neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and reproduction.In this study,to explore the function of neuropeptide LFRFamide in Sepia pharaonis,the full-length cDNA of LFRFamide-like gene(named SpLFRFL,MG869822.1)was identified with rapid amplification of cDNA ends(RACE)method.The sequence of SpLFRFL was 860 bp in length and encoded 188 amino acids containing 4 different mature peptides:1 copy of PHTPFRFamide,NSLFRFamide,TIFRFamide,and 3 copies of GNLFRFamide.Multiple alignment and phylogenetic analysis results showed that SpLFRFL shared high identity with LFRFamides of Sepia officinalis and Sepiella japonica and had the closest relationship with them.Through quantitative Real-time PCR(qRT-PCR),it was found that the SpLFRFL gene was highly expressed in the optic lobe and brain at three different stages during gonad development in both genders.Moreover,the four mature peptides at a concentration of 0.01μmol L^(−1) could inhibit the protein synthesis in the Chinese hamster ovary cell strain-K1(CHOK1)induced by SpGnRH.These data suggest that SpLFRFL might be involved in the development and reproduction of S.pharaonis.The results can contribute to future studies on neuropeptide evolution and function and benefit the cuttlefish farming.

Numerous physiological processes in insects are tightly regulated by neuropeptides and their receptors.Although they form an ancient signaling system,there is still a great deal of variety in neuropeptides and their receptors among different species within the same order.neuropeptides and their receptors have been documented in many hemipteran insects,but the differences among them have been poorly characterized.Commercial grapevines worldwide are plagued by the bug Daktulosphaira vitifoliae(Hemiptera:Sternorrhyncha).Here,33 neuropeptide precursors and 48 putative neuropeptide G protein-coupled receptor(GPCR)genes were identified in D.vitifoliae.Their expression profiles at the probe and feeding stages reflected potential regulatory roles in probe behavior.By comparison,we found that the Releasing Hormone-Related Peptides(GnRHs)system of Sternorrhyncha was differentiated from those of the other 2 suborders in Hemiptera.Independent secondary losses of the adipokinetic hormone/corazoninrelated peptide receptor(ACP)and corazonin(CRZ)occurred during the evolution of Sternorrhyncha.Additionally,we discovered that the neuropeptide signaling systems of Sternorrhyncha were very different from those of Heteroptera and Auchenorrhyncha,which was consistent with Sternorrhyncha's phylogenetic position at the base of the order.This research provides more knowledge on neuropeptide systems and sets the groundwork for the creation of novel D.vitifoliae management strategies that specifically target these signaling pathways.

The NLRP3 inflammasome plays an essential role in resistance to bacterial infection. The nervous system secretes multiple neuropeptides affecting the nervous system as well as immune cells. The precise impact of the neuropeptide CGRP on NLRP3 inflammasome activation is still unclear. Here, we show that CGRP negatively regulates the antibacterial process of host cells. CGRP prevents NLRP3 inflammasome activation and reduces mature IL-1β secretion. Following NLRP3 inflammasome stimulation that triggers endosome leakage, CGRP internalized to endosomal compartments is released into the cell cytosol. Cytosolic CGRP binds directly to NLRP3 and dismantles the NLRP3-NEK7 complex, which is crucial for NLRP3 inflammasome activation. CGRP administration exacerbates bacterial infection, while the treatment with a CGRP antagonist has the opposite effect. Our study uncovers a unique role of CGRP in inhibiting inflammasome activation during infections, which might shed new light on antibacterial therapies in the future.

Functional characterization of an orexin neuropeptide in amphioxus reveals an ancient origin of orexin/orexin receptor system in chordate

Amphioxus belongs to the subphylum cephalochordata, an extant representative of the most basal chordates, whose regulation of endocrine system remains ambiguous. Here we clearly demonstrated the existence of a functional orexin neuropeptide in amphioxus, which is able to interact with orexin receptor, activate both PKC and PKA pathways, decrease leptin expression, and stimulate lipogenesis. We also showed the transcription level of amphioxus orexin was affected by fasting or temperature,indicating a role of this gene in the regulation of energy balance. In addition, the expression of the amphioxus orexin was detected at cerebral vesicle, which has been proposed to be a homolog of the vertebrate brain. These data collectively suggest that a functional orexin neuropeptide has already emerged in amphioxus, which provide insights into the evolutionary origin of orexin in chordate and the functional homology between the cerebral vesicle and vertebrate brain.

In sect n europeptides are a group of brain n euro-regulatory factors,which plays very important roles in growth and developme nt,molting and metamorphosis,as well as mating and reproduction.The neuropeptide F(NPF),a multi-functional neuropeptide,is one of neuropeptides identified in numerous insect species,which plays important roles in feeding,metabolism,courtship,reproduction,aggression,ethanol sensitivity,locomotor circadian rhythms,learning and stress responses.These roles of NPF are implemented through NPF receptors(NPFR).The NPFR1,a G protein-coupled receptor with 7 transmembrane domai ns,is one of these receptors and is found to be imports nt for NPF regulation.The NPF usually is con sisted of around 36-40 amino acid residues,but the short neuropeptide F(sNPF)consisted of 7-16 amino acid residues have also been found in some insects.In this review,the structure and function of both NPF and sNPF in in sects are discussed.

