A study of efficacy of nigella sativa in treatment of Meniere's disease: A randomized, placebo controlled clinical trial

Purpose:Meniere's disease(MD),first introduced by Prosper Meniere,is characterized mainly by vertigo,tinnitus,aural fullness and sensorineural hearing loss.Though the exact pathophysiology of MD is unknown,immunologic and inflammatory interactions are possible underlying mechanisms involved in MD.This study is aimed to investigate the immunomodulatory and anti-inflammatory effect of nigella sativa on MD as a therapeutic agent.Methods:We divided 40 patients with definite MD into two groups of 20 cases.The study group received 1 g of nigella sativa oil daily for three months and the control group received a placebo.Changes in hearing,tinnitus and vertigo were estimated by pure tone audiometry,tinnitus handicap inventory questionnaire and dizziness handicap inventory questionnaire,respectively.Results:At the end of the study we did not observe any significant improvement in study's group hearing threshold,tinnitus and vertigo compared to the control group.Conclusions:In this study,statistical analysis showed that nigella sativa failed to improve signs and symptoms of MD.However,further investigations with a larger study population are needed to ascertain the current conclusion.

Drought-Mediated Modulation in Metabolomic Profiling of nigella sativa Leaf, Growth, Ecophysiology and Antioxidants

Abiotic stresses,including drought,have been found to affect the growth and medicinal quality of numerous herbs.The proposed study aims to study the effects of different drought regimes on the metabolic profile,growth,ecophysiology,cellular antioxidants,and antioxidant potential of nigella sativa(Black cumin)leaf.Forty-day-old seedlings of N.sativa were exposed to three regimes of drought(control,moderate and high)for a week.UPLCMS/MS metabolic profile of the leaf reveals the presence of more than a hundred metabolites belonging to anthocyanins,chalcones,dihydro flavonoids,flavonoids,flavanols,flavones,flavonoid carbonoside,isoflavones,etc.Drought was found to alter the contents of identified metabolites.Drought stress-induced oxidative stress and increased production of hydrogen peroxide and superoxide anions.Physiological changes,activities of antioxidant enzymes,contents of antioxidants,and proline were significantly high under drought to protect against the low water regimes.Furthermore,stressed leaf extract had higher antioxidant potential.Thus,N.sativa leaf bears multiple metabolic pathways and can tolerate a higher degree of drought or osmotic stress.

Antidepressant and anti-nociceptive effects of nigella sativa and its main constituent,thymoquinone:A literature review

Medicinal plants and their ingredients have beneficial effects on human health.nigella sativa is a herbal plant with multiple biological and pharmacological activities.Previous studies demonstrated the anti-inflammatory and antioxidant properties of nigella sativa and its main constituent thymoquinone significantly contributes to the antidepressant and anti-nociception effects of this plant.It has been reported that thymoquinone may achieve its antidepressant effect by preventing the elimination of brain neurotransmitters affecting depression such as serotonin.The role of brain-derived neurotrophic factors in the antidepressant effects of thymoquinone has also been documented.Additionally,thymoquinone can attenuate pain by upregulation of intracellular signaling pathways related to nitric oxide and K_(ATP)^(+)channels.The present review summarizes the antidepressant and anti-nociceptive activity of nigella sativa and its main constituent thymoquinone by searching literature on electronic databases such as PubMed,Web of Science,Scopus,and Google Scholar from the beginning of 2010 until the end of August 2022.

Molecular Interaction Study to Explore the nigella sativa Bioactive Components as an Inhibitor of Peptide Deformylase to Inhibit the Xanthomonas oryzae pv.oryzae via Applying Computational Approach

