Composition of ophiopogon Polysaccharide,Notoginseng Total Saponins and Rhizoma Coptid is Alkaloids Inhibits Myocardial Apoptosis in Diabetic Atherosclerosis Rabbits

Objective: To investigate the effects of Composition of ophiopogon polysaccharide, Notoginseng total saponins and Rhizoma Coptidis alkaloids(CONR) on myocardial apoptosis of diabetic atherosclerosis(DA) rabbits Methods: Sixty male New Zealand white rabbits were randomly divided into 6 groups [control group, model group, CONR high-dose group(450 mg/kg), CONR medium-dose group(150 mg/kg), CONR low-dose group(50 mg/kg), and simvastatin group] by using a completely random method, 10 in each group. DA model was established by intravenously injected alloxan combined with high-fat diet and abdominal aortic balloon injury. After mediation for 10 weeks, fasting blood glucose(FBG), glycosylated hemoglobin(GHB), glycosylated serum protein(GSP), fructoseamine(FRA), aldose reductase(AR), advanced glycation end products(AGEs) in serum were measured by enzyme linked immunosorbent assay(ELISA) method;the expression of receptor of AGEs(RAGE) in myocardial tissue were observed by immunohistochemical method;and p-Jun N-terminal kinase(p-JNK), caspase-3, B-cell lymphoma-2(bcl-2) protein expression in myocardial tissue were measured by Western blotting. The myocardial apoptosis was detected by Td T-mediated d UTPnick-end labeling(TUNEL) method, and apoptosis index(AI) was calculated. Results: Compared with the control group, serum FBG, GHB, GSP, FRA, AR, AGEs and the expression of myocardium RAGE, p-JNK, caspase-3 proteins, as well as apoptosis index(AI) were significantly increased and bcl-2 protein was significantly decreased in the model group(P<0.01). Compared with the model group, the levels of serum FBG, GHB, GSP, FRA and AR showed a significant decline in CONR high-and medium-dose groups(P<0.01). FBG and GHB showed a significant decline in CONR low-dose group(P<0.01). Compared with the model group, the expression of serum AGEs and myocardium RAGE, p-JNK and caspase-3 protein as well as AI were significantly decreased and bcl-2 protein was significantly up-regulated in all treatme

Characterization and Antioxidant Properties of OJP2, a Polysaccharide Isolated from ophiopogon japonicus

A water-soluble polysaccharide (OJP2) obtained from the roots of ophiopogon japonicas, was precipitated with 95% ethanol and purified by DEAE-52 cellulose anion-exchange and Sephadex G-100 gel filtration chromatography. The characteristics of OJP2 were determined by chemical analysis, high performance gel permeation chromatography (HPGPC), and gas chromatography-mass spectrometry (GC-MS). The results showed that the average molecular weight (Mw) of OJP2 was 35.2 kDa, and five kinds of monosaccharides including rhamnose, arabinose, xylose, glucose and galactose in a molar ratio of 0.5:5:4:1:10. Furthermore, the antioxidant activity of OJP2 was evaluated in H2O2-treated HaCaT cells and glucose-treated LO2 cells. The results show that OJP2 can increase the activity of SOD and NO production, and decrease the level of MDA in these two kinds of injury cells. OJP2 should be explored as a novel and potential natural antioxidant agent for use in functional foods or medicine.

Antioxidant Activity in Vitroof Polysaccharide Extracted by Ultrasound with Different Powers from ophiopogon japonicas

The polysaccharides were extracted by different power ultrasound from ophiopogon japonicas. The extraction yield and in vitro antioxidant activities including scavenging effect on hydroxyl radical, superoxide anion free radicals and DPPH were investigated. The results showed that with the increase of ultrasonic power, the yield of the polysaccharides decreased first and then increased. The antioxidant activity of polysaccharides increased first and then decreased with the increase of ultrasonic power. When the power was 400 W, the scavenging effect on superoxide anion and DPPH of the polysaccharides was the strongest, and the scavenging ability of hydroxyl radical was the strongest when the power was 560 W. Therefore, different ultrasonic power could affect the extraction yield and antioxidant activity of ophiopogon japonicas polysaccharide. It will provide theoretical basis and experimental support for the application of ultrasonic extraction on polysaccharides from ophiopogon japonicas.