BACKGROUND： Studies in recent years have indicated that several neuropeptide-like substances, such as arginine vasopressin （AVP）, somatostatin （SS）, and β-endorphine （β-EP）, are involved in the process of cerebral ischemic damage to cranial nerves. OBJECTIVE： To observe the effects of electropuncture on back-shu points, as well as the influence on learning and memory, AVP, SS, and β-EP levels in plasma and brain were measured in a rat model of vascular dementia （VD）. DESIGN： Randomized controlled trial. SETTING： College of Acupuncture and Massage of Guangzhou University of Chinese Medicine. MATERIALS： This experiment was performed at the Animal Experiment Center of Guangzhou University of TCM from December 2005 to December 2006. A total of 48 healthy adult male Sprague Dawley rats of SPF-grade, 180-220 g, were provided by The Animal Experiment Center of Guangzhou University of Traditional Chinese Medicine. The following instruments were used： SDQ-30 Dipolar Radio-frequency Electrocoagulator （Shanghai Operation Instrument Factory）, Morris Water Maze （The Animal Experiment Center of Guangzhou University of Traditional Chinese Medicine）, Type G6805-1 Treating Equipment （Huasheng Equipment Factory, Qingdao, China）. METHODS： ① Eight rats were randomly selected for the control group; the remaining 40 rats underwent 4-vascular occlusion to establish a cerebral ischemia model. Due to the death of 13 rats and 2 hemiplegies during model establishment, there was a total of 25 model rats available for testing. The model rats were divided randomly into 3 groups according to their body weight： electropuncture group （n = 9）, medication group （n = 8）, and VD group （n = 8）. ② Electropuncture group： 25 mm needles （28 gauge） were used to electropuncture （150 Hz, continuous waves, 1.0-2.0 mA, duration of 20 minutes） the following acupoints： Baihui （GV20）, Geshu （BLIT）, Pishu （BL20）, and Shenshu （BL23）. The acupoints were located according to Experimental acupuncturology and were stimulated electrically by G6805-1 treating equipment.

We have investigated the effects of magnetic fields on nitric acid (NO) and neuropeptide Y (NPY) on adrenal glands of the rat using NO nitric acid reductase-spec-trophotometry and histochemistry techniques. We found that all cellular layers of the adrenal cortex, including zona glomerulosa, zona fasciculata and zona reticularis were stained by NADPH-d, and some chromaffin cells of the medulla were positive for NPY. Furthermore, magnetic fields increased NO so strongly that high NO levels could be maintained for several hours, as well as some neuroganglion cells in medulla that were double-stained for NPY and NADPH-d. Our data showed that the magnetic field can regulate endocrine and neuroendocrine directly by some action on parenchyma cells, or indirectly by action to NO-ergic, NPY-ergic neurons in the adrenal gland.

Mechanism Research of Reducing Obesity-Induced Insulin Resistance in the White Adipose Tissue by Knockdown of neuropeptide Y Expression in the Dorsomedial Hypothalamus

This study investigated the specific mechanism of knockdown of neuropeptide Y(NPY) in reducing obesity-induced insulin resistance in the white adipose tissue. Adeno-associated virus(AAV)-mediated RNAi was utilized to downregulate NPY expression in rats fed either regular chow or high fat diet. By investigating the differences in rat body weight and food intake, we assessed the effect of knockdown of NPY expression on insulin sensitivity and β-cell proliferation. Glucose consumption and 2-[3 H]DG uptake in 3 T3-L1 adipocytes were assessed to determine the molecular mechanisms. The results showed that knockdown of NPY expression in the dorsomedial hypothalamus(DMH) reduced obesity-induced insulin resistance, increased glucose consumption, and decreased 2-[3 H]DG uptake in 3 T3-L1 adipocytes via the PI3 K/Akt/GSK-3β signaling pathways and the NPY Y5 receptor.

Baculoviruses constitute a large group of invertebrate DNA viruses, predominantly infecting larvae of the insect order Lepidoptera. During a baculovirus infection, the virus spreads throughout the insect body producing a systemic infection in multiple larval tissues, included the central nervous system (CNS). As a main component of the CNS, neuropeptides are small protein-like molecules functioning as neurohormones, neurotransmitters, or neuromodulators. These peptides are involved in regulating animal physiology and behavior and could be altered after baculovirus infection. In this study, we have investigated the effect of Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV) infection on expression of Spodoptera exigua neuropeptides and neuropeptide-like genes. Expression of the gene encoding a polypeptide that resembles the well-known insect neuropeptide proctolin and named as proctolin-like peptide (PLP), was downregulated in the larval brain following infection and was chosen for further analysis. A recombinant Autographa californica multiple nucleopolyhedrovirus (AcMNPV) overexpressing the C-terminal part of the PLP was generated and used in bioassays using S. exigua larvae to study its influence on the viral infection and insect behavior. AcMNPV-PLP-infected larvae showed less locomotion activity and a reduction in growth compared to larvae infected with wild type AcMNPV or mock-infected larvae. These results are indicative of this new peptide as a neuromodulator that regulates visceral and skeletal muscle contractions and offers a novel effector involved in the behavioral changes during baculovirus infection.