Bacterial leaf blight(BLB)caused by Xanthomonas oryzae pv.oryzae(Xoo)is one of the most damaging diseases to rice across the world.Various chemicals have been employed so far for the management of bacterial leaf blight.On the other hand,these compounds are damaging to the ecosystem and have an impact on non-target species such as humans and animals.As a result,there is a need to create a new natural inhibitor for BLB management.Deformylase(PDF)enzyme is present in all eubacteria and its necessity in bacterial protein synthesis reveals it as an attractive target for drug development.In this study,the active components of nigella sativa have been selected based on their previously reported antimicrobial activity and screened on the active site of bacterial PDF by the in silico art of techniques.Among these compounds,dithymoquinone and p-cymene strongly bind with the PDF enzyme with binding energy values of 7.77 kcal/mol and 7.26 kcal/mol,respectively,which is comparatively higher than the control compound(−6.73 kcal/mol).Hence,the“dithymoquinone-PDF”and“p-cymene-PDF”complexes were selected for further study,and their stability was assessed by molecular dynamic(MD)simulation.In MD simulation,both selected compounds exhibited steady-state interaction with PDF for 20 ns.It has been hypothesized that p-cymene and dithymoquinone inhibit peptide deformylase and could be used as antibacterials or pesticides against Xoo against the BLB disease.

nigella sativa protects against isoproterenol-induced myocardial infarction by alleviating oxidative stress,biochemical alterations and histological damage

Objective: To evaluate the cardioprotective effect of nigella sativa L.(N.sativa) in isoproterenol-induced myocardial infarction(MI).Methods: Groups were treated with different doses of ethanol extract of N.sativa(EENS)and N.sativa oil alone and along with enalapril for 28 days.MI was induced by subcutaneous administration of isoproterenol(85 mg/kg) in two consecutive doses.Levels of cardiac biomarkers and antioxidant enzymes such as creatine kinase–N-acetyl-L-cysteine, lactate dehydrogenase, aspartate aminotransferase, malondialdehyde, superoxide dismutase, reduced glutathione and catalase were evaluated along with gross histopathological examination.Results: Isoproterenol(85 mg/kg) induced MI by causing the significant(P < 0.01)reduction in the activity of cardiac biomarkers(creatine kinase–N-acetyl-L-cysteine,lactate dehydrogenase, aspartate aminotransferase) and antioxidant markers(superoxide dismutase, catalase, glutathione) along with significant(P < 0.01) increase in the level of malondialdehyde.Furthermore, histopathological evaluation also confirmed the isoproterenol-induced MI.Pretreatment with EENS(800 mg/kg) and combination of EENS(800 mg/kg) with enalapril(1 mg/kg) significantly(P < 0.01) prevented the development of these alteration and restored activity of cardiac biomarkers as well as antioxidant markers almost near to normal levels.Histopathological evaluation of cardiac tissue further confirmed the restoration of biochemical activity.Conclusions: Experimental findings thus indicate that EENS(800 mg/kg) demonstrated cardioprotective effect against isoproterenol-induced MI by restoring cardiac biomarkers and antioxidant status.

Effects of the Hydro-Alcoholic Extract of nigella sativa on Scopolamine-Induced Spatial Memory Impairment in Rats and Its Possible Mechanism

Objective： To evaluate the effect of nigella sativa （NS） extract on memory performance and its possible mechanisms in scopolamine （Sco）-induced spatial memory impairment model using Morris water maze test. Methods： Thirty-two male Wistar rats were randomly divided into four groups. The control group received saline instead of both NS extract and Sco. The Sco group was treated by saline for two weeks, and was injected by Sco （2 mg/kg, intraperitoneally） 30 min before each trail in Morris water maze test. Sco＋NS 200 and SCO＋NS 400 groups were daily treated by 200 or 400 mg/kg of NS （intraperitoneally） for two weeks, respectively, and were finally injected by Sco 30 min before Morris water maze test. The brains of animals were removed to determine the acetylcholinesterase （ACHE） activity and oxidative stress criteria in cortical tissues. Results： Time latency and path length in the Sco group were significantly higher than in the control group （P〈0.01）, while the SCO＋NS 400 group showed a significantly shorter traveled path length and time latency compared with the Sco group （P〈0.01）. AChE activity in the cortical tissues of the Sco group was significantly higher than the control group （P〈0.01）, while AChE activity in the Sco＋NS 200 and Sco＋NS 400 groups was lower than the Sco group （P〈0.01）. Following Sco administration, malondialdehyde （MDA） concentrations were increased （P〈0.01） in comparison with the control group, while cortical total thiol content decreased （P〈0.01）. Pretreatment with extracts caused a significant elevation in cortical total thiol content （P〈0.01） and reduction in cortical MDA concentration （P〈0.01） compared with the Sco group. Conclusions： Hydro-alcoholic extract of NS prevents Sco-induced spatial memory deficits and decreases the AChE activity as well as oxidative stress of brain tissues in rats. Our results support the traditional belief about the beneficial effects of NS in nervous system. Moreover, further investigations are needed for better understanding of this protective effect.

