Fenofibrate is a member of such fibrate class agents as bezafibrate and it work as a ligand of pparα, and also shows a potent triglyceride-lowering effect. The elevation of aminotransferase levels has been frequently observed after the administration of fenofibrate and this phenomenon is considered to be non-pathological because fenofibrate activates the gene expression of the aminotransferases. Recently, fenofibrate has been used not only for hypercholesterolemia but also for primary biliary cirrhosis （PBC）. However, the occurrence of liver injury induced by fenofibrate has not yet been reported written in the English literature. We herein report a rare case of liver injury due to the oral use of this drug.

Nuclear receptor corepressor(NCoR1)interacts with various nuclear receptors and regulates the anabolism and catabolism of *** imbalance in lipid/energy homeostasis is also an important factor in obesity and metabolic syndrome *** this study,we found that the deletion of NCoR1 in intestinal epithelial cells(IECs)mainly activated the nuclear receptor ppara and attenuated metabolic syndrome by stimulating *** increase in brown adipose tissue thermogenesis was mediated by gut-derived tricarboxylic acid cycle intermediate succinate,whose production was significantly enhanced by ppara activation in the fed ***,NCoR1 deletion derepressed intestinal LXR,increased cholesterol excretion,and impaired duodenal lipid absorption by decreasing bile acid hydrophobicity,thereby reversing the possible negative effects of intestinal ppara ***,the simultaneous regulatory effect of intestinal NCoR1 on both lipid intake and energy expenditure strongly suggests that it is a promising target for developing metabolic syndrome treatment.

Silencing effect of recombinant plasmids pparγ-pSUPER-EGFP on the expression of peroxisome proliferator-activated receptor γ

To investigate the silencing effect of gene encoding peroxisome proliferator-activated receptor γ （ppar-γ） on the expression of tumor necrosis factor alpha （TNFα） by constructing vectors for RNA interference in RAW264.7 cells. Methods： The pSUPER-EGFP vectors were used to transcribe functional small interfering RNA （ siRNA ）. Four pairs of oligonucleotides （ 64 nt ） targeting ppar-γ gene were inserted into the downstream of the HI promotor, with their veracity confirmed by double digestion and sequencing. Western blotting and immunofluorescence assay were used to examine the silencing effect of ppar-γ gene in RAW264.7 cells. Meanwhile, the TNFα level was determined by Sandwich ELISA. Results ： Compared with other recombinant pSUPER- EGFP vectors （R-pSUPER. EGFP）, R-pSUPER. EGFP2 induced the best silencing effect on the expression of pparT in RAW264.7 cells, which played an obvious inhibitory role in down-regulating the TNFα expression after the curcumin and lipopolysaccharide （LPS） stimulation. Conclusions： pparγ-pSUPER-EGFP inducing a silencing effect on the expression of pparT can efficiently play a negative role in controlling the inflammatory responses of RAW264.7 cells.
