Genetic analysis on 3'- terminal flanking region of uncoupling protein 3 in different pig breeds

The 3’-terminal flanking region of porcine uncoupling protein 3 (UCP3) was cloned, the sequence data revealed 15 nucleotide substitutions among Landrace and three Chinese native pig breeds named Neijiang, Minpig and Erhualian. The continuous 9 polymorphic sites were checked by pcr-rflp. the results indicated that Erhualian had extraordinary gene frequency, presented most significant difference by x2 test compared with Landrace, Largewhite, Neijiang and Minpig respectively, significant level compared with Meishan; and Meishan also had significant difference compared with Landrace and Minpig respectively. These results can be concluded that Taihu pigs have special genetic characteristics among pig breeds.

Genetic Polymorphism of Kappa Casein （K-CN）in Iraqi Buffalo Using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism

This study was carried out the animal production department, genetic engineering lab, college of agriculture, （UoB）, Iraq. The aim of this study was to use the polymerase chain reaction restriction fragment length polymorphism （pcr-rflp. as a fast, efficient and low cost method to detect the genetic variants of kappa-casein gene （k-CN） in Iraqi buffalo using three different primers specific for bovine k-CN to amplify the gene segment, followed by digestion using restriction enzyme （Hind III） for genotyping. DNA from 50 Iraqi buffaloes was extracted by phenol chloroform method. pcr was carried out in a final reaction volume of 25 μL and the reaction mixture was subjected to standard pcr protocol. The results of this work show that among the examined 50 Iraqi Buffalo were homozygous for the K-CN and genotyped as BB for all three primers but gave different bands. Thus pcr-rflp.using Hind III revealed all the samples to be monomorphic for this locus. The restriction digestion analysis of 397 bp pcr product of k-CN indicates the presence of two fragments of 154 bp and 225 bp for BB-genotype. A 437 bp fragment of the bovine genomic K-CN gene was amplified. One Hind III restriction site is found in position 346 of the amplified fragment of allele k-CN B, yielded 91 bp and 346 bp. Amplified products from Iraqi buffalo （530）, after being digested with Hind III, yielded two separate DNA fragments of different sizes i.e., 160 bp and 370 bp. For the first time completed research such specifications in Iraq, for the first time using molecular biology in genetic identification. Our objectives of this study have been to aid in understanding domestication, Buffalo origin and their history and evolution, to identify genetically unique breeds, to provide an objective basis for conservation decisions and to aid the formulation of breeding plans.

Provincial distribution of three HIV-1 resistant polymorphisms (CCR5-Δ32, CCR2-64I, and SDF1-3' A) in China

CCR5-Δ2, CCR2-641, mutants in two chemokine receptors and SDF1-3’ A, mutant in a ligand gene, can delay AIDS pathogenesis. The distribution of the three polymorphic loci was studied in 1 046 DNA samples from 26 provinces (cities) in China. No CCR5-Δ32 was observed. CCR2-64I and SDF1-3’ A had reverse distribution cline from south to north in China, with average frequency of 20.8% and 24.8% respectively. Relative hazard was evaluated. Important information to the epidemiology of AIDS and the origin and spread of these polymorphic loci in Chinese was provided.

＜正＞pcr direct identification and pcr-rflp.analysis from single nematodes were employed to discriminate Bursaphelenchus xylophilus from B. mucronatus. For pcr direct identification, two specific primer sets from ITS amplified a 220bp and 330bp fragment from DNA of B. xylophilus, respectively. But no amplification band obtained from DNA of B. mucronatus. Restriction patterns of pcr-rflp.have revealed that Hinfl and MspI could be used to discriminate B. xylophilus from B. mucronatus. Both methods employed here could be used to identify the single nematode of B. xylophilus sensitively and accurately, and pcr direct analysis is more convenient. pcr-rflp.analysis could be used as an alternate method to confirm the result of pcr direct analysis when necessary.