Potential immunomodulation effect of the extract of nigella sativa on ovalbumin sensitized guinea pigs

Several different pharmacological effects have been described for nigella sativa(Siah-Daneh) ,including an anti-inflammatory effect.In the present study,the effect of the extract of N.sativa on lung pathology and blood interleukin-4(IL-4) and interferon-γ(IFN-γ) of sensitized guinea pigs was examined.Three groups(n=8 for each group) of guinea pigs sensitized to ovalbumin(OA) were given drinking water alone,and drinking water containing low and high concentrations of the plant extract,respectively.The animals of the control group(n=8) were treated with saline instead of OA and were given drinking water.The pathological changes of the lung,including infiltration of eosinophils and lymphocytes,local epithelial necrosis,the presence of oedema,thickening of the basement membrane,smooth muscle layer hypertrophy,mucosal secretion,and the presence of mucosal plug,and blood IL-4 and IFN-γof sensi-tized guinea pigs were evaluated.The lungs of the sensitized group showed significant pathological changes(Psativa extract on lung inflammation of sensitized guinea pigs.

Black seed(nigella sativa) is an annual flowering plant from Ranunculaceae family, native to southwest Asia. This plant has many food and medicinal uses. The use of its seeds and oil is common for treatment of many diseases, including rheumatoid arthritis, asthma, inflammatory diseases, diabetes and digestive diseases. The purpose of this study was to provide a comprehensive review on the scientific reports that have been published about N. sativa. The facts and statistics presented in this review article were gathered from the journals accessible in creditable databases such as Science Direct, Medline, Pub Med, Scopus, EBSCO, EMBASE, SID and Iran Medex. The keywords searched in Persian and English books on medicinal plants and traditional medicine, as well as the above reputable databases were "Black seed", "nigella sativa", "therapeutic effect", and "medicinal plant". The results showed that N. sativa has many biological effects such as anti-inflammatory, anti-hyperlipidemic, anti-microbial, anti-cancer, anti-oxidant, anti-diabetic, anti-hypertensive, and wound healing activities. It also has effects on reproductive, digestive, immune and central nervous systems, such as anticonvulsant and analgesic activities. In summary, it can be used as a valuable plant for production of new drugs for treatment of many diseases.

Protective effect of nigella sativa oil against binge ethanolinduced oxidative stress and liver injury in rats

AIM: nigella sativa L.(Ranunculaceae) is considered as a therapeutic plant-based medicine for liver damage. In this study, the aim was to study the effect of nigella sativa oil(NSO) pretreatment on ethanol-induced hepatotoxicity in rats. METHOD: Rats were given nigella sativa oil at doses of 2.5 and 5.0 mL·kg-1, orally for 3 weeks, followed by oral ethanol(EtOH) administration(5 g·kg-1) every 12 h three times(binge model). RESULTS: Binge ethanol application caused significant increases in plasma transaminase activities and hepatic triglyceride and malondialdehyde(MDA) levels. It decreased hepatic glutathione(GSH) levels, but did not change vitamins E and vitamin C levels and antioxidant enzyme activities. NSO(5.0 mL·kg-1) pretreatment significantly decreased plasma transaminase activities, hepatic MDA, and triglyceride levels together with amelioration in hepatic histopathological findings. CONCLUSION: NSO pretreatment may be effective in protecting oxidative stress-induced hepatotoxicity after ethanol administration.

A case study report of acute renal failure associated with nigella sativa in a diabetic patient

1 Introduction nigella sativa, known as black seed, has analgesic, anti-inflammatory, antioxidant and anticancer effects. It has been shown to reduce the development of kidney failure when given prior to the use of nephrotoxic drugs particularly due to its antioxidant action. However, as far as the authors could ascertain, there is no human study in literature showing these effects. Here we present a case of acute renal failure after the use of N. sativa, rather than exhibiting antioxidant or antidiabetic effects.
