Grain number and seed-setting rate are components of crop *** influences grain ***,emerging studies suggest that high cytokinin signals often lead to reduced branching or seed-setting rate,leading to reduced grain yield,although the mechanisms remain *** this study,we identified and characterized the rice(Oryza sativa L.)gene LONELY GUY-LIKE 8(LOGL8),based on analysis of the LOGL8-pm(promoter mutant of LOGL8)mutant,which harbors a T-DNA insertion in the promoter of this *** mutation in LOGL8-pm causes ectopic hyperexpression of LOGL8 in inflorescence organs,resulting in plants with smaller panicles and defective ovules lacking archesporial cells and *** of LOGL8 caused pollen abortion,leading to a reduced seed-setting ***8 encodes a putative cytokinin-activating *** results showed that LOGL8 directly catalyzes the biosynthesis of bioactive ***,we propose that the ectopic expression of LOGL8 disrupts cytokinin spatiotemporal distribution and causes inhibition of LONELY GUY(LOG),which affects panicle branching and female organ *** findings reveal the important role of LOGL8 in male development,and highlight the delicate balance of local cytokinin levels during panicle branching and female organ development.

In order to investigate the cause of poor fruit set in 'Zuili' plums,anatomical examinations of post-bloom pistils were conducted and the dates of young fruit drop were recorded during the growing seasons of 2008 and *** of cv.'Black Amber' were also examined as an abundant setting *** major dropping periods were detected in 'Zuili':one during the first 5 d after full bloom (DAF) and another between 10 and 17 *** analyses of the pistils at the full bloom stage revealed that half of the ovules had not developed embryos,which may have caused their early *** most dropped pistils collected at 17 DAF,the micropyle had not been penetrated by a pollen tube,indicating that they were not fertilized.'Zuili' ovules initiated embryo division at 10-12 DAF,although thereafter embryo development was retarded when compared to the rates observed in 'Black Amber'.ovule fertilization failure and inactive embryo development after ovule fertilization may be the major causes of the later fruit drop observed in 'Zuili' plum trees.

Nicotiana tabaccum ovule extracts induced nuclear reconstitution of demembranated Xenopus leavis sperm in a cell-free system. Demembranated Xenopus sperm began to swell after 15 min of incubation with Nicotiana ovule extracts. Accompanying the process of incubation, Xenopus sperm decondensed and their shapes changed gradually from long and ellipse to round. The completely decondensed chromatin was surrounded with membrane structure, which was a mixture envelope of a double membrane and a single membrane. Nucleosome assembly was verified by means of micrococcal nuclease digestion to re-constituted nuclei and DNA electrophoresis. Nicotiana ovule extracts supplied one more experimental model and system. The new system could promote powerfully the research on mechanisms of cell division and cell cycle regulation.

Nfbp6 was a floral homeotic gene isolated from tobacco. It showed high homology with other class C genes. Using in situ hybridization technique, the expression of Nfbp6 was investigated in various stages of flower development. The results showed that Nfbp6 was expressed at low level during the initiation period of stamen and carpel primordia. In the later stage of flower bud development, the transcription level of Nfbp6 gene increased especially during the formation of pollen and ovule. The Nfbp6 gene expression was also observed in the transmitting tissue of style, stomium and circular cell clusters of the anther.

Distribution of nuclei of different ploidy levels was studied at different developmental stages in the embryonic tissue of the ovule, seed and protocorm of Phalaenopsis aphrodite subsp. formosana (Miwa) E.A. Christ. by a combination of flow cytometry and fluorescence microscopy with Apo Tome Slider. Three stages of ploidy patterns were identified in the ovular tissue at different days after pollination (DAP). Firstly, between pollination and fertilization (0 to 50 DAP), 2C nuclei were dominant over 4C nuclei and resulted in low level of cycle value. Secondly, between fertilization and seed maturation (50 to 110 DAP), amount of 4C nuclei increased rapidly, maintained at a high level and then decreased gradually to a low level. Small amount of 8C nuclei was also detected at this stage. Thirdly, at seed maturation (110 to 130 DAP), 2C nuclei became dominant over 4C nuclei again and the cycle value remained at a low but significant level at this stage. After seed sowing, nuclei with ploidy levels of 2C, 4C and 8C were observed in the developing protocorms as early as at 4 DAS (days after sowing). Nuclei with high ploidy levels (8C and 16C) increased gradually until 40 DAS in this study. Significant level of cycle value at this stage of protocorm development indicated the presence of endopolyploidy. 4,6-diamido-2-phenylindol (DAPI) staining showed large and prominent nuclei in the basal portions of the mature seeds before sowing and in the developing protocorms at 20 DAS. These findings clearly demonstrate the occurrence of different distribution patterns of nuclei with different ploidy levels during ovule, seed and protocorm de-velopment in Phalaenopsis aphrodite. These observations will provide fundamental information for further studies in Phalaenopsis orchids.

Phytolacca is the biggest and most original genus in Phytolaccaceae and an important genus in plant systematic *** microscopy results show that the Phytolacca americana *** arises from the caulis(floral receptacle).The perisperm and hypostase are simultaneously initiated from the top several layers of cells of chalaza after fertilization,and the perisperm is located between the nucellus and *** the early stages of development,the hypostase cells are thin-walled with dense cytoplasm,clear nuclei,and some reserve ***,at the heart-shaped embryo stage,the hypostase cells are dead and *** main functions of the hypostase may be to maintain cellular division and perisperm growth without delivering nutrient materials to the *** evolutionary picture of placentation in Caryophyllales is also presented.
