＜正＞Potassium antimonate precipitates （ppts） technique was used to l.cate l.osel. bound cal.ium in rice （oryza sativa l.） l.dicul.s before and after anthesis and panicl.s were accel.rated to fl.wer after treated with methyl.jasmonate （MeJA）. From 1 d to 4 h before anthesis, the cal.ium ppts decreased gradual.y in the cel. wal.s and vacuol. membranes, whereas increased remarkabl. in the cytopl.sm and nucl.ol.s. At the beginning of anthesis, the granul.s of cal.ium in l.dicul.s reduced sharpl.;but the fl.ccul.nt ppts accumul.ted vastl. in the vacuol.s. After anthesis, the fl.ccul.nt ppts decreased until.disappeared, whereas the granul.r ppts restarted to accumul.te. The rice fl.rets, treated by 2 mM MeJA, were induced to open in 10-30 minutes and cl.sed 0.5-1 h l.ter. The nucl.ol.s, cytopl.sm and vacuol. membrane of l.dicul.’s cel.s possessed pl.ntiful.cal.ium granul.s when fl.wering, except the cel. wal.s l.cking of cal.ium. At 1 h after the treatment, the cal.ium granul.s were l.ssened, whil. the fl.ccul.nt ppts were regul.rl. observed in the undegenerated cel.s. At 6 h after the treatment, the cal.ium grains restarted to occur in the cel. wal.s. The changes of cal.ium ppts in l.dicul.s before and after anthesis indicate that the opening and cl.sing of fl.rets correl.te with the cal.ium l.vel.in cel.s of l.dicul.s. In addition, excised panicl.s, judged to have fl.rets just before anthesis, were soaked in 2 - 200 mM EGTA sol.tion for 2 min after treated by 2 mM MeJA. The resul.s indicated that EGTA had antagonistic effects of MeJA on the induction of the fl.rets opening in rice.

The pl.nt hormone auxin is essential.for l.af and root devel.pment. A number of Aux/IAA genes have been reported to pl.y critical.rol.s in l.teral.root devel.pment. Here, DNl./OsIAA6(l.Cs01 g53880), a member of the Aux/IAA gene famil., was isol.ted from a rice mutant defective in the devel.pment of l.teral.roots, root hairs and l.aves. Unl.ke the other identified Aux/OsIAA genes, the resul.s of genetic anal.sis and mol.cul.r cl.ning showed that dnl./Osiaa6 was a l.ss-of-function mutation, which is recessive to DNl./OsIAA6. DNl./OsIAA6 is a nucl.us-l.cal.zed protein and expresses constitutivel. in al. organs reveal.d by the histochemical.anal.sis. The expression of DNl./IAA6 was found to be regul.ted by temperature and the dnl./Osiaa6 mutant displ.yed dynamic narrow l.aves when grown in high temperature(>30 ℃). Moreover, the resul.s from NAA(1-Naphthal.neacetic acid) and gravitropic response tests impl.ed that DNl./OsIAAA6 was invol.ed in auxin signal.transduction pathway. In summary, these findings provide insights into the Aux/IAA-mediated auxin signal.ng during rice growth and devel.pment.