Effects of Biological Fertilizer and Paclobutrazol on Total Flavonoids in ophiopogon japonicus

[Objectives] The research aimed to study the effects of applying biofertilizer instead of paclobutrazol on the content of flavonoids in ophiopogon japonicas and determine its quality characteristics,in order to provide the scientific basis for the quality control of GAP and the comprehensive utilization of resources. [Methods] O. japonicas collected in Lingxing Town and Huayuan Town of Sichuan Province was divided into three groups: blank control group,paclobutrazol group and biofertilizer group. Root tuber of O. japonicas was obtained,and the enzyme was killed by the microwave,and then it was dried in 60℃. The content of flavonoids in root tuber of O. japonicus was determined by ultraviolet spectrophotometry. Then,SPSS19. 0 was used to analyze the results. [Results]The average contents of flavonoids in different treatments were as follows: 0. 184%-0. 198% in control group; 0. 283%-0. 330% in the paclobutrazol group; 0. 404%-0. 428% in the biofertilizer group. Biological bacterial fertilizer had a significant role in promoting the synthetic accumulation of homoisoflavonoids in O. japonicus( P <0. 001**). [Conclusions]The application of biological fertilizer can not only significantly improve the content of homoisoflavonoids in root ruber of O. japonicus,but also significantly optimize the micro ecological environment of soil. Therefore,biofertilizer should be widely used to gradually replace paclobutrazol in the main production areas of O. japonicas.

Preliminary Research of the Regular Pattern between Flavonoids Content and the Age of Erect Type ophiopogon jaonicus

[Objective] The experiment aimed to inquire into the regular pattern of the flavonoids content in erect type ophiopogon jaonicus preliminarily.[Methods]In the main ophiopogon jaonicus-producing area,we use the method of home position dynamic method to gather sample for experiments,use UV to determine the flavonoids content in ophiopogon jaonicus and use correlation and regression method to analyze the relation between flavonoids content and the growth period of ophiopogon jaonicus.[Results]The relation between flavonoids content(y1)and the growth period(x∥d)of erect type ophiopogon jaonicus is:y=1.235 6lnx-4.748 9(n=7,50≤x≤730,0≤y≤3.128,r0.001=0.950 7,r=0.954 2).[Conclusion]During a determinate time(50≤x≤730),positive correlation exists between flavonoids content and the growth period of ophiopogon *** research has a great importance in directing the scientific harvest and GAP production of erect type ophiopogon jaonicus.

Effects of Plant Growth Regulators on Tillering Ability of ophiopogon japonicus cv

[Objectives]This study was conducted to discuss the effects of tillering law and its controlling factors on the growth of ophiopogon japonicus cv.[Methods]With *** cv as an experimental material,its average number of tillers,proliferation coefficient,total number of tillers,death number,germination index,germination rate,fresh root weight,root volume and survival rate were determined.[Results] The reproductive ability of the NAA treatments was significantly higher than that of the 6-BA treatments,and the average tillering capacity of the 5 mg/L NAA treatment was significantly higher,90% higher than that of the *** proliferation coefficient of *** cv was significantly better in the NAA treatments than in the 6-BA treatments,and the 5 mg/L-NAA treatment was more *** NAA treatments showed total numbers of tillers significantly higher than those of the 6-BA treatments,and exhibited significantly reduced *** 1 mg/L NAA and 10 mg/L 6-BA experimental groups had the strongest germination-promoting effects on *** cv,which were significantly different from those of other experimental groups,and were 92% and 95% higher than that of the CK,*** to the germination rate indicator of *** cv,the 1 mg/L NAA and 10 mg/L 6-BA experimental groups exhibited the highest values,which increased by 48% and 43%,respectively,compared with the *** 6-BA treatments significantly improved the root growth of *** cv compared with the NAA treatments,and they significantly promoted root volume of *** *** survival rate of *** cv treated with hormone NAA was 23.3% higher than that of the 6-BA treatment group.[Conclusions] This study provide a theoretical basis for improving the reproductive ability of *** cv and expanding its commercial production.

Relevance of Carbon Sequestration to the Physiological and Morphological Traits of Several Green Roof Plants during the First Year after Construction

Different vegetation types used for the extensive green roofs have characteristic physiological and morphological traits (e.g., C3, C4, or CAM photosynthesis, deciduous or evergreen). Several Sedum species are recognized as “inducible CAM” type plants. These differences in the physiological and morphological traits have a considerable effect on the carbon sequestration in the green roofs. The objective of the present study was to quantify the carbon sequestration in several green roof plants during the first year after the construction of the green roofs and to clarify the relevance of the physiological and morphological traits to each plant’s ability to sequester carbon in its body using the growth analysis method. We used Zoysia matrella , ophiopogon japonicus , and Sedum mexicanum species for the study wherein, S. mexicanum was assigned to the wet, dry, and non-irrigation treatments, and Z. matrella and O. japonicus only received the wet treatment. During the first year after the construction, carbon sequestration in the plants and the substrate of S. mexicanum was in the range of 276 to 364 g-C/m2/year, which was similar to that of O. japonicus and the finding of a previous study. In contrast, Z. matrella exhibited the highest carbon sequestration (670 g-C/m2/year), which is also expressed as the relative plant C-sequestration rate per whole-plant C-content (RGRc), because Z. matrella is a C4 plant and exhibits the highest net assimilation rate (NARc) of all species. Significant differences were not observed in RGRc , NARc , and RMF (root mass fraction) in S. mexicanum between the wet and dry treatments. These results suggest that in countries with high rainfall, a high frequency of irrigation has an insignificant effect on the physiological and morphological characteristics, and carbon sequestration in the Sedum green roofs.
