed of one glucopyranosyl ring and five six-membered rings with ring junctures of A/B trans, B/C trans, and D/E cis. There are two C36H50O10 molecules in an asymmetric unit, and the title compound is stacked into a 2D layer structure through hydrogen bonds.

Chemical Constituents of Millettia speciosa

Objective To study the chemical constituents from Millettia speciosa. Methods The compounds were isolated and purified by silica gel, Sephadex LH-20, ODS column chromatography, and recrystallization. The structures were identified using physicochemical and spectral data. Results Thirteen compounds were isolated from M. speciosa and identified as docosanoic acid （1）, tetracosane （2）, octadecane （3）, hexacosanoic acid （4）, β-sitosteroi acetate （5）, β-sitosterol （6）, syringin （7）, maackiain （8）, formononetin （9）, ψ-baptigenin （10）, rotundic acid （11）, pedunculoside （12）, and daucosterol （13）. Conclusion Compounds 5, 7, and 10-12 are obtained from this plant for the first time.