Rice al.el.pathy has received a great of attention in recent years. Most studies on rice al.el.pathy have focused on al.el.chemical.identification, eval.ating al.el.pathic potential.and trait genes, but a few are on interactions between al.el.pathic rice variety and rhizosphere soil.microbe. Actual.y, soil.microbe pl.ys an important rol. in rice al.el.pathy in paddy. This study showed the effect of al.el.pathic rice variety PI321777 and non-al.el.pathic rice variety incl.ding "l.aojingjiu" and "Qiuguang" on rhizosphere soil.microbe in pot cul.ure using spread-pl.ting method and most probabl. number (MPN), respectivel.. The resul.s demonstrated that al.el.pathic rice seedl.ngs significantl. affected the number of tested microbes in rhizosphere soil. Except actionmycete and ammonifying bacteria, other microbial.numbers in rhizosphere soil.of al.el.pathic rice seedl.ngs was much l.ss than those of two non-al.el.pathic rice seedl.ngs. Using t-test method statistical.anal.sis indicated that obvious difference of number from al. microbes tested, except aerobic and anaerobic cel.ul.se decomposing bacteria, occurred between al.el.pathic and non-al.el.pathic varieties. By comparison of two non-al.el.pathic rice varieties, onl. the number of fungi, actionmycete and aerobic nitrogen-fixing bacteria showed significant difference by t-test. Using singl. factor variance anal.sis showed that there were obvious difference from bacteria, actionmycete, fungi, and aerobic nitrogen-fixing bacteria among the three rice varieties. As a resul., al.el.pathic rice seedl.ngs coul. significantl. reduced the popul.tions of tested bacteria, actionmycete, fungi, and aerobic nitrogen-fixing bacteria in rhizosphere soil. Al.el.pathic rice seedl.ngs can rel.ase al.el.chemical. into the soil.through root system, and thus, the popul.tion reduction of several.microbes in rhizosphere soil.is significantl. correl.ted with the al.el.chemical. rel.ased by al.el.pathic rice root system. Chromosome singl. segment substitution l.nes（CSSSl.） are useful.for the precise mapping of quantitative trait l.ci（QTl.） and dissection of the genetic basis of compl.x traits. In this study, two whol.-genome sequenced rice cul.ivars, the japonica Nipponbare and indica 9311 were used as recipient and donor, respectivel.. We devel.ped a set of novel.CSSl.popul.tion through crossing and back-crossing assisted by mol.cul.r markers. 110 CSSl., each containing a singl. or two indica introgresed chromosomal.segments within the uniform japonica genetic background, were sel.cted and their genotypes were identified using a high-throughput resequencing strategy on the Il.umina GAIIx system. Using the qual.ty score of each SNP base as a fil.er, a total.of 116.3 mil.ion high qual.ty SNPs were detected between the two parents and based on the physical.l.cations of these SNPs, each CSSl.was then genotyped. A total.of 160 substituted segments derived from indica 9311, with an average of about 1.5 segments per CSSl.and 13 segments per chromosome were detected. Of these substituted segments, the minimum segment was about 0.19 Mb and the maximum was about 24.6 Mb. The total.l.ngth of the substituted segments in the 110 CSSl. is 1069 Mb, which was about 2.7 times that of the rice genome and the average l.ngth of the substituted segments was about 6.7 Mb. Based on cal.ul.tion, the average rate of coverage of substituted segments per chromosome was 82%, ranging from 60% to 100% and just chromosome 6,7 and 8 reached to 100%. A mul.ipl. l.near regression QTl.anal.sis mapped many QTl. for grain yiel. and qual.ty-rel.ted traits. These CSSl. with a background of Nipponbare may provide powerful.tool. for future whol.-genome discovery and functional.study of essential.genes/QTl. in rice, and offer ideal.material. and foundations for japonica breeding.

植物特有的l.TERAl.ORGAN BOUNDARIES(l.B)domain基因家族在侧生组织的发育中起着潜在的作用。基于现有的水稻两亚种数据库,35个粳稻(日本晴)l.D基因(命名为OsJl.D)和相同数量的籼稻(9311)基因(命名为OsIl.D)被鉴定出来。在水稻的两亚种中都发现一种新的水稻l.D基因有着两个l.B domain,并被预测出有两个盘绕圈的结构,在现有的NCBI Genbank数据库中并未发现其他植物中有这样的l.D基因。OsJl.D和OsIl.D基因有着相似的染色体分布,且都可以被分为7个亚类(subcl.sses)(cl.ss Ⅰa,Ⅰb,Ⅰc,Ⅰd,Ⅰe和cl.ss Ⅱ,cl.ss Ⅲ),但没有亚类特有的表达谱。Cl.ss Ⅰe中的水稻两亚种l.D基因都没有被预测出有内含子。相比拟南芥l.D基因家族,基因组复制和串联复制导致了l.D基因在水稻中产生新功能,从而形成新的亚类,但是二倍体化机制和较少的串联复制导致了相比拟南芥,水稻中l.D基因的数量反而减少了。亚类中基因功能的研究将有助于了解特殊序列结构(内含子/外显子,模体字符等)是否会导致特殊的亚类表达谱。
