Objective: To better investigate the protective role of branched-chain amino acids(BCAAs)and Cymbopogon schoenanthus(CS) extract against the potassium dichromate(PDC)-induced oxido-nitrosative nephrotoxic insult in the experimental rat model. Methods: Thirty male rats were randomly divided into five equal groups: The 1 st group served as control; the 2^(nd)was injected with a single dose of PDC(15 mg/kg b.w i.p.);the 3^(rd), 4^(th), and 5^(th) groups were respectively treated with BCAAs, CS, and their combination for 15 d prior to induction of renal insult via PDC single dose(15 mg/kg b.w s.c.). The experimental period was terminated in all groups 2 d after induction of renal insult. The harvested kdney samples were divided for biochemical assays and histological examination. Results: The PDC-induced nephrotoxic effect caused a depletion of renal oxidative scavengers glutathione, superoxide dismutase with consequent lipo-oxidative cellular membrane deterioration manifested by a rise in malonaldehyde, oxidized glutathione, myeloperoxidase and the concomitant increase in inflammatory response elements tumor necrosis factor α, nitric oxide, and interleukin 1 β.Moreover, the comet assay and increased 8-hydroxy-2-deoxyguanosine proved an accelerated apoptotic DNA fragmentation. These local renal changes were met with global altered blood biochemistry. The BCAAs and CS or their compiled administration showed an ameliorative effect against PDC-induced nephrotoxic in a synergistic pattern. Conclusions: Both BCAAs and CS or their combined administration afford potential competitors against renal insult induced by polyvalent anion pollutants in experimentally studied animals model. As a route for novel drug discovery, further investigation should be attempted to optimize their augmenting reno-protecting potential.

Objective: To evaluate the toxic potential of Launaea taraxacifolia leaf extract(LTE) in rats within 14 d of oral administration and also assess the potential of LTE in protecting against kidney injury induced by gentamicin using rat model.Methods: The protective ability of LTE was done after sub-acute toxicity evaluation has been carried out. Acute Kidney Injury(AKI) was induced by gentamicin at a dose of 160 mg/kg intraperitoneal i.p. Parameters and indicators considered include mortality,clinical signs, body and organ weights, haematological and clinical chemistry parameters.Gross examination and histopathological assessment was also done on selected internal organs.Results: There were no treatment-related deaths or changes in clinical signs, haematological and clinical chemistry indices during sub-acute toxicity studies with the exception of creatinine levels. This was confirmed by micrographs obtained from histopathological analysis. Co-administration of LTE with 160 mg/kg of gentamicin(i.p) markedly decreased the levels of urea and creatinine when compared to negative control group.Histological studies of kidney tissues showed an insignificant change in tubular epithelium in LTE plus gentamicin treated group compared to LTE treated only.Conclusions: Data obtained show that ethanolic leaf extract of Launaea taraxacifolia is non-toxic within a 14 d administration at a maximum dose of 1 000 mg/kg bwt and also possesses the ability to protect against gentamicin-induced kidney damage in rats at a dose of 300 mg/kg bwt.

p-Coumaric acid ameliorates ethanol-induced kidney injury by inhibiting inflammatory cytokine production and NF-κB signaling in rats

Objective: To examine the effects of p-coumaric acid on ethanol-induced kidney injury in Swiss Wistar rats.Methods: Ethanol(25% v/v) was used to induce nephrotoxicity in rats. p-Coumaric acid was orally administered at 50, 100, or 200 mg/kg body weight. The levels of oxidative parameters were determined; pro-inflammatory biomarkers were analyzed by Western blotting and apoptotic protein was analyzed by immunohistochemistry. Results: Ethanol treated rats showed decreased levels of antioxidants and aberrant production of pro-inflammatory cytokines(IL-6, IL1β, TNF-α), NF-κB activation and imbalance of proand anti-apoptotic proteins(Bcl-2, Bax, caspase 3). Meanwhile, p-coumaric acid restored antioxidant levels and decreased the levels of inflammatory cytokines, NF-κB, and proapoptotic proteins and increased Bcl-2 expression. Conclusions: p-Coumaric acid ameliorates ethanol-induced kidney injury by restoring antioxidant production and suppressing cellular apoptosis and inhibiting NF-κB expression.p-Coumaric acid should be further investigated as a promising candidate for ethanol-induced kidney toxicity.

BACKGROUND Interferons(IFNs)are characterized by a wide range of biological effects,which justifies their potential therapeutic use in several pathologies,but also elicit a wide array of adverse effects in almost every organ system.Among them,renal involvement is probably one of the most complex to identify.CASE SUMMARY We describe four cases of kidney damage caused by different IFN formulations:IFN-β-related thrombotic microangiopathy,IFN-β-induced systemic lupus erythematosus,and two cases of membranous nephropathy secondary to pegylated-IFN-α2B.In each case,we carefully excluded any other possible cause of renal involvement.Once suspected as the casual relationship between drug and kidney damage,IFN treatment was immediately discontinued.In three cases,we observed a complete and persistent remission of clinical and laboratory abnormalities after IFN withdrawal,while the patient who developed thrombotic microangiopathy,despite IFN withdrawal and complement-inhibitor therapy with eculizumab,showed persistent severe renal failure requiring dialysis.CONCLUSION This case series highlights the causal relationship between IFN treatment and different types of renal involvement and enables us to delineate several peculiarities of this association.

