Characterization of a Novel Annexin Gene from Cotton (Gossypium hirsutum cv CRI 35) and Antioxidative Role of its Recombinant Protein

plant annexins represent a multigene family involved in cellular elongation and development. A cDNA encoding a novel annexin was isolated from a cotton （Gossypium hirsutum） fiber cDNA library and designated GhAnxl. This gene encodes a 316 amino acid protein with a theoretical molecular mass of 36.06 kDa and a theoretical pl of 6.19. At the amino acid level, it shares high sequence similarity and has evolutionary relationships with annexins from higher plants. The purified recombinant protein expressed in Escherichia coil was used to investigate its physicochemical properties. Circular dichroism spectrum analyses showed a positive peak rising to the maximum at 196 nm and a broad negative band rounding 215 nm, suggesting that the GhAnxl protein was prominently α-helical. The fluorescence measurements indicated that it could bind to Ca2 in vitro. These results demonstrated that GhAnxl was a typical annexin protein in cotton. A bioassay experiment was conducted to analyze its potential function and showed that E. coil cells expressing GhAnxl were protected from tert-butyl hydroperoxide （tBH） stress, suggesting that it had a potential antioxidative role. Northern blot analyses revealed that GhAnxl was highly expressed in fibers, especially during the elongation stage, suggesting that it might be important for fiber elongation.

Use of flue gas desulfurization gypsum to reduce dissolved phosphorus in runoff and leachate from two agricultural soils

Controlling dissolved phosphorus(DP)loss from high P soil to avoid water eutrophication is a worldwide high priority.A greenhouse study was conducted in which flue gas desulfurization gypsum(FGDG)was applied by using different application methods and rates to two agricultural *** fertilizer was incorporated into the soils at 2.95 g kg^(-1)to simulate soil with high P *** FGDG was then applied at amounts of 0,1.5,and 15 g kg^(-1)soil on either the soil surface or mixed throughout the soil samples to simulate no-tillage and tillage,*** was planted after treatment *** study showed that FGDG reduced runoff DP loss by 33%and leachate DP loss 38%in silt loam soil,and runoff DP loss 46%and leachate DP loss 14%in clay loam soil,at the treatment of 15 g kg^(-1)*** applied method(tillage)provided strong interaction with higher *** overall effect,the mixing-applied method performed better in controlling DP loss from silt loam soil,while surface-applied(no tillage)showed its advantage in controlling DP loss from clay loam *** practice it is necessary to optimize FGDG concentrations,application methods,and DP sources(runoff or leachate)to get maximized benefits of FGDG *** FGDG application had no negative effects on the soil and ryegrass.