Objective:To evaluate the effect of resveratrol against CCl4-induced nephrotoxicity.Methods:Forty-two male Wistar rats were divided into seven groups randomly.After six weeks,kidney weight,body weight,blood urea,serum creatinine,oxidative stress markers,and gene expression of renal transforming growth factor-beta1(TGF-β1),TGF-βreceptor type 1(TGF-βR1)and Smad3 were determined.In addition,the protein level of TGF-β1 in the tissue lysate was measured.Results:Resveratrol had a protective role in renal tissue by the improvement of antioxidant balance and reduction of renal parameters such as creatinine and urea(Pnephrotoxicity in CCl4 treated rats by reducing oxidative stress status and modulating the TGF-βsignaling.

Objective: To observe the clinical effect of Yiqi Jianpi Bushen recipe (YQJPBS, 益气健脾补肾方) in antagonizing nephrotoxicity and hematological toxicity by cisplatin (CDDP). Methods: The patients with different types of carcinomas were divided into two groups. One group was treated by western medicine (WM), i.e. cisplatin combined with diuretic therapy. The other group by Chinese medicine (CM). One day before chemotherapy, YQJPBS was given orally. Urinary enzyme (NAG, γ-GT, ALP) and serum erythropoietin (Epo) were used as the key monitoring indicator. Results: The early renal damage by CDDP remains near proximal convoluted tubule, which leads to the rising of urinary enzyme. So urinary enzyme of the CM group was obviously lower than that of the WM group. The lesion of renal interstitial cells that produce Epo did not cause the serum Epo of WM group to rise obviously 15 days later. As a result there was 40% patients whose RBC and Hb became lower than the standard count. While serum Epo of CM group was higher than that of WM group, only 5% patients in the CM group had their RBC and Hb lowered. Conclusion: YQJPBS can effectively antagonize nephrotoxicity and hematological toxicity induced by CDDP.

The development of nephrotoxicity largely limits the clinical use of chemotherapy.MiRNAs are able to target various genes and involved in the regulation of diverse cellular processes,including cell apoptosis and death.Our study showed that miR-181 a expression was significantly increased after 5-fluorouracil（5-FU） treatment in renal mesangial cells and kidney tissue,which was associated with decreased baculoviral inhibition of apoptosis protein repeat containing 6（BIRC6） expression and increased apoptotic rate.Enforced miR-181 a expression enhanced 5-FU-induced p53-dependent mitochondrial apoptosis,including declined Bcl-2/Bax ratio,loss of mitochondrial membrane potential,cytochrome c release,and caspase-9 and caspase-3 activation.However,inhibition of miR-181 a was associated with reduced p53-mediated mitochondrial apoptosis induced by 5-FU.Moreover,miR-181 a increased BIRC6 downstream gene p53 protein expression and transcriptional activity by reducing ubiquitin-mediated protein degradation.We found that miR-181 a directly targeted 3’-UTR of BIRC6 mRNA and negatively regulated BIRC6 expression.In vivo study,knockdown of miR-181 a with adeno-associated virus harboring miR-181 a-tough decoy attenuated 5-FU-induced renal cell apoptosis,inflammation and kidney injury.In conclusion,these results demonstrate that miR-181 a increases p53 protein expression and transcriptional activity by targeting BIRC6 and promotes5-FU-induced apoptosis in mesangial cells.Inhibition of miR-181 a ameliorates5-FU-induced nephrotoxicity,suggesting that miR-181 a may be a novel therapeutic target for nephrotoxicity treatment during chemotherapy.

The aminolycoside Gentamicin is a widely used antibiotic,applied in equine medicine.Despite its clinical use,concerns remain regarding the potential toxic side-effects,such as nephrotoxicity.Early detection of renal damage is critical in preclinical drug development.This study was aimed to determine whether kidney injury molecule-1(KIM-1) and neutrophil gelatinase-associated lipocalin(NGAL) may be early indicators in the assessment of Gentamycin-induced nephrotoxicity.In our study,a model of Gentamicin-induced nephrotoxicity in male Sprague Dawley rats treated for up to 7 days at 50 or 100mg/kg/day was used to monitor the expressions of novel biomarkers of renal toxicity during the progression of acute kidney injury(AKI).Additionally,biomarkers were assessed in human kidney proximal epithelial cells(HK-2)treated with Gentamicin for 2,6,12,24,36 or 48 h in vitro.Repeated administration of Gentamicin to rats for 1,3,or 7 days resulted in a dose-and time-dependent increase in the expression of KIM-1 and NGAL.The expressions of the two biomarkers changed prior to proximal tubule damage and increases in serum creatinine(SCr)and blood urea nitrogen(BUN)levels,suggesting their usefulness for predicting Gentamicin-induced acute kidney injury(AKI)in vivo.In contrast,no significant increase in the expression of the biomarker genes and proteins were evident in HK-2 cells after treated by Gentamycin for up to 48 h,suggesting that they may not be suitable endpoints for sensitive detection of nephrotoxic effects in vitro.
